Summary During the autumn migratory period a juvenile Griffon Vulture was followed by satellite tracking from the breeding grounds in the Pyrenees to the area of Valencia. Satellite tracking was performed on the basis of a Toyocom transmitter connected to the Argos system. The bird could be traced over a two-month period from end of September to end of November 1990. After an initial south-ward movement the bird stayed in an arid lowland close to the Ebro valley for a longer period and later again moved southward to an area near Valencia. Before the transmitter's lifetime ended due to battery insuffiency the bird had turned north into an area where Griffon Vultures are common. The experimental bird may have joined a large group of conspecifics, wintering there. The bird could be traced over a total distance of about 2000 km and the longest stage per day was about 80 km. As in the case of another European pilot study that we have recently carried out on Bewick's Swans, satellite tracking of a Griffon Vulture proved to be a highly promising method to examine in detail migratory movements as well as ecological aspects on route.  相似文献

An interesting property of certain peptides presented by major histocompatibility complex (MHC) molecules is their acquisition of a dual binding mode within the peptide binding groove. Using x-ray crystallography at 1.4 A resolution, we show here that the glucagon receptor-derived self-peptide pGR ((412)RRRWHRWRL(420)) is presented by the disease-associated human MHC class I subtype HLA-B*2705 in a dual conformation as well, with the middle of the peptide bent toward the floor of the peptide binding groove of the molecule in both binding modes. The conformations of pGR are compared here with those of another self-peptide (pVIPR, RRKWRRWHL) that is also displayed in two binding modes by HLA-B*2705 antigens and with that of the viral peptide pLMP2 (RRRWRRLTV). Conserved structural features suggest that the N-terminal halves of the peptides are crucial in allowing cytotoxic T lymphocyte (CTL) cross-reactivity. In addition, an analysis of T cell receptors (TCRs) from pGR- or pVIPR-directed, HLA-B27-restricted CTL clones demonstrates that TCR from distinct clones but with comparable reactivity may share CDR3alpha but not CDR3beta regions. Therefore, the cross-reactivity of these CTLs depends on TCR-CDR3alpha, is modulated by TCR-CDR3beta sequences, and is ultimately a consequence of the conformational dimorphism that characterizes binding of the self-peptides to HLA-B*2705. These results lend support to the concept that conformational dimorphisms of MHC class I-bound peptides might be connected with the occurrence of self-reactive CTL.  相似文献

Dendritic cells (DC) generated from human umbilical cord blood might replace patients' DC in attempts to elicit tumor-specific immune response in cancer patients. We studied the efficiency of transfection of human cord blood DC with plasmid DNA carrying the enhanced version of green fluorescent protein (EGFP) as a reporter gene, to test if nonviral gene transfer would be a method to load DC with protein antigens for immunotherapy purposes. Cord blood mononuclear cells were cultured in serum-free medium in the presence of granulocyte-monocyte colony stimulating factor (GM-CSF), stem cell factor (SCF) and Flt-3 ligand (FL), to generate DC from their precursors, and thereafter transfected by electroporation. Maturation of DC was induced by stimulation with GM-CSF, SCF, FL and phorbol myristate acetate (PMA). Transfected DC strongly expressed EGFP, but transfection efficiency of DC, defined as HLA-DR(+) cells lacking lineage-specific markers, did not exceed 2.5%. Expression of the reporter gene was also demonstrated in the DC generated from transfected, purified CD34(+) cord blood cells, by stimulation with GM-CSF, SCF, FL, and tumor necrosis factor alpha (TNF-alpha). Transfection of CD34(+) cells was very efficient, but proliferation of the transfected cells was much reduced as compared to the untransfected cells. Therefore, the yield of transgene-expressing DC was relatively low. In conclusion, nonviral transfection of cord blood DC proved feasible, but considering the requirements for immunotherapy in cancer patients, transfection of differentiated DC or generation of DC from transfected hematopoietic stem cells provide only a limited number of DC expressing the transgene.  相似文献

