Reports about diurnal changes of assimilates in phloem sap are controversial. We determined the diurnal changes of sucrose and amino acid concentrations and fluxes in exudates from cut aphid stylets on tansy leaves (Tanacetum vulgare), and sucrose, amino acid and K(+) concentrations and fluxes in bleeding sap of castor bean pedicel (Ricinus communis). Approximately half of the tansy sieve tubes exhibited a diurnal cycle of sucrose concentrations and fluxes in phloem sap. Data from many tansy plants indicated an increased sucrose flux in the phloem during daytime in case of low N-nutrition, not at high N-nutrition. The sucrose concentration in phloem sap of young Ricinus plants changed marginally between day and night, whereas the sucrose flux increased 1.5-fold during daytime (but not in old Ricinus plants). The amino acid concentrations and fluxes in tansy sieve tubes exhibited a similar diurnal cycle as the sucrose concentrations and fluxes, including their dependence on N-nutrition. The amino acid fluxes, but not the concentrations, in phloem sap of Ricinus were higher at daytime. The sucrose/amino acid ratio showed no diurnal cycle neither in tansy nor in Ricinus. The K(+)-concentrations in phloem sap of Ricinus, but not the K(+) fluxes, decreased slightly during daytime and the sucrose/K(+)-ratio increased. In conclusion, a diurnal cycle was observed in sucrose, amino acid and K(+) fluxes, but not necessarily in concentrations of these assimilates. Because of the large variations between different sieve tubes and different plants, the nutrient delivery to sink tissues is not homeostatic over time.  相似文献

Peroxisomes undergo rapid, selective autophagic degradation (pexophagy) when the metabolic pathways they contain are no longer required for cellular metabolism. Pex3 is central to the formation of peroxisomes and their segregation because it recruits factors specific for these functions. Here, we describe a novel Saccharomyces cerevisiae protein that interacts with Pex3 at the peroxisomal membrane. We name this protein Atg36 as its absence blocks pexophagy, and its overexpression induces pexophagy. We have isolated pex3 alleles blocked specifically in pexophagy that cannot recruit Atg36 to peroxisomes. Atg36 is recruited to mitochondria if Pex3 is redirected there, where it restores mitophagy in cells lacking the mitophagy receptor Atg32. Furthermore, Atg36 binds Atg8 and the adaptor Atg11 that links receptors for selective types of autophagy to the core autophagy machinery. Atg36 delivers peroxisomes to the preautophagosomal structure before being internalised into the vacuole with peroxisomes. We conclude that Pex3 recruits the pexophagy receptor Atg36. This reinforces the pivotal role played by Pex3 in coordinating the size of the peroxisome pool, and establishes its role in pexophagy in S. cerevisiae.  相似文献

The 5898 nucleotide single-strand RNA genome of Sugarcane yellow leaf virus (SCYLV) contains one long open reading frame, which is translated into a 120.6 kDa polyprotein. The sequences of SCYLV isolates from the two SCYLV-susceptible cultivars from Hawaii had a deletion of 48–51 nt in ORF1. SCYLV from 12 sugarcane hybrid cultivars from different origins were tested by RT-PCR using a specific set of primers, to investigate the genome segment for this deletion. Only three cultivars were found not to have the deletion (H87-4319, JA-605 and CP52-43), while SCYLV from nine cultivars (H73-6110, H87-4094, H78-7750, GT54-9, G84-47, H78-4153, H65-7052, C1051-73, Ph-8013) along with aphid (Melanaphis sacchari), which fed on SCYLV-infected H73-6110, contained a deletion of about 50 nt. The deleted sequence was located in the overlap frameshift of ORF1 and ORF2. Thus, ORFs 1 and 2 of SCYLV are translated via ribosomal frameshift and yield the 120.6 kDa viral replicase. ORF1 plays most likely a role in the replication and is a source of large variability among the virus population. To identify possible recombination events located in the RdRp domain of the Hawaiian isolates, two programs were used: RDP v.4.3 and RECCO. It is noteworthy that according both methods Haw73-6110 was found as a potential recombinant. On the other hand, opposed to the RDP package, RECCO revealed that Haw87-4094 isolate was also a recombinant whereas Haw87-4319 was not.  相似文献

The field of plant-made therapeutics in South Africa is well established in the form of exploitation of the country's considerable natural plant diversity, both in the use of native plants in traditional herbal medicines over many centuries, and in the more modern extraction of pharmacologically-active compounds from plants, including those known to traditional healers. In recent years, this has been added to by the use of plants for the stable or transient expression of pharmaceutically-important compounds, largely protein-based biologics and vaccines. South Africa has a well-developed plant biotechnology community, as well as a comprehensive legislative framework for the regulation of the exploitation of local botanic resources, and of genetically-modified organisms. The review explores the investigation of both conventional and recombinant plants for pharmaceutical use in South Africa, as well as describing the relevant legislative and regulatory frameworks. Potential opportunities for national projects, as well as factors limiting biopharming in South Africa are discussed.  相似文献

Magnaporthe oryzae is a major pathogen of rice (Oryza sativa L.) but is also able to infect other grasses, including barley (Hordeum vulgare L.). Here, we report a study using Magnaporthe isolates collected from other host plant species to evaluate their capacity to infect barley. A nonhost type of resistance was detected in barley against isolates derived from genera Pennisetum (fontaingrass) or Digitaria (crabgrass), but no resistance occurred in response to isolates from rice, genus Eleusine (goosegrass), wheat (Triticum aestivum L.), or maize (Zea mays L.), respectively. Restriction of pathogen growth in the nonhost interaction was investigated microscopically and compared with compatible interactions. Real-time polymerase chain reaction was used to quantify fungal biomass in both types of interaction. The phylogenetic relationship among the Magnaporthe isolates used in this study was investigated by inferring gene trees for fragments of three genes, actin, calmodulin, and beta-tubulin. Based on phylogenetic analysis, we could distinguish different species that were strictly correlated with the ability of the isolates to infect barley. We demonstrated that investigating specific host interaction phenotypes for a range of pathogen isolates can accurately highlight genetic diversity within a pathogen population.  相似文献

