Animal experimentation contributes significantly to the progression of science. Nonhuman primates play a particularly important role in biomedical research not only because of their anatomical, physiological, biochemical, and behavioral similarities with humans but also because of their close phylogenetic affinities. In order to investigate the use of New World primates (NWP) in biomedical research over the last four decades (1966–2005), we performed a quantitative study of the literature listed in bibliographic databases from the Health Sciences. The survey was performed for each genus of NWP that has been bred in the National Center of Primates in Brazil. The number of articles published was determined for each genus and sorted according to the country from which the studies originated and the general scientific field. The great challenges for researchers working in the field of vaccinology are optimizing DNA vaccines for use in humans or large animals and creating effective single-dose vaccines using appropriated controlled delivery systems. Plasmid DNA encoding the heat-shock protein 65 (hsp65) (DNAhsp65) has been shown to induce protective and therapeutic immune responses in a murine model of tuberculosis (TB). Despite the success of naked DNAhsp65-based vaccine to protect mice against TB, it requires multiple doses of high amounts of DNA for effective immunization. In order to optimize this DNA vaccine and simplify the vaccination schedule, we coencapsulated DNAhsp65 and the adjuvant trehalose dimycolate (TDM) into biodegradable poly (DL-lactide-co-glycolide) (PLGA) microspheres for a single dose administration. Moreover, a single-shot prime-boost vaccine formulation based on a mixture of two different PLGA microspheres, presenting faster and slower release of, respectively, DNAhsp65 and the recombinant hsp65 protein was also developed. These formulations were tested in mice as well as in guinea pigs by comparison with the efficacy and toxicity induced by the naked DNA preparation or BCG. The single-shot prime-boost formulation clearly presented good efficacy and diminished lung pathology in both mice and guinea pigs.  相似文献

Background

The electroencephalography (EEG) is an attractive and a simple technique to measure the brain activity. It is attractive due its excellent temporal resolution and simple due to its non-invasiveness and sensor design. However, the spatial resolution of EEG is reduced due to the low conducting skull. In this paper, we compute the potential distribution over the closed surface covering the brain (cortex) from the EEG scalp potential. We compare two methods – L-curve and generalised cross validation (GCV) used to obtain the regularisation parameter and also investigate the feasibility in applying such techniques to N170 component of the visually evoked potential (VEP) data.

Methods

Using the image data set of the visible human man (VHM), a finite difference method (FDM) model of the head was constructed. The EEG dataset (256-channel) used was the N170 component of the VEP. A forward transfer matrix relating the cortical potential to the scalp potential was obtained. Using Tikhonov regularisation, the potential distribution over the cortex was obtained.

Results

The cortical potential distribution for three subjects was solved using both L-curve and GCV method. A total of 18 cortical potential distributions were obtained (3 subjects with three stimuli each – fearful face, neutral face, control objects).

Conclusions

The GCV method is a more robust method compared to L-curve to find the optimal regularisation parameter. Cortical potential imaging is a reliable method to obtain the potential distribution over cortex for VEP data.

Progesterone (P(4)), 17beta- estradiol (E(2)) and androstenedione (A(4)) plasma concentrations were correlated with palpated corpora lutea (CL), recovered embryos and viable embryos in 13 Nelore cows induced to superovulate with FSH, starting on Day 10 of the estrous cycle. Administration of FSH increased the number of ovulations and recovered embryos. Plasma P(4), E(2) and A(4) levels on Day 0 and of P(4) on Days 10 and 11 of the cycle were not correlated with the superovulatory response. Determination of CL by palpation per rectum was used to estimate the number of recovered embryos. Plasma P(4) levels higher than 1 ng/ml on the induced estrus day (Day 14) had an adverse effect on the embryo viability rate. Plasma E(2) concentrations on Day 14 were positively correlated with the number of viable embryos collected, a correlation that has not been previously reported. The present data indicate that plasma P(4) and E(2) concentrations in FSH-PGF2alpha-treated Nelore cows are useful for the identification of 2 different populations of Nelore donors and are correlated with superovulatory response and, particularly, with the number of viable embryos.  相似文献