We present a novel approach to the analysis of fluctuations in human myoelectrical gastric activity measured noninvasively from the surface of the abdomen. The time intervals between successive maxima of the wavelet transformed quasi-periodic electrogastrographic waveform define the gastric rate variability (GRV) time series. By using the method of average wavelet coefficients, the statistical fluctuations in the GRV signal in healthy individuals are determined to scale in time. Such scaling was previously found in a variety of physiological phenomena, all of which support the hypothesis that physiological dynamics utilize fractal time series. We determine the scaling index in a cohort of 17 healthy individuals to be 0.80 +/- 0.14, which compared with a set of surrogate data is found to be significant at the level P < 0.01. We also determined that the dynamical pattern, so evident in the spectrum of average wavelet coefficients of the GRV time series of healthy individuals, is significantly reduced in a cohort of systemic sclerosis patients having a scaling index 0.64 +/- 0.17. These results imply that the long-term memory in GRV time series is significantly reduced from healthy individuals to those with systemic sclerosis. Consequently, this disease degrades the complexity of the underlying gastrointestinal control system and this degradation is manifest in the loss of scaling in the GRV time series.  相似文献

Abstract. This paper presents results of a long-term study on natural forest dynamics in the Bia?owieza National Park (BNP), northeastern Poland. Five permanent sample areas were used, each consisting of a transect of varying width (40 - 60 m) and length (200 - 1380 m). The total sample area is 14.9 ha. The study covers the period 1936–1992. During this period measurements were made on five occasions at approximately 10-yr intervals. On each measurement date all trees with DBH > 5 cm were identified and their spatial location, diameter, crown condition and position in the canopy determined. During the study period the stands underwent noticeable changes, mainly in terms of tree species composition. The major change was a quantitative increase of the late-successional species: Tilia cordata and Carpinus betulus, also to a lesser degree Fraxinus excelsior and, in the last period, of the early successional Alnus glutinosa. Declining species included both early- and late-succession species. Among the latter group, Picea abies ranked first. This species lost much of its importance during the last few decades. P. abies was followed by Pinus sylvestris which is an important component of the climax vegetation under the conditions prevailing in Bialowieza, at least on more oligotrophic sites. Still, this species has not been able to regenerate during the whole study period. Some other late-succession species, Acer platanoides and Quercus robur, were also amongst the declining species. Although the basal area of Q. robur increased, its population was getting older and the process of natural regeneration was markedly impeded. All typical pioneer, short-lived species: Betula pendula and B. pubescens, Salix caprea and Populus tremula also decreased, which was probably caused by a lack of major disturbances during the study period. In general, the results obtained for the semi-natural conditions of Bialowieza during the 56-yr observation period suggest a rather high compositional instability of the forest stands there. A more precise identification of the role of particular factors in the observed stand dynamics is difficult because of the paucity of appropriate historical and environmental data which refer directly to the study plots; moreover, the data are generally incompatible and of different resolution.  相似文献

This study investigated the problem of the influence of 50 Hz magnetic field (MF) on human heart rate variability (HRV). The exposure system was a commercial device for magnetotherapy, generating field of the strength of 500 microT at the center of the coil, 150-200 microT at the position of human subjects' heart and 20-30 microT at the position of subjects' head. The exposure protocols, applied randomly, were either "half hour MF-off/half hour MF-on" or "half hour MF-off/half hour MF-off." The phonocardiographic (PhCG) signal of 15 volunteers were obtained during exposure and used for calculation of time-domain HRV parameters (mean time between heart beats (N-N), standard deviation of time between heart beats (SDNN), and the number of differences of successive beat-to-beat intervals greater than 50 ms, divided by the total number of beat-to-beat intervals (pNN50)) and nonlinear HRV measures (approximate entropy (ApEn), detrended fluctuation scaling exponents). The protocol MF-off/MF-on was applied in nine subjects. Repeated measures ANOVA (RMANOVA) performed for Mf-off/MF-off protocol indicated no statistical difference among four 15 min intervals of HRV data (P value >20% for all parameters except for N-N, where P = 3.7%). RMANOVA followed by the post hoc Tukey test performed for Mf-off/MF-on protocol indicated a statistically significant difference during MF on for N-N (8% increase, P <.1%), SDNN (40% increase, P = 1.1%), and pNN50 (110% increase, P <.1%). The results of the analysis indicate that the changes of these parameters could be associated with the influence of MF.  相似文献

