Extensively ball-milled cellulose fibers were used as natural substrate for the determination of cellulase activity. This physical treatment breaks the large cellulose fibers to small but insoluble particles yielding a substrate accessible for complete enzymatic breakdown. The parameters studied to estimate the activity of cellulases were (a) the decrease in optical density of ball-milled suspension of fibers and (b) simultaneous measurement of liberated sugars during hydrolysis. A good correlation was found between the initial rate of reaction and the amount of sugar released at given times.  相似文献

The results of experiments on the infection of many different plants with seven biologic strains of known history of the nematode, Anguillulina dipsaci (Kühn), are given, together with the results obtained by other investigators. The technique by which the nematode was induced to infect other host plants is described in detail. The biologic strain theory in relation to A. dipsaci is discussed, together with the merits of the trinomial system of nomenclature in connexion with unspecialized races. The factors concerned with nematode outbreaks receive consideration. The relationship that exists between flower colour in herbaceous phloxes and the degree of infection by A. dipsaci is presented.  相似文献

Cells from cranial and spinal arachnoid membranes of humans were grown in culture. Their growth characteristics, morphology and details of their cytoskeletal composition are described. Arachnoid membranes, obtained at autopsy, were finely minced and incubated in tissue culture medium. Monolayers of cells of homogeneous morphology grew from these tissue fragments. The cells were flat and polygonal. They divided slowly to form non-overlapping monolayers of low cell density. Electron microscopic examination of cultured arachnoid cells revealed numerous desmosome-like tight junctions and abundant intermediate filaments (tonofilaments). Both morphological features are characteristic of arachnoid cells in situ, but not of cells in the fibroblast-rich dura mater. Immunofluorescence microscopy with monoclonal antibodies demonstrated cytokeratin in the cytoplasm of primary cultures of arachnoid cells. Thus we demonstrated that these cultured cells retained certain of the specific differentiated properties of arachnoid cells in situ and that they are not fibroblasts (which lack tight junctions and cytokeratins). To our knowledge, there have been no previous reports of in vitro growth of arachnoid cells. This in vitro model should be useful in studying the response of arachnoid cells to a variety of substances thought to be involved in the chronic inflammatory condition of the meninges known as arachnoiditis.  相似文献

Complexity and stability are the ubiquitous characteristics of all biological phenomena. The theoretical suggestions presented here acknowledge this and attempt to capitalize on the property of selectiveness that complexity induces. It is from this that a belief in the existence of selection rules emerges. What is required of a viable theory is a linguistic means of maneuvering known information so that these selection or limitation rules may be obtained, and thence, by their manipulation, to generate the properties exhibited by biological systems.  相似文献

The de Broglie wave equation has been applied to the study of amoeboid motility. This leads to precise predictions of wavelengths displayed by the cellular membrane during motility. A fundamental challenge in robotics today is building robots that can learn new skills by observing humans and imitating human actions. We propose a new Bayesian approach to robotic learning by imitation inspired by the developmental hypothesis that children use self-experience to bootstrap the process of intention recognition and goal-based imitation. Our approach allows an autonomous agent to: (i) learn probabilistic models of actions through self-discovery and experience, (ii) utilize these learned models for inferring the goals of human actions, and (iii) perform goal-based imitation for robotic learning and human-robot collaboration. Such an approach allows a robot to leverage its increasing repertoire of learned behaviors to interpret increasingly complex human actions and use the inferred goals for imitation, even when the robot has very different actuators from humans. We demonstrate our approach using two different scenarios: (i) a simulated robot that learns human-like gaze following behavior, and (ii) a robot that learns to imitate human actions in a tabletop organization task. In both cases, the agent learns a probabilistic model of its own actions, and uses this model for goal inference and goal-based imitation. We also show that the robotic agent can use its probabilistic model to seek human assistance when it recognizes that its inferred actions are too uncertain, risky, or impossible to perform, thereby opening the door to human-robot collaboration.  相似文献
