The affinity of yeast glycogen synthetase to glycogen modified by crosslinking has been studied under various experimental conditions. It was found that the higher the degree of crosslinking, the lower the affinity of glycogen synthetase to glycogen. The amount of glycogen synthetase adsorbed from the solution depends on the amount of crosslinked glycogen added and is inversely proportional to the concentration of the soluble glycogen. The stability of the complex formed between yeast glycogen synthetase and the crosslinked glycogen was found to be maximal at neutral pH range. The presence of glucose 6-phosphate, uridine 5′-di-phosphate, and uridine 5′-diphosphate glucose enhanced the stability of the complex. 相似文献

Exercise is associated with release of catecholamines and vasoactive intestinal polypeptides. Recurrent exposure to catecholamines modifies the sensitivity of adrenoceptors. To test the hypothesis that exercise training may affect the sensitivity of the epicardial coronary arteries, we performed studies on isolated coronary arteries from male dogs capable of running on a treadmill. The animals were separated randomly into two groups: sedentary and exercise training. After 11 wk, rings of left circumflex and left anterior descending coronary arteries were studied in vitro. Contractions to alpha 1-adrenergic agonists (norepinephrine and phenylephrine) were not affected by exercise training. During contractions with prostaglandin F2 alpha, endothelium-dependent relaxations to alpha 2-adrenergic agonists (norepinephrine and UK 14304) were not reduced significantly by exercise training. The concentration-relaxation curves to beta-adrenergic agonists (norepinephrine, isoproterenol, and epinephrine) were shifted to the right after training. The concentration-response curves to vasoactive intestinal polypeptide, but not that to substance P, were shifted to the right in rings with endothelium from exercise-trained animals. These findings demonstrate a decrease in responsiveness of canine vascular smooth muscle to beta-adrenergic agonists and to vasoactive intestinal polypeptide after exercise training. 相似文献

Small resting B lymphocytes were highly enriched and completely depleted of all preactivated large B lymphocytes using countercurrent centrifugal elutriation and free flow electrophoresis. They required T lymphocytes, monocytes, and a mitogen to produce antibodies after 5 days of preincubation. Large activated B lymphocytes were obtained in cell fractions which were free of resting ones. They produced antibodies even in the absence of a mitogen. Two groups were distinguished, differing in their stage of differentiation and their negative surface charge density. The cells of one group had an electrophoretic mobility (EM) like resting B lymphocytes ranging from 0.85 to 0.99 X 10(-4) (cm2 V-1 s-1). They took 2 to 3 days of preincubation before they started to secrete antibodies. Interleukin 2 and pokeweed mitogen enhanced their antibody production capability. The cells of the other group had an EM between 0.99 and 1.13 X 10(-4) (cm2 V-1 s-1). They secreted antibodies even during the first day of incubation. The quantity of the antibodies which they produced depended only on the blood donor. It could not be influenced by a mitogen or by interleukin 2. The study shows that large B lymphocytes with high negative surface charge density are in a later maturation stage than those with lower negative surface charge density. 相似文献

Observations on functional morphology and results from experiments demonstrate that setiferous epipods compose the major gill-cleaning mechanism in a penaeoid shrimp, Rimapenaeus similis. Epipods on the second maxillipeds and on pereopods 1-3 are equipped with long setae bearing an array of digitate scale setules. These multidenticulate setae reach to most gills and are jostled among them during limb movements. Experiments were performed in which epipods were removed from the gill chamber on one side (experimental) but not the other (control); treated animals were exposed to fouling in a recirculating water system for 2 weeks. Particulate fouling, measured by reduction in relative gill transparency, was significantly greater on experimental than control gills. The pereopodal exopods, not previously implicated in gill cleaning in any decapod, were similarly identified as important gill-cleaning structures. Equipped with long multidenticulate setae like those on the epipods, exopods sweep back and forth over the gill filaments just under the gill cover, areas not reached by the epipods. Exopod-ablation experiments were conducted that showed that exopods prevent particulate fouling on gill surfaces over which they sweep. The similarity in action of the passive gill-cleaning system of R. similis to that of crayfish (Bauer [1998] Invert Biol 117:29-143) suggests the hypothesis that the epipodal and exopodal cleaning setae of R. similis are ineffective against epibionts. The reduction in epipodal and exopodal cleaning systems that occurs in the Penaeoidea is hypothesized to be compensated for by increased development of gill-cleaning setae on the branchiostegite, scaphognathite, or other structures. 相似文献
