Vinblastine (VLB) is an antineoplastic agent contained in leaves of Catharanthus roseus. One-pot synthesis of VLB from vindoline and catharanthine, biosynthetic precursors in the plant, was achieved under biomimetic conditions. FMN-mediated coupling of vindoline and catharanthine under irradiation with near-ultraviolet light was followed by NAD(P)H-mediated direct conversion of the coupling product to VLB in the dark. The yield of VLB was governed by the level of NAD(P)H added, and the highest yield was obtained by incubation with 10 mM NAD(P)H for 5 h. It is postulated, as one concept of VLB biosynthesis, that similar non-enzymic VLB synthesis may occur in the plant leaves. 相似文献

Myeloperoxidase (MPO)-generated halogenating molecules, such as hypochlorous acid and hypobromous acid (HOBr), in inflammatory regions are postulated to contribute to disease progression. In this study, we showed that ergothioneine (EGT), derived from an edible mushroom, inhibited MPO activity as well as the formation of 8-bromo-2′-deoxyguanosine in vitro. The HOBr scavenging effect of EGT is higher than those of ascorbic acid and glutathione. We initially observed that the administration of Coprinus comatus, an edible mushroom containing a high amount of EGT, inhibited the UV-B-induced inflammatory responses and DNA halogenation, suggesting that EGT is a promising anti-inflammatory agent from mushrooms. 相似文献

Background
Uterine serous endometrial intraepithelial carcinoma (SEIC) is an immediate precursor of invasive carcinoma. The majority of stage IA SEICs are curable, but those with latent peritoneal metastasis and/or capillary lymphatics invasion may have poor prognoses Careful pathologic staging is thus needed to predict the risk of recurrence and to determine postoperative therapeutic strategies.Case Presentation
A 71-year-old woman was hospitalized for the treatment of peritoneal carcinoma. She had undergone total hysterectomy and bilateral salpingo-oophorectomy due to SEIC (stage IA) at age 63 years, and had received medical check-ups every year since. Elevated serum CA125 (184 U/mL) was detected for the first time 8 years after surgery. A thorough workup revealed no potential primary lesion other than that in the peritoneum. Tumor reduction surgery was performed. Histologic analysis of the peritoneal lesion was high-grade serous carcinoma. The peritoneal carcinoma was diffusely immunostained for p53; thus, possible recurrence of SEIC was suspected. Tumor DNAs were microdissected from the uterine and peritoneal lesions and p53 mutation analysis was done. SEIC and peritoneal carcinomas had distinct p53 mutations that were mutually exclusive.Conclusions
The present case raised a concern about the difficulty of histologic staging for SEICs. Although SEICs confined to the uterine endometrium in most cases predict a good prognosis, microscopic metastasis to the peritoneum may not be detectable at hysterectomy. If secondary malignancies of a serous phenotype develop years later, comprehensive reexamination of SEIC is mandated, with the help of DNA analysis.106.

Human immunodeficiency virus (HIV) exhibits immunological hypervariability, which has been an obstacle to successful production of effective anti-HIV vaccines. In this study, we estimated patterns of nucleotide and amino acid substitutions in the env gene of HIVs, with the aim of finding characteristics of the mechanism which generates the immunological diversity of the env protein of HIVs. We found that nucleotide changes between A and G are predominant compared to those between other nucleotides. Since this feature is consistent with the pattern of nucleotide substitutions of other retroviral genes but is quite different from those of most eukaryotic genes, a high rate of nucleotide substitution between A and G appears to be specific for retroviruses including HIVs. We discuss the biological relationship between this biased substitution and the mechanism generating hypervariability of epitopes on the env protein of HIVs. 相似文献

Microtubule (MT) binding accelerates the rate of ATP hydrolysis in kinesin. To understand the underlying mechanism, using charged‐to‐alanine mutational analysis, we identified two independent sites in tubulin, which are critical for kinesin motility, namely, a cluster of negatively charged residues spanning the helix 11–12 (H11–12) loop and H12 of α‐tubulin, and the negatively charged residues in H12 of β‐tubulin. Mutation in the α‐tubulin‐binding site results in a deceleration of ATP hydrolysis (kcat), whereas mutation in the β‐tubulin‐binding site lowers the affinity for MTs (K0.5MT). The residue E415 in α‐tubulin seems to be important for coupling MT binding and ATPase activation, because the mutation at this site results in a drastic reduction in the overall rate of ATP hydrolysis, largely due to a deceleration in the reaction of ADP release. Our results suggest that kinesin binding at a region containing α‐E415 could transmit a signal to the kinesin nucleotide pocket, triggering its conformational change and leading to the release of ADP. 相似文献

Effects of aging on the biological discrimination between calcium (Ca) and strontium (Sr) by the kidneys and bone were studied in male and female rats of 5 to 50 wk of age by examining Sr/Ca ratios in the plasma, urine, and bone. The Ca-Sr discrimination at the reabsorption process in the kidneys was not affected by aging in male or female rats. On the other hand, discrimination between the two elements was shown to be age-related at the absorption process in the digestive tract, and became more strict with age. The reverse situation was observed in the discrimination of Ca and Sr in the femur; younger rats discriminated the two elements more strictly than older animals. 相似文献

The quality of protein function predictions relies on appropriate training of protein classification methods. Performance of these methods can be affected when only a limited number of protein samples are available, which is often the case in divergent protein families.
