Chemopreventive approaches for the treatment of breast cancer have been validated clinically and with in vitro studies. The combined action of tamoxifen/all‐trans retinoic acid was advantageous in MCF‐7 cells, reducing cell proliferation, Bcl‐2 and c‐Myc protein levels and increasing E‐Cadherin protein levels and Gap junctional Intercellular Communication. We further investigated their combined effect in the presence of bradykinin, a pro‐inflammatory agent, previously reported to contribute to the proliferation of breast cancer cells. Bradykinin increased MCF‐7 cell proliferation, c‐Myc levels and ERK1/2 activity. The co‐incubation of bradykinin‐MCF‐7 cells with tamoxifen/all‐trans retinoic acid reduced cell proliferation, ERK1/2 activity, as well as Bcl‐2, c‐Myc, and bradykinin receptor‐2 levels, without altering the enhanced E‐cadherin levels induced by tamoxifen/all‐trans retinoic acid. We showed that the anti‐tumoral effect of tamoxifen/all‐trans retinoic acid is beneficial in MCF‐7 breast cancer cells grown in a bradykinin‐pro‐mitogenic environment, an effect that might be, at least in part, through the MAPK pathway and B2‐bradykinin receptor inhibition. Animals for survival in complex, time-evolving environments can estimate in a “single parallel run” the fitness of different alternatives. Understanding of how the brain makes an effective compact internal representation (CIR) of such dynamic situations is a challenging problem. We propose an artificial neural network capable of creating CIRs of dynamic situations describing the behavior of a mobile agent in an environment with moving obstacles. The network exploits in a mental world model the principle of causality, which enables reduction of the time-dependent structure of real situations to compact static patterns. It is achieved through two concurrent processes. First, a wavefront representing the agent’s virtual present interacts with mobile and immobile obstacles forming static effective obstacles in the network space. The dynamics of the corresponding neurons in the virtual past is frozen. Then the diffusion-like process relaxes the remaining neurons to a stable steady state, i.e., a CIR is given by a single point in the multidimensional phase space. Such CIRs can be unfolded into real space for execution of motor actions, which allows a flexible task-dependent path planning in realistic time-evolving environments. Besides, the proposed network can also work as a part of “autonomous thinking”, i.e., some mental situations can be supplied for evaluation without direct motor execution. Finally we hypothesize the existence of a specific neuronal population responsible for detection of possible time-space coincidences of the animal and moving obstacles.  相似文献

The autotransporter family of proteins is an important class of Gram-negative secreted virulence factors. Their secretion mechanism comprises entry to the periplasm via the Sec apparatus, followed by formation of an outer membrane beta barrel, which allows the N-terminal passenger domain to pass to the extracellular space. Several groups have identified a region immediately upstream of the beta domain that is important for outer membrane translocation, the so-called linker region. Here we characterize this region in EspP, a prototype of the serine protease autotransporters of enterobacteriaceae. We hypothesized that the folding of this region would be important in the outer membrane translocation process. We tested this hypothesis using a mutagenesis approach in conjunction with a series of nested deletions and found that in the absence of a complete passenger, mutations to the C-terminal helix, but not the upstream linker, significantly decrease secretion efficiency. However, in the presence of the passenger mutations to the amino-terminal region of the linker decrease secretion efficiency. Moreover, amino acids of hydrophobic character play a crucial role in linker function, suggesting the existence of a hydrophobic core or hydrophobic interaction necessary for outer membrane translocation of autotransporter proteins.  相似文献

Forty-six Mesorhizobium strains associated with the leguminous plants Leucaena leucocephala and Sesbania herbacea in an uncultivated Mexican field were characterized using a polyphasic approach. The strains were identified as Mesorhizobium plurifarium based upon the close relationships with the reference strains for this species in PCR-based restriction fragment length polymorphism analyses, sequencing of 16S rRNA genes, multilocus enzyme electrophoresis, and DNA-DNA hybridization. Although the strains isolated from both plants formed the same group in multilocus enzyme electrophoresis and cross-nodulations were observed in the laboratory, different electrophoretic types were obtained from the two plants grown in natural soils, indicating the existence of a preferable association between the plants and the rhizobia. The M. plurifarium strains from Mexico and the reference strains from Africa and Brazil formed different phenotypic clusters in a numerical taxonomy. The Mexican strains did not grow at 37 °C and were sensitive to salty-alkaline conditions, while the reference strains from Africa and Brazil grew at 42 °C and were more resistant to salty-alkaline conditions. These results demonstrate that both the plants and environmental factors affected the evolution of rhizobia and that the Mexican strains had adapted to the neutral soils and the cool climate where they were isolated.  相似文献