An anthropoid-specific segmental duplication on human chromosome 1q22   总被引:1,自引:0,他引:1  
Segmental duplications (SDs) play a key role in genome evolution by providing material for gene diversification and creation of variant or novel functions. They also mediate recombinations, resulting in microdeletions, which have occasionally been associated with human genetic diseases. Here, we present a detailed analysis of a large genomic region (about 240 kb), located on human chromosome 1q22, that contains a tandem SD, SD1q22. This duplication occurred about 37 million years ago in a lineage leading to anthropoid primates, after their separation from prosimians but before the Old and New World monkey split. We reconstructed the hypothetical unduplicated ancestral locus and compared it with the extant SD1q22 region. Our data demonstrate that, as a consequence of the duplication, new anthropoid-specific genetic material has evolved in the resulting paralogous segments. We describe the emergence of two new genes, whose new functions could contribute to the speciation of anthropoid primates. Moreover, we provide detailed information regarding structure and evolution of the SD1q22 region that is a prerequisite for future studies of its anthropoid-specific functions and possible linkage to human genetic disorders.  相似文献

Saccharomyces cerevisiae Pex3p has been shown to act at the ER during de novo peroxisome formation. However, its steady state is at the peroxisomal membrane, where its role is debated. Here we show that Pex3p has a dual function: one in peroxisome formation and one in peroxisome segregation. We show that the peroxisome retention factor Inp1p interacts physically with Pex3p in vitro and in vivo, and split-GFP analysis shows that the site of interaction is the peroxisomal membrane. Furthermore, we have generated PEX3 alleles that support peroxisome formation but fail to support recruitment of Inp1p to peroxisomes, and as a consequence are affected in peroxisome segregation. We conclude that Pex3p functions as an anchor for Inp1p at the peroxisomal membrane, and that this function is independent of its role at the ER in peroxisome biogenesis.  相似文献

We present insights from a study on communicating Synthetic Biology conducted in 2008. Scientists were invited to write press releases on their work; the resulting texts were passed on to four journalists from major Austrian newspapers and magazines. The journalists in turn wrote articles that were used as stimulus material for eight group discussions with select members of the Austrian public. The results show that, from the lab via the media to the general public, communication is characterized by two important tendencies: first, communication becomes increasingly focused on concrete applications of Synthetic Biology; and second, biotechnology represents an important benchmark against which Synthetic Biology is being evaluated.  相似文献

A new species Ganoderma carocalcareus (Basidiomycota, Ganodermataceae) was collected on living trunk and dead stumps of Anthocleista nobilis (Gentianaceae) in waterlogged swamps in the Mbalmayo Forest Reserve, Cameroon, and identified on the basis of morphology and phylogenetic analyses inferred from mitochondrial small subunit (mtSSU) and internal transcribed spacer (ITS1-5.8S-ITS2) rDNA sequences. Distinct phenotypic characteristics of the new species are dimorphism of basidiomata and variability in context structure and texture over developmental stages. The young basidiomata is ungulate to punk-shaped with context composed of vegetative hyphae attended by scattered, orbicular, smooth, thick-walled chlamydospores, and the mature basidiomata is cushion- to bracket-like with context entirely consisting of chlamydospores masses. This ontogeny intimates the origin of chlamydospores, for which the biogenesis correlates the vanishing of vegetative hyphae throughout the basidiomata maturation. Morphological comparison included Tomophagus colossus (=G. colossus), G. subamboinense and G. weberianum, the known Ganodermataceae species producing chlamydospores and or gasterospores in basidiomata tissues, and G. resinaceum, the closest species with regard to morphology. It followed that G. carocalcareus could not be assigned to these or any other known Ganoderma species. Analyses of mtSSU and ITS rDNA sequence data resolved G. carocalcareus in the G. resinaceum group as a distinct species, but being a close relative of both G. subamboinense and G. weberianum.  相似文献

Biodiversity inventories unravel insightful taxonomic and biogeographic patterns that provide valuable inputs in guiding conservation practices and policymaking. Here we present an updated inventory of the native and alien woody flora of the Kashmir Himalaya, India, together with analyses of the patterns of taxonomic composition, geographic distribution, and invasion status of the alien species. The data was assembled from the floristic literature of the last two centuries, supplemented with herbarium records and validated by field surveys over the past decade. In total, the woody flora comprised of 520 species; out of these 322 species were native and 198 alien, yielding a proportion of 38% aliens. Amongst the natives, 16 species are on the IUCN red list 2009. While 37 genera included both native and alien species indicating a potential for hybrid formation, only 15 families were represented by alien species exclusively. Regarding the invasion status, presently 37 alien woody species are naturalized and 7 are invasive in the region. The results show that the Kashmir Himalaya harbours a diverse woody flora. It is concluded that a distinction between the native and the alien flora, as adopted in the present study, is crucial for a meaningful analysis of any biodiversity patterns. We predict that in the near future many alien woody species, presently naturalized in the region, can become serious plant invaders, and therefore need further research efforts and timely management.  相似文献
