Six magnetic polarity sections have been established over the Potwar Plateau region of Pakistan, including the major stratotypes of the Siwalik Group. In all six sections the dominant feature of the magnetic polarity stratigraphy is a long normal polarity zone, which is contained within the Nagri Formation. This conspicuous normal polarity zone has been radiometrically dated at 9.5 ± 0.6 m.y., which identifies it as magnetic Chron 9. Radiometric dates from the Upper Siwalik Formation have also been used to identify the Chron 2–3 boundary in two of the sections. The magnetic polarity stratigraphy of three of the sections has been correlated securely with the accepted magnetic polarity time scale, so that the ages of the local stratigraphy are indexed accordingly. Based exclusively on data from stratotype sections, the Chinji, Nagri and Dhok Pathan Formations have nominal age ranges of 10.1–13.1, 7.9–10.1 and 5.1–7.9 m.y. Age fluctuations on the order of 105 years may be anticipated for these formational boundaries within radius of some 20 km of the designated stratotype. Mean sediment accumulation rates during the Chinji, Nagri and Dhok Pathan interval range from 13 to 52 cm/103 yr.Essentially linear sediment accumulations are locally maintained over time intervals of several million years. The Chinji-Nagri lithofacies boundary marks a transition from slow to faster sediment accumulation over much of the Potwar Plateau, indicating a fundamental sedimentary-tectonic change at this time.  相似文献

Replication initiator proteins (Reps) from the HUH-endonuclease superfamily process specific single-stranded DNA (ssDNA) sequences to initiate rolling circle/hairpin replication in viruses, such as crop ravaging geminiviruses and human disease causing parvoviruses. In biotechnology contexts, Reps are the basis for HUH-tag bioconjugation and a critical adeno-associated virus genome integration tool. We solved the first co-crystal structures of Reps complexed to ssDNA, revealing a key motif for conferring sequence specificity and for anchoring a bent DNA architecture. In combination, we developed a deep sequencing cleavage assay, termed HUH-seq, to interrogate subtleties in Rep specificity and demonstrate how differences can be exploited for multiplexed HUH-tagging. Together, our insights allowed engineering of only four amino acids in a Rep chimera to predictably alter sequence specificity. These results have important implications for modulating viral infections, developing Rep-based genomic integration tools, and enabling massively parallel HUH-tag barcoding and bioconjugation applications.  相似文献

Wild male house mice Mus musculus domesticus were collected from the hybrid zone between the John o'Groats race (2n = 32) and the standard race (2n = 40) in northern Scotland. Meiosis in both homozygotes (2n = 32, 36, and 40) and single Robertsonian heterozygotes (2n = 33, 35, and 37) was found to be orderly. At prophase/metaphase I in heterozygotes, a trivalent was formed from the metacentric and two homologous acrocentrics. At pachytene, this trivalent usually had a single side arm at the position of the centromeres, as a result of nonhomologous pairing of the acrocentrics. This side arm persisted into diplotene. Generally only a single chiasma was formed between each acrocentric and the metacentric. Anaphase I nondisjunction frequencies were estimated as 1.5% for the homozygotes and 2.7% for the heterozygotes. The extent of germ cell death between the pachytene and round spermatid stages was 18% greater in heterozygotes than in homozygotes. Our results concur with previous studies which indicate that single Robertsonian heterozygotes in wild house mice have near-normal fertility.  相似文献

A coordinated study of water chemistry, sediment mineralogy, and sediment microbial community was conducted on four >73°C springs in the northwestern Great Basin. Despite generally similar chemistry and mineralogy, springs with short residence time (~5–20 min) were rich in reduced chemistry, whereas springs with long residence time (>1 day) accumulated oxygen and oxidized nitrogen species. The presence of oxygen suggested that aerobic metabolisms prevail in the water and surface sediment. However, Gibbs free energy calculations using empirical chemistry data suggested that several inorganic electron donors were similarly favorable. Analysis of 298 bacterial 16S rDNAs identified 36 species-level phylotypes, 14 of which failed to affiliate with cultivated phyla. Highly represented phylotypes included Thermus, Thermotoga, a member of candidate phylum OP1, and two deeply branching Chloroflexi. The 276 archaeal 16S rDNAs represented 28 phylotypes, most of which were Crenarchaeota unrelated to the Thermoprotei. The most abundant archaeal phylotype was closely related to “Candidatus Nitrosocaldus yellowstonii”, suggesting a role for ammonia oxidation in primary production; however, few other phylotypes could be linked with energy calculations because phylotypes were either related to chemoorganotrophs or were unrelated to known organisms.