Sporulation in Bacillus subtilis can be triggered by carbon catabolite limitation. Conversely, carbon source excess can repress the production of extracellular enzymes, motility, and sporulation. Recent studies have implicated a pH-sensing mechanism, involving AbrB, the TCA cycle, Spo0K, and ÏH in controlling the catabolite repression of sporulation gene expression. In an accompanying paper, we demonstrate that the AbrB-dependent pH-sensing mechanism may not be the only means by which carbon catabolites affect sporulation. In the studies reported here, we have examined the molecular basis underlying the catabolite repression phenotype of mutations in the hpr (scoC), rpoD (crsA47), and spo0A (rvtA11) loci. Loss of function mutations in hpr (scoC) restored sporulation gene expression and sporulation in the presence of excess catabolite(s), suggesting that Hpr (ScoC) has a pivotal role in mediating catabolite repression. Moreover, hpr gene expression increased substantially in the presence of excess catabolite(s), further supporting the involvement of Hpr (ScoC) in the carbon catabolite response system. Human astrovirus is an important cause of acute gastroenteritis. We have generated, for the first time, a vaccinia virus recombinant expressing the astrovirus 87-kDa structural polyprotein. The results demonstrate that this expression results in the formation of virus-like particles in the absence of other astrovirus proteins and genomic RNA. The purified trypsin-activated virus-like particles strongly resemble the complete astrovirus particles.  相似文献

A wide variety of in vitro models have been used for studying rabies infection, however, currently, no central nervous system (CNS) adult neuron cultures are available. The current study determined the susceptibility to rabies infection in an adult CNS neuron cell line (CAD-R1). Cultures of CAD-R1 cells were held for 5 days in medium containing serum (undifferentiated CAD-R1 cells) or in serum-free medium (differentiated CAD-R1 cells). They were then infected with highly neurotropic rabies virus (RV) strain (CVS), obtained from fibroblastic cells (CVS-BHK) or from adult mouse brain (CVS-MB). Undifferentiated and differentiated cells were infected with the two RV strains, but the percentage of infected cells in differentiated cultures was significantly greater (83% and 79%, respectively) than in undifferentiated cells (51% and 60%) (Student's t test<0.05). Susceptibility to infection apparently depended on cellular differentiation state, possibly due to acquisition of additional morphological and biochemical characteristics during the differentiation process that made them more susceptible to RV infection. Therefore, CAD R1 cells may represent a good model for RV infection, making them a useful tool for studying RV neurotropism, infection pathogeny, isolation of street virus or producing safer and most potent vaccines.  相似文献

Eighteen clinical isolates of Trichomonas vaginalis were obtained from women who attended health centers of the Government of Madrid. A total of 1,848 vaginal specimens recovered during the gynaecological examination were seeded in culture tubes containing liquid Diamond medium. Pathogenicity to mice was determined after intraperitoneal inoculation of mice by quantification of mortality and gross damage to abdominal organs. As could be expected, a broad variability was obtained, being some of the isolates more virulent than the reference strain. Regarding to metronidazole susceptibility, none resistant isolate was found but different degrees of susceptibility were determined.  相似文献

Acetobacter diazotrophicus isolates that originated from different sugarcane cultivars growing in diverse geographic regions of Mexico and Brazil were shown to have limited genetic diversity. Measurements of polymorphism in the electrophoretic mobilities of metabolic enzymes revealed that the mean genetic diversity per enzyme locus (among the four electrophoretic types distinguished) was 0.064. The results of the genetic analysis indicate that the genetic structure of A. diazotrophicus is clonal, with one largely predominant clone. Plasmids were present in 20 of 24 isolates, and the molecular sizes of the plasmids ranged from 2.0 to 170 kb. Two plasmids (a 20- to 24-kb plasmid detected in all 20 plasmid-containing isolates and a 170-kb plasmid observed in 14 isolates) were highly conserved among the isolates examined. Regardless of the presence of plasmids, all of the isolates shared a common pattern of nif structural gene organization on the chromosome.  相似文献