A powerful IFN-gamma response is triggered upon infection with the protozoan parasite, Toxoplasma gondii. Several cell populations, including dendritic cells (DCs), macrophages, and neutrophils, produce IL-12, a key cytokine for IFN-gamma induction. However, it is still unclear which of the above cell populations is its main source. Diphtheria toxin (DT) injection causes transient DC depletion in a transgenic mouse expressing Simian DT receptors under the control of the CD11c promoter, allowing us to investigate the role of DCs in IL-12 production. T. gondii-inoculated DT-treated and control groups were monitored for IL-12 levels and survival. We show in this study that DC depletion abolished IL-12 production and led to mortality. Furthermore, replenishment with wild-type, but not MyD88- or IL-12p35-deficient, DCs rescued IL-12 production, IFN-gamma-induction, and resistance to infection in DC-depleted mice. Taken together, the results presented in this study indicate that DCs constitute the major IL-12-producing cell population in vivo during T. gondii infection.  相似文献

Urbanization is often associated with homogenization, including the homogenization of biodiversity and overpopulation by generalist species that can change community and disease dynamics. In Madrid, nestlings of Eurasian Scops Owl Otus scops frequently suffer necrotic oropharyngeal disease as a result of infection by Gongylonema sp., a parasitic nematode transmitted by adult owls to their offspring through the diet, more specifically through consumption of a pest species: the oriental cockroach Blatta orientalis. We studied how the presence of cockroaches and owl infection rates are affected by urban features such as green areas. We found that a higher number of birds were affected by the parasite in the initial period of the breeding season and in areas with greater surface area of green spaces. We suggest that urban management should promote the diversity of insects in green areas so that the owls diversify their dietary offer to their offspring, to reduce the prevalence of the disease and improve their breeding success in the city of Madrid, as this problem has not yet been described in other cities.  相似文献

Species-specific climate sensitivity of tree growth in Central-West Germany   总被引:1,自引:0,他引:1  
Growth responses to twentieth century climate variability of the three main European tree species Fagus sylvatica, Quercus petraea, and Pinus sylvestris within two temperate low mountain forest sites were analyzed, with particular emphasis on their dependence upon ecological factors and temporal stability in the obtained relationships. While site conditions in Central (~51°N, 9°E, KEL) and West (50.5°N, 6.5°E, EIF) Germany are similar, annual precipitation totals of ≅700 mm and ≅1,000 mm describe a maritime-continental gradient. Ring-width samples from 228 trees were collected and PCA used to identify common growth patterns. Chronologies were developed and redundancy analysis and simple correlation coefficients calculated to detect twentieth century temperature, precipitation, and drought fingerprints in the tree-ring data. Summer drought is the dominant driver of forest productivity, but regional and species-specific differences indicate more complex influences upon tree growth. F. sylvatica reveals the highest climate sensitivity, whereas Q. petraea is most drought tolerant. Drier growth conditions in KEL result in climate sensitivity of all species, and Q. petraea shifted from non-significant to significant drought sensitivity during recent decades at EIF. Drought sensitivity dynamics of all species vary over time. An increase of drought sensitivity in tree growth was found in the wetter forest area EIF, whereas a decrease occurred in the middle of the last century for all species in the drier KEL region. Species-specific and regional differences in long-term climate sensitivities, as evidenced by temporal variability in drought sensitivity, are potential indicators for a changing climate that effects Central-West German forest growth, but meanwhile hampers a general assessment of these effects.  相似文献

Physical exercise stimulates the release of endogenous opioid peptides supposed to be responsible for changes in mood, anxiety, and performance. Exercise alters sensitivity to these effects that modify the efficacy at the opioid receptor. Although there is evidence that relates exercise to neuropeptide expression in the brain, the effects of exercise on opioid receptor binding and signal transduction mechanisms downstream of these receptors have not been explored. Here, we characterized the binding and G protein activation of mu opioid receptor, kappa opioid receptor or delta opioid receptor in several brain regions following acute (7 days) and chronic (30 days) exercise. As regards short‐ (acute) or long‐term effects (chronic) of exercise, overall, higher opioid receptor binding was observed in acute‐exercise animals and the opposite was found in the chronic‐exercise animals. The binding of [35S]GTPγS under basal conditions (absence of agonists) was elevated in sensorimotor cortex and hippocampus, an effect more evident after chronic exercise. Divergence of findings was observed for mu opioid receptor, kappa opioid receptor, and delta opioid receptor receptor activation in our study. Our results support existing evidence of opioid receptor binding and G protein activation occurring differentially in brain regions in response to diverse exercise stimuli.
