Here we present a brief conception of biosensors. Structural peculiarities and properties of single- and double-stranded nucleic acids that are to be taken into account when creating biosensors on the basis of these biomolecules are considered. A novel technique for the assessment of cytotoxic capacity, which is a characteristic of the state of immune-system cells, has been proposed. Using coherent phase microscopy, perforin granules have been detected and described in the cytoplasm of live CD8+ lymphocytes. According to the analysis of phase images, the parameters of perforin granules in the cell were quantitatively assessed. Their average values were calculated as follows: the projected area on the image plane, 15 ± 7 μm2; phase volume, 0.94 ± 0.24 μm3; and the dry weight of the granules, 0.52 ± 0.11 pg. Taking the advantage of this technique, it appears possible to determine cytotoxic capacity using a small number of live cells without employing fluorescent-labeled probes. 相似文献

A thermodynamic model of the formation of bridges consisting of alternating daunomycin molecules and copper ions and connecting neighboring nucleic acid molecules in a particle of a liquid crystalline dispersion was constructed. The model is based on the conception that ligands are adsorbed on lattices of reaction centers which are formed in a liquid crystal at a particular spatial arrangement of adjacent nucleic acid molecules (phasing). Equations were derived that describe typical experimentally obtained S-shaped dependences of bridge concentration on the concentration of copper ions and daunomycin molecules in an initial solution. It was shown that dependences of this kind take place in two variants of the adsorption model: when the binding of daunomycin with adjacent nucleic acid molecules is considered to be independent on the formation of bridges and when bridges compete with single daunomycin molecules for the sites on adjacent nucleic acid molecules. 相似文献

The binding of chitosan molecules to DNA in solutions of different ionic strength has been studied. The data were analyzed in terms of the model of ion condensation and the thermodynamic theory of the binding of protracted ligands to DNA. Combining these approaches made it possible to estimate the sterical and energetic characteristics of chitosan-DNA binding and establish the dependence of the chitosan-DNA binding constant on the ionic strength of solution. 相似文献

The typology of temporary waterbodies in Saratov oblast has developed with a similarity in the composition of zooplankton
species that serve as its foundation. Seven waterbody groups have been distinguished: waterbodies of flood lands, river valley
terraces, steppe watersheds, brackish lagoons, brackish lagoons connected to irrigation networks, excavated ponds, and puddles.
The specificity of zooplankton species composition in different types of temporary waterbodies is shown. A significant influence
of waterbody lifetime on the number of zooplankton species has been revealed, with the waterbody capacity and surface area

A new series of potent 8-hydroxyquinolines was designed based on the newly resolved X-ray crystal structure of EGLN-1. Both alkyl and aryl 8-hydroxyquinoline-7-carboxyamides were good HIF-1alpha prolyl hydroxylase (EGLN) inhibitors. In subsequent VEGF induction assays, these exhibited potent VEGF activity. Ubiquitin has been used in protein expression for enhancing yields and biological activities of recombinant proteins. Biotin binds tightly and specifically to avidin and has been widely utilized as a tag for protein purification and monitoring. Here, we report a versatile system that takes the advantages of both biotin and ubiquitin for protein expression, purification, and monitoring. The tripartite system contained coding sequences for a leader biotinylation peptide, ubiquitin, and biotin holoenzyme synthetase in two reading frames under the control of T7 promoter. The expression and purification of several large mammalian enzymes as biotin-ubiquitin fusions were accomplished including human ubiquitin activating enzyme, SUMO activating enzymes, and aspartyl-tRNA synthetase. Expressed proteins were purified by one-step affinity column chromatography on monomeric avidin columns and purified proteins exhibited active function. Additionally, the ubiquitin protein hydrolase UBP41, expressed and purified as biotin-UBP41, efficiently and specifically cleaved off the biotin-ubiquitin tag from biotin-ubiquitin fusions to produce unmodified proteins. The present expression system should be useful for the expression, purification, and functional characterization of mammalian proteins and the construction of protein microarrays. 相似文献
