Absence of actin in the cilia of Tetrahymena pyriformis   总被引:1,自引:0,他引:1  
An extensive electrophoretic analysis of the proteins of highly purified Tetrahymena pyriformis cilia has been undertaken. No component which would specifically bind rabbit muscle myosin could be identified. Furthermore, no peptides from acetone powders of these cilia could be found which co-electrophoresed with rabbit muscle actin. Lastly, the components which most closely resembled actin in molecular weight were quantitated using densitometry and found to represent less than 0.8% of the tubulin in these preparations.  相似文献

Most estrous cycles in cows consist of 2 or 3 waves of follicular activity. Waves of ovarian follicular development comprise the growth of dominant follicles some of which become ovulatory and the others are anovulatory. Ovarian follicular activity in cows during estrous cycle was studied with a special reference to follicular waves and the circulating concentrations of estradiol and progesterone. Transrectal ultrasound examination was carried out during 14 interovulatory intervals in 7 cows. Ovarian follicular activity was recorded together with assessment of serum estradiol and progesterone concentrations. Three-wave versus two-wave interovulatory intervals was observed in 71.4% of cows. The 3-wave interovulatory intervals differed from 2-wave intervals in: 1) earlier emergence of the dominant follicles, 2) longer in length, and 3) shorter interval from emergence to ovulation. There was a progressive increase in follicular size and estradiol production during growth phase of each wave. A drop in estradiol concentration was observed during the static phase of dominant anovulatory follicles. The size of the ovulatory follicle was always greater and produced higher estradiol compared with the anovulatory follicle. In conclusion, there was a predominance of 3-wave follicular activity that was associated with an increase in length of interovulatory intervals. A dominant anovulatory follicle during its static phase may initiate the emergence of a subsequent wave. Follicular size and estradiol concentration may have an important role in controlling follicular development and in determining whether an estrous cycle will have 2 or 3-waves.  相似文献

We have investigated the structural basis for the phenotype of a native rat Slo (rSlo) potassium channel (BK(Ca); KCNMA1) in a rat pituitary cell line, GH(4)C(1). Opposing regulation of these calcium- and voltage-activated potassium channels by cAMP- and cGMP-dependent protein kinases requires an alternatively spliced exon (strex) of 59 amino acids in the cytoplasmic C terminus of the pore-forming alpha subunit encoded by rslo. However, inclusion of this cysteine-rich exon produces a 10-fold increase in the sensitivity of the channels to inhibition by oxidation. Inclusion of the strex exon also increases channel sensitivity to stimulation by calcium, but responses in the physiological ranges of calcium and voltage require coassembly with beta(1) subunits. With strex present, however, beta(1) subunits only stimulated channels assembled from rSlo alpha subunits with a truncated N terminus beginning MDALI-. Thus N-terminal variation and strex exon splicing in rSlo interact to produce BK(Ca) channels with a physiologically relevant phenotype.  相似文献

The O-antigen chain from the lipopolysaccharide of Helicobacter pylori strain UA861 was determined to be composed of an elongated type 2 N - acetyllactosamine backbone, -[-->3)-beta-D-Gal-(1-->4)-beta-D-GlcNAc-(1- ]n-->, with approximately half of the GlcNAc units carrying a terminal alpha-d-Glc residue at the O -6 position. The O-chain of H.pylori UA861 was terminated by a N -acetyllactosamine [beta-D-Gal-(1-->4)-beta-D- GlcNAc] (LacNAc) epitope and did not express terminal Lewis X or Lewis Y blood-group determinants as previously found in other H.pylori strains. The absence of terminal Lewis X and Lewis Y blood-group epitopes and the replacement of Fuc by Glc as a side chain in the O- chain of H.pylori UA861 represents yet another type of lipopolysaccharide structure from H.pylori species. These structural differences in H.pylori lipopolysaccharide molecules carry implications with regard to possible different pathogenic events between strains and respective hosts.   相似文献

The first Australian record of the lily thrips, Liothrips vaneeckei Priesner, is reported from a bulb farm in Warragul South, Victoria. Results of a field survey of southern Wisconsin shallow lakes suggested that watershed (catchment basin) land use has a significant and adverse effect on zooplankton species richness. Zooplankton communities in lakes with no riparian buffer zone, in agriculture-dominated watersheds, contained about half as many species as lakes in least-impact watersheds. In that study, the age of the lake was not taken into account. It is possible that agricultural lakes, often artificial, were so recently-constructed that they had not yet accumulated the equilibrium number of species characteristic of older lakes. In other words, it is possible that the interpretation of the results of the previous study is fatally flawed, if the results were an artifact of lake age, rather than an effect of land use. The major aim of this current study was to determine the ages of agricultural lakes and of lakes in least-impact watersheds, to test for an effect of lake age on zooplankton species richness, using the same sites from the previous study. We used an anova approach to test the null hypothesis that two factors, watershed land use and lake age, had no systematic effect on zooplankton species richness. We determined the age of 35 shallow lakes, using aerial photos, satellite images, and interviews of resource managers and land owners. We identified five artificial agricultural sites and five artificial sites in least-impact prairie watersheds. The artificial sites in this study ranged from 3 to 37 years in age, while natural lakes dated from the melting of the last glacier, about 9500 years ago. Our results suggest, that because artificial lake made up only about a third of the sites, and for the range of lake age and watershed land use, lake age did not have a significant effect on zooplankton species richness, while land use had a highly significant adverse effect. These results pose a larger question for future research. Namely, how quickly do newly-constructed lakes attain the equilibrium number of species seen in the previous study, and what is the quantitative relationship between lake age and zooplankton richness?  相似文献

Achromatopsia is a rare autosomal recessive disorder which shows color blindness, severely impaired visual acuity, and extreme sensitivity to bright light. Mutations in the alpha subunits of the cone cyclic nucleotide-gated channels (CNGA3) are responsible for about 1/4 of achromatopsia in the U.S. and Europe. Here, we test whether gene replacement therapy using an AAV5 vector could restore cone-mediated function and arrest cone degeneration in the cpfl5 mouse, a naturally occurring mouse model of achromatopsia with a CNGA3 mutation. We show that gene therapy leads to significant rescue of cone-mediated ERGs, normal visual acuities and contrast sensitivities. Normal expression and outer segment localization of both M- and S-opsins were maintained in treated retinas. The therapeutic effect of treatment lasted for at least 5 months post-injection. This study is the first demonstration of substantial, relatively long-term restoration of cone-mediated light responsiveness and visual behavior in a naturally occurring mouse model of CNGA3 achromatopsia. The results provide the foundation for development of an AAV5-based gene therapy trial for human CNGA3 achromatopsia.  相似文献
