Macrolophus pygmaeus Rambur (Hemiptera: Miridae) is a generalist natural enemy that is used to control multiple pest species in a variety of horticultural crops. The bugs are released at the start of the crop cycle to allow them to establish and build up a population in the crop that can control pest infestations later in the season. To facilitate population growth and dispersal in protected sweet pepper crops, Capsicum annuum L. (Solanaceae), food should be supplemented in a full‐field fashion during the first 6–8 weeks after introduction. To reduce the costs of food supplementation, we investigated whether fewer applications could produce similar results in terms of population growth and dispersal within the greenhouse. First, a cage experiment was carried out in which a weekly and biweekly application rate was tested for three food sources: cysts of brine shrimps Artemia spp. (Anostraca: Artemiidae), eggs of the Mediterranean flour moth, Ephestia kuehniella Zeller (Lepidoptera: Pyralidae), and a commercial mix of the two. Artemia spp. cysts resulted in the largest M. pygmaeus populations. There was no difference in population size between the two application rates for any of the food sources. Second, a greenhouse experiment was set up to test both application rates for Artemia spp. cysts under conditions mimicking commercial practice. Again, no difference in population size was observed between a weekly and a biweekly application rate. This insight is good news for growers, as they can use the least expensive food source and they need fewer applications to successfully introduce M. pygmaeus in protected sweet pepper crops.  相似文献

Pregnancy is a hypercoagulable state which carries an excess risk of maternal venous thrombosis. Endothelial injury, alterations in blood flow and activation of the coagulation pathway are proposed to contribute to the hypercoagulability. The risk for thrombosis may be accentuated by certain drugs and device implants that directly or indirectly affect the coagulation pathway. To help ensure that these interventions do not result in adverse maternal or fetal outcomes during pregnancy, gravid experimental animals can be exposed to such treatments at various stages of gestation and over a dosage range that would identify hazards and inform risk assessment. Circulating soluble biomarkers can also be evaluated for enhancing the assessment of any increased risk of venous thrombosis during pregnancy. In addition to traditional in vivo animal testing, efforts are under way to incorporate reliable non‐animal methods in the assessment of embryofetal toxicity and thrombogenic effects. This review summarizes hemostatic balance during pregnancy in animal species, embryofetal development, biomarkers of venous thrombosis, and alterations caused by drug‐induced venous thrombosis. Synthesis and preliminary in vitro biological evaluation of a selective high-affinity CRTH2 antagonist is described. The stability of an N-benzyl group facilitated synthesis of the corresponding radioligand by tritiation of a brominated precursor. The compound [(3)H]TRQ11238 represents the first selective CRTH2 antagonist radioligand and exhibited a specific radioactivity of 52 Ci/mmol and a pK(d) of 9.0.  相似文献

We previously showed that production of reactive oxygen species (ROS) caused by overexpression of the mitochondrial electron transfer protein adrenodoxin (Adx) induces apoptosis in mammalian cells. In the fission yeast Schizosaccharomyces pombe, ROS are also produced in cells that undergo an apoptotic-like cell death, but it is not yet clear whether they are actually causative for this phenomenon or whether they are merely produced as a by-product. Therefore, the purpose of this study was to trigger mitochondrial ROS production in fission yeast by overexpression of either wildtype Adx (Adx-WT) or of several activated Adx mutants and to investigate its consequences. It was found that strong expression of either Adx-WT or Adx-S112W did not produce any ROS, while Adx-D113Y caused a twofold and Adx1–108 a threefold increase in ROS formation as compared to basal levels. However, no typical apoptotic markers or decreased viability could be observed in these strains. Since we previously observed that an increase in mitochondrial ROS formation of about 60% above basal levels is sufficient to strongly induce apoptosis in mammalian cells, we conclude that S. pombe is either very robust to mitochondrial ROS production or does not undergo apoptotic cell death in response to mitochondrial ROS at all.  相似文献

Doppel (Dpl) is the first described homologue of the prion protein, the main constituent of the agent responsible for prion diseases. The cellular prion protein (PrP(C)) is predominantly present in the central nervous system. Although its role is not yet completely clarified, PrP(C) seems to be involved in Cu(2+) recycling from synaptic clefts and in preventing neuronal oxidative damage. Conversely, Dpl is expressed in heart and testis and has been shown to regulate male fertility by intervening in gametogenesis and sperm-egg interactions. Therefore, despite a high sequence homology and a similar three-dimensional fold, the functions of PrP(C) and Dpl appear unrelated. Here we show by electron paramagnetic resonance and fluorescence spectroscopy that the in vitro binding of copper(II) to human recombinant Dpl occurs with a different pattern from that observed for recombinant PrP. At physiological pH values, two copper(II)-binding sites with different affinities were found in Dpl. At lower pH values, two additional copper(II)-binding sites can be identified as follows: one complex is present only at pH 4, and the other is observed in the pH range 5-6. As derived from the electron paramagnetic resonance characteristics, all Dpl-copper(II) complexes have a different coordination sphere from those present in PrP. Furthermore, in contrast to the effect shown previously for PrP(C), addition of Cu(2+) to Dpl-expressing cells does not cause Dpl internalization. These results suggest that binding of the ion to PrP(C) and Dpl may contribute to the different functional roles ascribed to these highly homologous proteins.  相似文献

