Multigene transformation (MGT) is becoming routine in plant biotechnology as researchers seek to generate more complex and ambitious phenotypes in transgenic plants. Every nuclear transgene requires its own promoter, so when coordinated expression is required, the introduction of multiple genes leads inevitably to two opposing strategies: different promoters may be used for each transgene, or the same promoter may be used over and over again. In the former case, there may be a shortage of different promoters with matching activities, but repetitious promoter use may in some cases have a negative impact on transgene stability and expression. Using illustrative case studies, we discuss promoter deployment strategies in transgenic plants that increase the likelihood of successful and stable multiple transgene expression. 相似文献

Runx1 is highly expressed in chondroprogenitor and osteoprogenitor cells and in vitro experiments suggest that Runx1 is important in the early stages of osteoblast and chondrocyte differentiation. However, because Runx1 knockout mice are early embryonic lethal due to failure of hematopoiesis, the role of Runx1 in skeletogenesis remains unclear. We studied the role of Runx1 in skeletal development using a Runx1 reversible knockout mouse model. By crossing with Tie2-Cre deletor mice, Runx1 expression was selectively rescued in the endothelial and hematopoietic systems but not in the skeleton. Although Runx1Re/Re embryos survived until birth and had a generally normal skeleton, the development of mineralization in the sternum and some skull elements was significantly disrupted. In contrast to wild-type embryos, the sternum of E17.5 Runx1Re/Re embryos showed high levels of Sox-9 and collagen type II expression and lack of development of hypertrophic chondrocytes. In situ hybridization analysis demonstrated that, in contrast to the vertebrae and long bones, the sternum of wild-type embryos expresses high levels of Runx1, but not Runx2, the master regulator of skeletogenesis. Thus, although Runx1 is not essential for major skeletal development, it does play an essential role in the development of the sternum and some skull elements. 相似文献

The habituation of soybean (Glycine max) callus can be induced rapidly, by exposing the tissue to small amounts (10−9molar) of compounds including 2.4-dinitrophenol and phenoxyisobutyric acid for brief periods of time. Such compounds reportedly exhibit antiauxin activity. Various soybean callus phenotypes have been isolated which continue to exhibit hormone habituation 14 months following the initiation of the experiment. Protein changes in habituated tissue under selected hormonal regimes were detected indicating changes at the level of gene expression. Habituated tissue exhibits hormonal autonomy in a manner similar to crown gall tissue, suggesting that such studies may help elucidate the mechanism of induction of crown gall disease and genetic transformation by Agrobacterium. 相似文献

Purification and immunological characterization of a novel cytochrome P450 from C3H/10T1/2 cells 总被引:5,自引:0,他引:5
The major polycyclic aromatic hydrocarbon-metabolizing cytochrome P450 in the mouse embryo fibroblast-derived C3H/10T1/2 CL8 cell line (P450-EF) has been partially purified from benz[a]anthracene (BA)-induced 10T1/2 cells (40 pmol P450/mg). The purification of P450-EF was carried out by sequential chromatography of solubilized microsomes over hydrophobic aminohexyl-Sepharose 4B, anion exchange DE-52 cellulose, and cation exchange carboxymethyl trisacryl columns. The final preparation (1700 pmol/mg) appeared as a single major 55-kDa band by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Reconstitution of detergent-free partially purified P450-EF yielded a relatively high turnover for 7,12-dimethylbenz[a]anthracene (DMBA) metabolism (5.4 nmol/nmol/min). Polyclonal antibodies to purified P450-EF (anti-P450-EF), raised in, respectively, rabbit and chicken, detected a single 55-kDa band in 10T1/2 cell microsomes that was highly inducible by BA (approximately 20-fold) and TCDD (approximately 5-fold). Rabbit anti-P450-EF was much more effective than the corresponding chicken antibody at binding denatured P450-EF protein on Western blots. Conversely, only the chicken antibody was effective at inhibiting DMBA metabolism catalyzed by microsomal P450-EF. This antibody did not inhibit P450IA1-mediated DMBA metabolism. Rabbit anti-P450-EF recognized very weakly (less than 1% of homologous protein response) pure P450IA1, IIB1, IIC7, IIE1, and IIIA1 proteins on Western blots but exhibited substantial cross-reactivity (approximately 10%) with pure P450IIA1 and very strong cross-reactivity (approximately 75%) with a hormonally regulated rat adrenal P450. Polyclonal antibodies to several major P450 subfamilies either did not recognize P450-EF (anti-P450IA, IIB, and IIC) or recognized it very weakly (anti-P450IIA1). Carolight® is a transgenic maize variety that accumulates extraordinary levels of carotenoids, including those with vitamin A activity. The development of Carolight® maize involved the technical implementation of a novel combinatorial transformation method, followed by rigorous testing for transgene expression and the accumulation of different carotenoid molecules. Carolight® was envisaged as a way to improve the nutritional health of human populations that cannot access a diverse diet, but this ultimate humanitarian application can only be achieved after extensive testing for safety, agronomic performance and nutritional sufficiency. In this article, we chart the history of Carolight® maize focusing on its development, extensive field testing for agronomic performance and resistance to pests and pathogens, and feeding trials to analyze its impact on farm animals (and their meat/dairy products) as well as animal models of human diseases. We also describe more advanced versions of Carolight® endowed with pest-resistance traits, and other carotenoid-enhanced maize varieties originating from the same series of initial transformation experiments. Finally we discuss the further steps required before Carolight® can fulfil its humanitarian objectives, including the intellectual property and regulatory constraints that lie in its path. 相似文献

Energy storage challenges have triggered growing interest in various battery technologies and electrocatalysis. As a particularly promising variety, the Li–O2 battery with an extremely high energy density is of great significance, offering tremendous opportunities to improve cell performance via understanding catalytic mechanisms and the exploration of new materials. Furthermore, focus on nonaqueous electrolyte‐based Li–O2 batteries has markedly intensified since there could be a higher probability of commercialization, compared to that of solid‐state or aqueous electrolytes. The recent advancements of the nonaqueous Li–O2 battery in terms of fundamental understanding and material challenges, including electrolyte stability, water effect, and noncarbon cathode materials are summarized in this review. Further, the current status of water impact on discharge products, possible mechanisms, and parasitic reactions in nonaqueous electrolytes are reviewed for the first time. The key challenges of noncarbon oxygen electrode materials, such as noble metals and metal oxides‐based cathodes, transition metals, transition metal compounds (carbides, oxides) based cathodes as well as noncarbon supported catalysts are discussed.
