Simultaneous immunohistochemical demonstration of antigen expression and 5-bromodeoxyuridine incorporation in plastic embedded sections

Summary In this study a double immunohistochemical staining procedure is described for the simultaneous demonstration of antigen expressing cells and replicating cells in rat thymus. As markers for cell surface antigen expression a monoclonal antibody against Ia-expressing cells (His 19) and a monoclonal antibody against cells of the monocyte-macrophage lineage (ED2) were used. Replicating cells were demonstrated by the incorporation of 5-bromodeoxyuridine (BrdUrd). Tissue pieces were fixed in a periodate-lysine-paraformaldehyde fixative and embedded in glycol methacrylate. To demonstrate Ia-expressing cells or ED2-positive macrophages in plastic embedded sections a digestion with trypsin is necessary. The staining procedure was applied sequentially and was performed with a peroxidase and an alkaline phosphatase labeled reagent yielding respectively a brown and a blue reaction product. Results with this staining procedure on plastic embedded sections of rat thymus, an organ with a high DNA synthesizing capacity, showed incorporation of BrdUrd predominantly in the cortex. ED2-positive macrophages were only found in the cortex. The la-positive epithelial reticular cells demonstrated extremely well their stellate form.     

Hypotensive activity of serotonin antagonists; correlation with alpha 1-adrenoceptor and serotonin receptor blockade

For a series of 12 serotonin antagonists, largely varying in potency, the decrease in diastolic pressure was determined after intravenous injection into pentobarbitone-anaesthetized normotensive rats. The hypotensive activity of these antagonists was correlated with their affinity for alpha 1-adrenoceptors, established by [3H]prazosin radioligand displacement, and the 5-HT2 serotonergic receptor, determined by inhibition of specific [3H]mianserin binding. The radioligand binding assays were performed since they correspond to the in vivo antagonistic potencies of the antagonists at alpha 1--and 5-HT2-receptors, respectively. A close correlation (r = 0.963) was found between the affinity for alpha 1-adrenoceptors and hypotensive activity. On the other hand, a negative correlation of lower statistical quality (r = -0.808) existed between the affinity for 5-HT2-receptors and the depressor potency. In this series of 12 compounds, the new antihypertensive drug ketanserin is included for which it has been speculated that it lowers blood pressure by virtue of its serotonin antagonistic activity. The results of the present study, however, point towards alpha 1-adrenolytic potency as an important mechanism in the hypotensive action of the drug.     

Computer analysis of chromatin arrangement and nuclear texture in follicular thyroid tumours

The differentiation of the thyroid glands follicular neoplasias into adenomas and carcinomas is currently done using the histological criteria recommended by WHO. This pilot study of 10 human follicular carcinomas and 10 folliculars adenomas demonstrates the possibility of a cytological classification using digital picture processing of high resolution cell images. Giemsa stained paraplast sections were scanned with a Colour-TV-camera, different channels were used with respect to staining and analyzing methods and computed with an image processing system. The computer aided cytophotometric methods detected significant differences in the chromatin arrangement and structure.

The lists do not claim completeness, but have been intended to provide a working platform for further listings and for comparison with data published earlier.     

Bovine and ovine leukemia viruses. I. Characterization of viral antigens.

A comparative study on several virus-specific antigens of bovine and ovine leukemia viruses is presented. In addition to the major immunologically reactive, nonglycosylated antigen p21, which has a molecular weight of 21,000, both viruses share two glycoproteins of apparent molecular weights of 60,000 (gp60) and 32,000 (gp32), respectively. Immunological cross-reaction suggests that ovine and bovine leukemia viruses are genetically highly related.
