The Mec1/ATR kinase is crucial for genome maintenance in response to a range of genotoxic insults, but it remains unclear how it promotes context‐dependent signaling and DNA repair. Using phosphoproteomic analyses, we uncovered a distinctive Mec1/ATR signaling response triggered by extensive nucleolytic processing (resection) of DNA ends. Budding yeast cells lacking Rad9, a checkpoint adaptor and an inhibitor of resection, exhibit a selective increase in Mec1‐dependent phosphorylation of proteins associated with single‐strand DNA (ssDNA) transactions, including the ssDNA‐binding protein Rfa2, the translocase/ubiquitin ligase Uls1, and the Sgs1‐Top3‐Rmi1 (STR) complex that regulates homologous recombination (HR). Extensive Mec1‐dependent phosphorylation of the STR complex, mostly on the Sgs1 helicase subunit, promotes an interaction between STR and the DNA repair scaffolding protein Dpb11. Fusion of Sgs1 to phosphopeptide‐binding domains of Dpb11 strongly impairs HR‐mediated repair, supporting a model whereby Mec1 signaling regulates STR upon hyper‐resection to influence recombination outcomes. Overall, the identification of a distinct Mec1 signaling response triggered by hyper‐resection highlights the multi‐faceted action of this kinase in the coordination of checkpoint signaling and HR‐mediated DNA repair.  相似文献

Vulture populations worldwide have suffered precipitous declines in recent decades. The Cinereous Vulture Aegypius monachus, a highly philopatric scavenger distributed across southern Europe and the central Asian plateau, is threatened in many parts of its range. Turkey holds the second largest population of this species in the Western Palaearctic, but there has been no research on its genetic structure and the possible implications of this structure for the future of the species. Here we report nuclear diversity and relatedness determined by short tandem repeat genotyping of 81 individuals from the four largest colonies. Our results demonstrated no significant genetic structuring, suggesting a single panmictic metapopulation connected by frequent dispersal. Furthermore, we show that the study population has retained moderate levels of genetic diversity, despite passing through a recent demographic bottleneck. We estimated the effective population size to be 112 individuals (95% confidence interval 74–201). Our results imply that the observed lack of increase in population size since the 1990s has not been caused by lowered fitness due to genetic inbreeding but rather by increased mortality via demographic processes. In the short term, we suggest that conservation efforts should treat the Turkish subpopulations as a single management unit and aim to increase population size through effective protection, especially during the breeding season.  相似文献

Cytological events in Zea mays root meristem were followed during 26 hr of anaerobic treatment. From 8 to 14 hr, mitochondria swelled drastically, Golgi apparatus actively produced vesicles, endoplasmic reticulum proliferated, and chromatin strongly condensed. Plastids appeared normal. By 26 hr, however, Golgi activity receded, mitochondria assumed long, polymorphic shapes, chromatin partly dispersed, and plastids swelled. ER remained prominent, and the cytoplasm contained long fibrous inclusions. This preliminary study emphasizes the need to examine quantitatively all cellular organelles periodically for longer periods when following events of stress or pathology. Our observations corroborate scattered reports in the literature for single organelles under anaerobic stress and represent the first set of correlated observations on the entire spectrum of cellular events.  相似文献

A theory of follicle selection is developed in which all follicles, ovulatory and atretic, inherit the same developmental plan for responding to circulating concentrations of estradiol and gonadotropins. In the model, this plan is represented by a maturation surface that determines the rate of follicle growth as a function of follicle maturity and circulating hormone concentration. Examples of maturation surfaces are constructed for single and multiple spontaneous ovulators. When model follicles are activated from the reserve pool at random times, spontaneous and coordinated cycles of maturation develop in which the number of ovulatory follicles is controlled within a small and predictable range. For a given maturation surface, this range is nearly independent of the number of follicles in the interacting population, their activation times, and their initial maturities. The theory is therefore consistent with Lipschütz's Law of Follicular Constancy. The theory can account for certain statistical effects that occur with age on the timing of ovulation and the control of ovulation number. Maturation surfaces are also constructed that exhibit spontaneous transitions from cyclic to steady anovulatory behavior. The theory predicts an important regulatory role for atretic follicles in controlling both the timing of maturation and the number of follicles that reach ovulatory maturity.  相似文献

We review the literature describing constitutive activity of the five muscarinic acetylcholine receptors in native and recombinant systems and discuss the effect of constitutive activity on muscarinic pharmacology in the context of modern models of receptor activation. We include a summary of mutations found to cause constitutive activity and discuss the implications of these data for the structure, function, and activation mechanism of muscarinic receptors. Finally, we discuss the possible physiological significance of constitutive activity of muscarinic receptors, incorporating information provided by targeted deletion of each of the muscarinic subtypes.  相似文献

Many of the cellular mechanisms underlying host responses to pathogens have been well conserved during evolution. As a result, Drosophila can be used to deconstruct many of the key events in host-pathogen interactions by using a wealth of well-developed molecular and genetic tools. In this review, we aim to emphasize the great leverage provided by the suite of genomic and classical genetic approaches available in flies for decoding details of host-pathogen interactions; these findings can then be applied to studies in higher organisms. We first briefly summarize the general strategies by which Drosophila resists and responds to pathogens. We then focus on how recently developed genome-wide RNA interference (RNAi) screens conducted in cells and flies, combined with classical genetic methods, have provided molecular insight into host-pathogen interactions, covering examples of bacteria, fungi and viruses. Finally, we discuss novel strategies for how flies can be used as a tool to examine how specific isolated virulence factors act on an intact host.  相似文献
