Trichoderma reesei is the industrial producer of cellulases and hemicellulases for biorefinery processes. Their expression is obligatorily dependent on the function of the protein methyltransferase LAE1. The Aspergillus nidulans orthologue of LAE1 - LaeA - is part of the VELVET protein complex consisting of LaeA, VeA and VelB that regulates secondary metabolism and sexual as well as asexual reproduction. Here we have therefore investigated the function of VEL1, the T. reesei orthologue of A. nidulans VeA. Deletion of the T. reesei vel1 locus causes a complete and light-independent loss of conidiation, and impairs formation of perithecia. Deletion of vel1 also alters hyphal morphology towards hyperbranching and formation of thicker filaments, and with consequently reduced growth rates. Growth on lactose as a sole carbon source, however, is even more strongly reduced and growth on cellulose as a sole carbon source eliminated. Consistent with these findings, deletion of vel1 completely impaired the expression of cellulases, xylanases and the cellulase regulator XYR1 on lactose as a cellulase inducing carbon source, but also in resting mycelia with sophorose as inducer. Our data show that in T. reesei VEL1 controls sexual and asexual development, and this effect is independent of light. VEL1 is also essential for cellulase gene expression, which is consistent with the assumption that their regulation by LAE1 occurs by the VELVET complex. 相似文献

Aggregation patterns of plants vary according to spatial scale and developmental stage, and are dependent on vegetation dynamics and species composition. We describe the aggregation patterns of the epiphytic orchid Psychilis monensis from two sites with different vegetation structure and composition on Mona Island, Puerto Rico. We analyzed spatial variation for seedlings, juveniles, and adults using the density-independent, standardized Morisita index (IMS ). We censused a total of 879 plants. Strong preferences for some phorophyte species were evident, including dead trees, but association with bark roughness was equivocal. The highest densities occurred in the site with the lowest fruit and seed production, suggesting that the best sites for pollination and seedling establishment were not the same. Seedlings at one site were significantly aggregated, but all other stages were indistinguishable from a random distribution. Nevertheless, adults at both sites had the lowest IMS values indicating that they tend to be the least aggregated of the three life history stages. The abundance and age structure of P. monensis were clearly affected by the frequency of their preferred hosts, but site-specific factors affecting seedling survival probably play a major role in site differences. 相似文献

A new system has been developed to study hormone-directed transport in intact plants during parthenocarpic fruit set induced by gibberellins. Gibberellic acid (GA3 ) and gibberellin A1 (GA1 ) applied to unpollinated ovaries of pea ( Pisum sativum L. cv. Alaska) promoted sucrose transport from the leaf to the site of hormone application. In vivo experiments showed an early (30 min) accumulation of [14 C]-sucrose in ovaries of pea stimulated by gibberellins. This activation of sucrose transport appears to be mediated by gibberellins (GA1 , GA3 ), increasing both loading of phloem with sucrose in the leaf (source) and sucrose unloading in the ovary (sink). The ability of pea tissue segments to take up sucrose in vitro was not affected by the hormonal treatment. 相似文献

Propionyl-CoA carboxylase (PCC) is a mitochondrial, biotin-dependent enzyme involved in the catabolism of branched chain
amino acids, odd chain fatty acids, and other metabolites. PCC consists of non-identical subunits, α and β, encoded by the
PCCA and PCCB genes, respectively. Inherited deficiency of PCC due to mutations in either the PCCA or the PCCB gene results
in propionic acidemia (PA), a clinically heterogeneous disorder with a severe, often lethal, neonatal form, and a mild, later
onset form. To characterize PCCA gene mutations responsible for PCC deficiency, we analyzed RT-PCR products obtained from
cultured fibroblasts from Spanish PCC-α deficient patients. In three patients, smaller than normal PCR products were observed,
and sequence analysis revealed the deletion of a 54-bp exon in the cDNA. Sequencing of genomic DNA from these three patients
led to the identification of three novel mutations in the PCCA gene, two short deletions and one small insertion, adjacent
to short direct repeats, and all of them affecting the consensus splice sites of the skipped exon. These mutations, 1771IVS-2del9,
1824IVS+3del4, and 1824IVS+3insCT, are the cause of the aberrant splicing of the PCCA pre-mRNA and result in an in-frame deletion
of 54 nucleotides in the cDNA, probably leading to an unstable protein structure which is responsible for the lack of activity
leading to PCC deficiency in these patients.

