In order to evaluate the taeniosis-cysticercosis complex in a population of a peasants' settlement, located at Teodoro Sampaio, state of S?o Paulo, Brazil (longitude 52 degrees 36'12 ", latitude 22 degrees 17'12 ") a series of laboratory markers were determined. After signing an informed consent, participants answered a standardized questionnaire. To determine anti-Taenia solium cysticercus antibodies, the samples were tested by enzyme linked immunoabsorbent assay using 18-and 14-kDa antigen proteins from vesicular fluid of Taenia crassiceps (VF-Tcra). The reactive and inconclusive ELISA samples were tested by immunoblotting. Total IgE levels were determined by chemmiluminescence's assay and hemogram by flow cytometer flux counter. A total of 84 individuals, 5.9% presented anti-T. solium cysticercus antibodies in ELISA and 3.6% were strongly reactive in the 18/14 kDa immunoblotting confirmatory test. All of the individuals with positive antibodies showed elevated Total IgE levels. We conclude that the frequency of anti-T. solium cysticercus antibodies in this population is higher than other regions considered endemic in S?o Paulo. Thus, it is important to carry out surveys in Peasants' settlement areas with the objective of establishing public health measures for prevention and control of infectious diseases such as taeniosis-cysticercosis.  相似文献

Chagas' disease is chiefly transmitted by feces of haematophagous bugs (Triatominae) that ingested Trypanosoma cruzi from blood of infected people or animals. Pyrethroids have been the main insecticides used against these insects. However, some populations of insects have shown significant levels of resistance to several pyrethroids, indicating the need of new insecticides for the control of triatomines. Insecticidal activity of 24 Cerrado plant extracts belonging to five species of four families were assayed on fourth instar nymphs of Rhodnius milesi Carcavallo, Rocha, Galv?o & Jurberg (Hemiptera: Reduviidae), under laboratory conditions. For the extract application on triatomines, 50 microg of the extract were topically applied in duplicate on dorsal tergites of ten insects. Insects topically treated with acetone, ethanol, as well as insects with no treatment were used as controls. Triatomines were observed over a 28-day period. Hexanic and ethanolic extracts of Simarouba versicolor, Guarea kunthiana, Guarea guidonia and Talauma ovata caused mortality between 20% and 95% of R. milesi in comparison with the controls, which showed no insect mortality. These preliminary data suggest that the ethanolic extract of the root bark of S. versicolor and the hexanic extract of the root of G. guidonia, responsible for a 95% and 75% insect mortality, respectively, should be chemically investigated and monitored through biological assays in order to determine their insecticidal components, that could be used as a molecular model or as biorational compounds for use in insect control programmes.  相似文献

Recent advances in fluorescent microscopy have revealed the non-random organization of chromosomes in bacterial cells. In Escherichia coli, segments included in two large regions show similar localization patterns allowing the definition of two macrodomains centered, respectively, on the centromere-like site migS (the Ori domain) and the replication terminus (the Ter domain). A genetic system measuring long distance DNA interactions confirmed the macrodomain organization of the chromosome and revealed the existence of two additional macrodomains flanking the Ter domain. Altogether, the E. coli chromosome appears to have an open ring-like conformation. Recent developments regarding various stages of chromosome biology such as replication, sister chromatid cohesion, segregation, and chromosome choreography, have considerably improved our understanding of the coordination of chromosome dynamics with the cell cycle. The possible involvement of macrodomains in these various processes is discussed.  相似文献

Food availability may affect metal toxicity for aquatic organisms. In the present study, the influence of high, medium and low densities of the algae Pseudokirchneriella subcapitata (10(6), 10(5) and 10(4) cells.mL(-1), respectively) on the chronic toxicity of chromium to the cladoceran Ceriodaphnia dubia was investigated. C. dubia was exposed to a range of chromium concentration from 2.71 to 34.04 microg.L(-1) and fed with algae at various densities. In another experiment, the green alga was exposed to chromium concentrations (94 to 774 microg.L(-1)) and supplied as food in different densities to zooplankton. The survival and reproduction of the cladoceran were measured in these toxicity tests. The IC50 for Cr to P. subcapitata and metal accumulated by algal cells were determined. The results of a bifactorial analysis (metal versus algal densities) showed that metal toxicity to zooplankton was dependent on algal densities. Significant toxic effects on the reproduction and survival of C. dubia were observed at 8.73, 18.22 and 34.04 microg.L(-1) Cr when the test organisms were fed with 10(6) cells.mL(-1) of P. subcapitata. Although the chlorophyta retain low chromium content, a decrease in the reproduction and survival of C. dubia occurred when they were fed with high algal density contaminated with 774 microg.L(-1) Cr. It was concluded that high algal density have an appreciable influence on chromium toxicity to daphnids.  相似文献

This paper deals with the hydrological variability effects on the primary commercial fish species inhabiting the main channel and the floodplain of the large Paraná River in its middle reaches in Argentina. Analysing more than eight decades (1935–2016) of information on the most frequent and abundant commercial species in conjunction with hydrological levels and temperature, our results show that spring–summer floods of a certain magnitude (c. 6 m) and durations (> 80 days) are crucial for sustaining commercial fisheries. Moreover, the frequency of these floods was modulated by the decadal climatic fluctuations that have occurred over the past 100 years in the Paraná Basin. An insight into the probable incidence of some anthropogenic influences is also provided.  相似文献

Glycogen is the main source of glucose for many biological events. However, this molecule may have other functions, including those that have deleterious effects on cells. The rate‐limiting enzyme in glycogen synthesis is glycogen synthase (GS). It is encoded by two genes, GYS1, expressed in muscle (muscle glycogen synthase, MGS) and other tissues, and GYS2, primarily expressed in liver (liver glycogen synthase, LGS). Expression of GS and its activity have been widely studied in many tissues. To date, it is not clear which GS isoform is responsible for glycogen synthesis and the role of glycogen in testis. Using RT‐PCR, Western blot and immunofluorescence, we have detected expression of MGS but not LGS in mice testis during development. We have also evaluated GS activity and glycogen storage at different days after birth and we show that both GS activity and levels of glycogen are higher during the first days of development. Using RT‐PCR, we have also shown that malin and laforin are expressed in testis, key enzymes for regulation of GS activity. These proteins form an active complex that regulates MGS by poly‐ubiquitination in both Sertoli cell and male germ cell lines. In addition, PTG overexpression in male germ cell line triggered apoptosis by caspase3 activation, proposing a proapoptotic role of glycogen in testis. These findings suggest that GS activity and glycogen synthesis in testis could be regulated and a disruption of this process may be responsible for the apoptosis and degeneration of seminiferous tubules and possible cause of infertility. Manganese (Mn) is an essential trace element and trivalent Mn complexes have been used as oxidation catalysts and enzyme mimetics. We studied the cytotoxicity of Mn(III) derivatives of citrate, pyrophosphate and salicylene diamine (respectively, MnCit, MnPPi and EUK8) toward HeLa cells stressed by ultraviolet irradiation and the effect of the co-administration of ascorbate and para-amino salicylate (PAS) on cell viability. Metal complexes enhanced the lethality of irradiated cells, and this effect was even more pronounced when ascorbate was co-administered with Mn(III) species. The active role of Mn(III) compounds in the antitumor activity was demonstrated by the treatment of the cells with the chelator PAS, which restored the viability of both non-irradiated and UV-irradiated cells. The association of the Mn(III) metallodrugs with radiation and an antioxidant proved to be a very effective approach to chemotherapy.  相似文献