The (lyso)phospholipid mediators sphingosine-1-phosphate (S1P), lysophosphatidic acid (LPA), sphingosylphosphorylcholine (SPC), and phosphatidic acid (PA) regulate diverse cellular responses such as proliferation, survival and death, cytoskeletal rearrangements, cell motility, and differentiation among many others. Signaling is complex and many signaling events are mediated through the activation of cell surface seven transmembrane (7TM) G protein coupled receptors. Five high affinity receptors for S1P have been identified so far and named S1P(1, 2,3,4,5) (formerly referred to as endothelial differentiation gene (edg)1, 5, 3, 6, 8). Recently, the orphan receptor GPR63 was identified a low affinity S1P receptor structurally distant from the S1P(1-5) family. The orphan GPR3, 6, 12 cluster, phylogenetically related to the edg and melanocortin receptors appears to be subject to modulation by S1P and SPC although all three receptors are strong constitutive stimulators of the Galphas-adenylyl cyclase (AC) pathway and would not require additional ligand stimulation but rather inverse agonism to control activity. Ovarian cancer G protein coupled receptor 1 (OGR1) and GPR4, two structurally closely related receptors were assigned in functional and binding studies as high affinity molecular targets for SPC. Very recently, however, both OGR1 and GPR4 were described as receptors endowed with the ability to signal cells in response to protons. LPA exerts its biological effects through the activation of G protein coupled LPA(1-3) receptors (formerly referred to as edg2, 4, 7). A fourth high affinity LPA receptor has been identified: P2Y9 (GPR23) structurally related to nucleotide receptors and phylogenetically quite distant from the high affinity LPA(1-3) cluster. This review attempts to give an overview about the existing families of lysophosholipid receptors and the spectrum of lipid agonists they use as high or low affinity ligands to relay extracellular signals into intracellular responses. Recently deorphaned lipid receptors, within and outside the known lipid receptor clusters will receive particular attention.  相似文献

This new and simple method of DNA extraction from composite soil allows the isolation of plant DNA with high efficiency, quality and reproductivity. The method is based on a simple CaCl2-precipitation step and requires no additional purification steps to eliminate humic acids. The extracted DNA was obtained in sufficient purity and quantity to allow direct detection of transgenes by PCR. Furthermore, the simple procedure allows the assay of many samples at the same time.  相似文献

The investigation of self‐resistance in antibiotic producers is important to understand the emergence of antibiotic resistance in pathogens and to improve antibiotic production. Lantibiotics are ribosomally synthesized antibiotics that mostly target peptidoglycan biosynthesis. The actinomycete Microbispora ATCC PTA‐5024 produces the lantibiotic NAI‐107, which interferes with peptidoglycan biosynthesis by binding bactoprenol‐pyrophosphate‐coupled peptidoglycan precursors. In order to understand how Microbispora counteracts the action of its own antibiotic, its peptidoglycan composition was analysed in detail. Microbispora peptidoglycan consists of muropeptides with D‐Ala and Gly in similar proportion at the fourth position of the peptide stems and alternative 3‐3 cross‐links besides the classical 4‐3 cross‐links. In addition, the NAI‐107 biosynthetic gene cluster (mlb) was analysed for the expression of immunity proteins. We show that distinct immunity determinants are encoded in the mlb cluster: the ABC transporter MlbYZ acting cooperatively with the transmembrane protein MlbJ and the lipoprotein MlbQ. NMR structural analysis of MlbQ revealed a hydrophobic surface patch, which is proposed to bind the cognate lantibiotic. This study demonstrates that immunity in Microbispora is not only based on one determinant but on the action of the distinct immunity proteins MlbQ, MlbYZ and MlbJ.  相似文献

To establish the capacity of the leaf mesophyll plasmalemma of Phaseolus vulgaris L. to supply ascorbate (ASC) into the cell wall by simple diffusion, a method for calculating plasmalemma diffusional conductivity to ascorbic acid (AA) in intact leaves was evaluated. The core of the approach is that in the presence of a sink for ascorbate in the cell wall, cell wall total ascorbic acid concentration [TAA]cw (=[ASC]cw+[AA]cw) reaches zero at some positive whole‐leaf total ascorbic acid concentration [TAA]l. It is shown that [TAA]l at [TAA]cw=0 is proportional to the sink for ASC in the cell wall and the reciprocal of plasmalemma conductivity. The predicted proportional relationship between [TAA]cw and [TAA]l was confirmed by decreasing TAA levels in leaves through predarkening. Furthermore, increasing the sink intensity for ASC in the cell wall by the acute exposure of leaves to 450 nmol ozone mol?1 during re‐illumination, [TAA]cw reached zero at 2.7‐fold higher [TAA]l than without ozone, and the slope of the relationship increased twofold. Plasmalemma diffusional conductivities to AA of 2.9×10?6 and 1.8×10?6 m s?1, needed to maintain [TAA]cw at the observed level, were calculated from the increase in [TAA]l at [TAA]cw=0 and from the two different estimates of the sink for ASC. A value of 1.3×10?6 m s?1 was calculated on the basis of the oil‐water distribution coefficient for TAA. It is concluded that the demand for ASC in the mesophyll cell wall of the investigated leaves could be met by simple diffusion of AA through the plasmalemma. From the measured increase in the slope of the relationship [TAA]cw versus [TAA]l, an increase in the cell wall pH of 0.3 units was estimated under the influence of ozone.  相似文献