Comparative plant successional studies on derelict sites are providing significant insights into vegetation dynamics to ensure the success of future revegetation projects in these areas and, in the short-term, by using a space-for-time substitution. In this paper we describe, in relation to site exposure, vegetation development on waste rock materials covered with biologically active soil media, and compare this development with that from a previous study at the same mine (CW Spain) on non-biologically modified waste rock materials. The succession under study is faster on the North slope, as expected, and does not differ significantly from the general pattern of primary revegetation, although it was characterised by its own sequence of plant species. The topsoiling of waste increases richness and diversity from the first year of revegetation, reduces the time required for recovery of a terminal stage, and highlights the influence of slope orientation on vegetation dynamics. A total of 237 plant taxa were recorded, showing one of four patterns of change: (1) ‘pioneer’, (2) ‘intermediate’, (3) ‘late coloniser’ and (4) ‘fluctuating’. 相似文献

Duvalius (sg. Neoduvalius) gejzadunayi
sp. n. from Pećina u Dubokom potoku cave ( Donje Biševo village near Rožaje, Montenegro), the first known representative of this subgenus from the territory of Montenegro is described, illustrated and compared with the related species of the subgenus Neoduvalius Müller, 1913. This new species is characterised by depigmented, medium sized body, totally reduced eyes, deep and complete frontal furrows, 3–4 pairs of discal setae in third elytral stria, as well as by the shape of aedeagus. Data on the distribution and the ecology of this remarkable species, as well as a check-list of the subgenus Neoduvalius are also provided. Recently described genera Serboduvalius Ćurčić, S. B. Visual cognitive responses (P300) to moving stimuli were tested in 36 subjects with the aim to find the normal range of P300 parameters. Concomitantly, the circadian intra-individual variability of the P300 was studied in a subgroup of 6 subjects. Visual stimuli consisted of either coherent (frequent stimulus) or non-coherent motion (random stimulus). The oddball paradigm was applied for recording cognitive responses. P300 to rare stimuli had an average latency of 447.3 +/- 46.6 ms and amplitude of 12.9 +/- 6.0 microV. The average reaction time was in the range from 322 to 611 ms and there was no correlation between the reaction time and P300 latency. We did not find any significant circadian changes of the P300 parameters in the 6 subjects tested four times during the same day. Cognitive (event-related) responses (P300) displayed distinctly greater inter-individual variability (S.D. of 50 ms) when compared with pattern-reversal and motion-onset VEPs (S.D. of 6.0 ms and 14 ms, respectively). For this reason, the clinical use of P300 elicited by this kind of visual stimuli seems to be rather restricted and the evaluation of its intra-individual changes is preferable. 相似文献

We assessed the role of nutrients and disturbance experienced by mothers (maternal effects) in the growth of progeny in a pot experiment using two Plantago species. Photosynthetic capacity, biomass allocation and fecundity were measured. Offspring of plants grown in nutrient poor conditions produced more leaves, spikes and longer leaves and in case of P. lanceolata , they had also higher photosynthetic capacity. The progeny of P. media mothers that had resprouted after disturbance was favored in nutrient poor conditions whereas the progeny of undisturbed plants was favored in nutrient rich conditions.
This study demonstrates that maternal effects may play a role in the success of either a seeding or a resprouting strategy in environments with different nutrient availability. Moreover, we showed that alteration of photosynthetic capacity, even during adult stages, is a mechanism through which maternal plants may impact their progeny. 相似文献
