The previously obtained insertion mutants of Azospirillum brasilense Sp245 in the genes mmsB1 and fabG1 (strains SK039 and Sp245.1610, respectively) were characterized by impaired flagellation and motility. The putative products of expression of these genes are 3-hydroxyisobutyrate dehydrogenase and 3-oxoacyl-[acyl-carrier protein] reductase, respectively. In the present work, A. brasilense strains Sp245, SK039, and Sp245.1610 were found to have differences in the content of 3-hydroxyhexadecanoic, hexadecanoic, 3-hydroxytetradecanoic, hexadecenoic, octadecenoic, and nonadecanoic acids in their lipopolysaccharide preparations, as well as in cell hydrophobicity and hemagglutination activity and dynamics of cell aggregation, in biomass amount, and in the relative content of lipopolysaccharide antigens in mature biofilms formed on hydrophilic or hydrophobic surfaces.  相似文献

Summary The presence of multiple molecular forms (MMF) of glutamine synthetase (GS) has been studied in pumpkin plants and in cotyledons of bean plants. Two MMF of GS have been found in pumpkin leaves and in green cotyledons: chloroplast GS and cytosol GS. Cotyledons of etiolated pumpkin seedlings contain only the cytosol GS. Illumination of etiolated pumpkin seedlings with white light results in the appearance, within one minute, of the second molecular form, the chloroplast GS, which appears to be due to activation rather than de novo synthesis of the enzyme. Cotyledons of resting seeds of horse bean, pea, soybean and lupine contain only one form of GS. The second form, chloroplast GS, appears after germination in the light, but only in those cotyledons of soybean and lupine that can become green.  相似文献

We have synthesized the 24-41 fragment of the preS region of the hepatitis B (subtype ayw) envelope. The peptide was prepared by the solid phase synthesis on perfluoropoly-ethylene polymer grafted with polystyrene. The peptide chain was elongated from C-terminus using pentafluorophenyl- and p-nitrophenyl esters of Boc-amino acids. The peptide was cleaved off the solid phase by HBr in CF3COOH, purified by gel filtration, and, after conjugation with serum albumin (BSA), inoculated into mice. The resultant antibodies were shown to react with the peptide. The blood sera from patients with acute hepatitis B reacted with the peptide-BSA conjugates in the immunoenzymic solid phase assay.  相似文献

The effect of water soluble tocopherol forms (dipotassium salt d,l-alpha-tocopheryl-phosphate) on lipid peroxidation induced by oxygen in membranes of human erythrocytes has been studied. An erythrocyte was considered as a structure reflecting the state of plasmatic membranes of organs and tissues. It has been established that this substance doses optimum for membrane protection against lipid peroxidation are not in excess of 1.5-5.0 mg per 1 kg of human body mass; the dose by an order higher may exert a toxic effect.  相似文献

Folding and assembling of newly synthesized proteins is directed and effected by a group of relatively recently discovered proteins called molecular chaperones. These proteins not only control the assembling of native structures; they also remodel protein molecules that have wrong conformations. All molecular chaperones perform the same function, but structurally they are divided into groups of chaperones and chaperonins. These proteins are highly conserved in evolution and display an ATPase activity. The specificity of perch (Perca fluviatilis) roe fucolectin was studied using the protein dot blot technique, followed by detection with colloidal gold-labeled neoglycoproteins bearing human milk polysaccharides. The strongest binding was noted with the H type 1 pentasaccharide lacto-N-fucopentaose (LNFP I, Fuc alpha 1-2 Gal beta 1-3 GlcNAc beta 1-3 Gal beta 1-4Glc); the interaction with the H type 6 trisaccharide 2'-fucosyllactose (2-FL, Fuc alpha 1-2 Gal beta 1-4 Glc) was weaker. Binding of the perch lectin to the Lewis antigens (associated with tumors and embryonic tissues) was also studied. It was found that the lectin weakly interacted with the hexasaccharide lacto-N-difucohexaose I, Fuc alpha 1-2 Gal beta 1-3[Fuc alpha 1-4]GlcNac beta 1-3 Gal beta 1-4 Glc), but not with Lea, Lec, Lex antigens. Thus, perch roe lectin exhibited pronounced differences in carbohydrate specificity from other fucolectins--a feature that may be used in structural studies and isolation of fucose-containing glycoconjugates.  相似文献

The effect of applying Lactobacillus buchneri, alone or in combination with Lactobacillus plantarum, at ensiling, on the aerobic stability of wheat and corn silages was studied in 50-l plastic containers. Treatments comprised control (no additives), L. plantarum, L. buchneri and a combination of L. plantarum+L. buchneri. After 3 months of storage, the wheat silages treated with L. buchneri had higher acetic acid contents than the control or L. plantarum-treated silages, and were free of mold, whereas the top layers of the control or L. plantarum-treated silages were moldy. In an aerobic stability test the L. buchneri-treated silages were stable, whereas those treated with L. plantarum deteriorated. In the corn silages the effects of L. buchneri were not as clear and the top layer was moldy in all silages. However, L. buchneri also improved the aerobic stability of the corn silage, as indicated by lower yeast numbers, less CO2 production and stable pH. It is concluded that L. buchneri has a potential as a silage additive that protects the silage upon aerobic exposure. The effect of exogenously added vitamin E and its synthetic analogues (the hydrophilic form of vitamin E and chromans C13 and C1) on the growth, lipogenic activity, and the fatty acid composition of the eicosapolyenoic acid-synthesizing oomycete Pythium debaryanum was studied. The effect was found to depend on the molecular structure of particular compounds. For instance, vitamin E and chroman C13 stimulated fungal growth, whereas chroman C1 inhibited it. The hydrophilic form of vitamin E enhanced the lipogenic activity of the oomycete. The studied compounds, which possess antioxidant activity, did not exert any noticeable effect on the level and the degree of the unsaturation of fungal lipids.  相似文献

Fossil pollen grains of Castaneoideae from Holocene sediments from the southern Sea of Japan (J-3) at a depth of 200–202, 165–167, and 120–122 cm were studied with a scanning electron microscope. Pollen of some extant species of Castaneoideae, native to the region (Castanopsis cuspidata, C. sclerophylla, and Lithocarpus glaber) were also studied. As a result, four types of fossil pollen grains were revealed; type I is identified to species as Castanopsis cuspidata, type II is referred to the genus Castanopsis, type III is assigned to the genus Lithocarpus, and type IV is only identifined to subfamily (Castaneoideae type).  相似文献