Mesenchymal stem cells derived from bone marrow or several fetal tissues can be expanded and differentiated into other cell lines. The fetal liver is the source of early hematopoietic cells and also, as a fetal tissue, may be considered as a source of pluripotent stem cells. The differentiation potential of fetal rat liver cells have been examined. Freshly isolated liver cells from 14-d fetuses were cultured in Dulbecco medium supplemented with 10% FCS. The plastic-adherent cells were then passaged up to 10 times. Freshly isolated cells and cells from every passage were cultured in hematopoiesis-promoting environment that consists of methylcelulose supplemented with FCS, rat IL-3, human IL-6 and Epo. Parallely these cells were incubated in co-culture with rat muscle satellite cells (Dulbecco medium with 10% FCS and 10% HS) to examine their myogenic potential. Culture in methylcelulose resulted in a high number of GM and Mix colonies in case of freshly isolated liver cells and the number of colonies decreased according to the number of passages. In case of cells from 4th passage, there ware no hematopoietic colonies in culture. In contrast--freshly isolated cells were not able to fuse with rat satellite cells and form the myotubes. This ability appeared in plastic-adherent cells just from the second passage and increases to 5th passage. The cells from every next passage up to 10th when co-cultured with satellite cells participated in myotube formation at the same high level. This result may suggest that in the 14-d rat liver there exist at least two subpopulations of cells: the non-adherent hematopoietic cell population, and the population of plastic-adherent cells capable of differentiating into myotubes. Since the attempts to redifferentiate hematopoietic subpopulation into myopoiesis, or myopoietic subpopulation into hematopoiesis failed, it may be concluded that at least under our experimental conditions the fetal liver cells do not reveal the "plasticity" features.  相似文献

The postillumination burst (PIB) of CO2 and light-enhanced dark respiration (LEDR) depending on oxygen concentration, temperature, respiratory substrates and photorespiratory inhibitor aminoacetonitrile (AAN) were investigated in detached leaves of tall fescue (Festuca arundinacea) using a closed circuit system with an infrared gas analyzer. No PIB was observed in 1 % O2 under temperature over the range from 15 °C to 35 °C. The rate of LEDR was about twice as low in 1 % O2 as that in 21 and 50 % O2 under all temperatures applied. The PIB was absent and LEDR decreased at 21 % O2 following illumination of leaves for 1 hour at 1 % O2. When 200 mM glycine or malate solutions were introduced into the leaves of tall fescue, the magnitudes of PIB increased by about 60 and 40 % and rate of LEDR by about 70 % and 40 %, respectively. Pyruvate and succinate were less effective in promotion of PIB and LEDR. AAN had a small stimulatory effect on PIB and LEDR (about 20 % and 10 %, respectively). The dependences between magnitudes of PIB and rates of LEDR were highly correlated (r=0.94). The results presented indicate that atmospheric concentration of oxygen during the period of photosynthesis of tall fescue leaves was necessary not only for occurrence of PIB and LEDR but also for production of substrate(s) (glycine and/or malate) for these phenomena.  相似文献

The antenna proteins in photosystem II (PSII) not only promote energy transfer to the photosynthetic reaction center (RC) but provide also an efficient cation sink to re-reduce chlorophyll a if the electron transfer (ET) from the Mn-cluster is inhibited. Using the newest PSII dimer crystal structure (3.0 A resolution), in which 11 beta-carotene molecules (Car) and 14 lipids are visible in the PSII monomer, we calculated the redox potentials (Em) of one-electron oxidation for all Car (Em(Car)) by solving the Poisson-Boltzmann equation. In each PSII monomer, the D1 protein harbors a previously unlocated Car (CarD1) in van der Waals contact with the chlorin ring of ChlZ(D1). Each CarD1 in the PSII dimer complex is located in the interface between the D1 and CP47 subunits, together with another four Car of the other PSII monomer and several lipid molecules. The proximity of Car bridging between CarD1 and plastoquinone/Q(A) may imply a direct charge recombination of Car+Q(A)-. The calculated Em(CarD1) and Em(ChlZ(D1)) are, respectively, 83 and 126 mV higher than Em(CarD2) and Em(ChlZ(D2)), which could explain why CarD2+ and ChlZ(D2)+ are observed rather than the corresponding CarD1+ and ChlZ(D1)+.  相似文献

The effect of Cd and Pb on endogenous and IAA-induced elongation growth and medium pH of maize coleoptile segments incubated at 20, 25 and 30 °C was studied. It was found that the elongation of coleoptile segments and proton extrusion increased with the temperature and reached its maximum at 30 °C. For Cd, the maximal inhibition of endogenous and IAA-induced growth as well as medium acidification of coleoptile segments was observed at 25 °C. Meanwhile, Pb, irrespective of the temperature, diminished the growth of the segments by ca. 20%, increasing the acidification of the incubation medium. It was also found that in contrast to Cd, Pb accumulation in maize coleoptile segments did not correlate with temperature. The results suggest that the toxic effect of Cd on elongation growth of coleoptile segments is connected with the decrease of the PM H(+)-ATPase activity and probably with Cd-induced high acivity of IAA oxidase, whereas the effect of Pb did not depend on activity of any of the enzymes.  相似文献
