Molecular adaptation of a leaf-eating bird: stomach lysozyme of the hoatzin   总被引:6,自引:1,他引:5  
This report describes a lysozyme expressed at high levels in the stomach of the hoatzin, the only known foregut-fermenting bird. Evolutionary comparison places it among the calcium-binding lysozymes rather than among the conventional types. Conventional lysozymes were recruited as digestive enzymes twice in the evolution of mammalian foregut fermenters, and these independently recruited lysozymes share convergent structural changes attributed to selective pressures in the stomach. Biochemical convergence and parallel amino acid replacements are observed in the hoatzin stomach lysozyme even though it has a different genetic origin from the mammalian examples and has undergone more than 300 million years of independent evolution.   相似文献

Astringency of aqueous solutions of phenolic compounds (grape seed tannins, tannic acid, catechin and gallic acid) increased upon addition of citric acid, whereas the astringency of alum was reduced. Astringency of alum was decreased equivalently by addition of equi-sour levels of lactic acid, citric acid or hydrochloric acid. The difference between alum and the phenolic compounds is speculated to result from chemical modifications affecting binding of the astringents with oral proteins rather than cognitive differences. Chelation of the aluminum ion in alum by acids reduces its availability for interacting with salivary proteins or epithelial proteins. In contrast, the increased astringency produced upon acidification of phenolic compounds is speculated to result from the pH driven increase in the affinity of the phenols for binding with proteins. These results suggest that alum cannot be used interchangeably with phenolic astringents in psychophysical studies.   相似文献

The heterobifunctional, photoactivatable, thiol-cleavable cross-linker sulfosuccinimidyl 2-(p-azido-salicylamido)ethyl-1,3'-dithiopropionate (SASD) was radioiodinated and used to determine whether endothelial albumin binding proteins (ABP) recently identified (Ghinea, N., Fixman, A., Alexandru, D., Popov, D., Hasu, M., Ghitescu, L., Eskenasy, M., Simionescu, M., and Simionescu, N. (1988) J. Cell Biol. 107, 231-239) are plasma membrane-associated components exposed on the cell surface. Microvascular endothelial cells (MEC) freshly isolated from rat epididymal fat were incubated with 125I-2-(p-azidosalicylamido)ethyl-1,3'-dithiopropionate (ASD)-albumin conjugate which upon photolysis by UV light was cross-linked to the receptor proteins. By cleaving the disulfide linkages of the cross-linker with 5% beta-mercaptoethanol and the ligand-receptor interactions with 0.1% sodium dodecyl sulfate, the radioiodinated ASD moiety remained attached to the receptor peptides which were further detected by 5% sodium dodecyl sulfate-polyacrylamide gel electrophoresis followed by autoradiography. In parallel, samples were examined by ligand blotting with albumin-gold complex. The results showed that in these experimental conditions ABP are represented by two major peptides of 31 and 18 kDa and two minor bands of 73 and 56 kDa. Densitometric scanning showed that the two major bands constitute more than 70% of the total ABP. The four peptides were not apparent if the samples were not UV-irradiated. The binding of the radioiodinated ligand to ABPs was reduced by approximately 82% in the presence of excess competitive unlabeled albumin. When MEC were incubated with unlabeled SASD and exposed to UV light, the autoradiographic banding pattern obtained was similar to that of either radioiodinated receptor proteins or MEC not treated with SASD. This indicated that the four albumin binding peptides are distinct proteins of the endothelial cell plasmalemma.  相似文献

The circulating cells arriving at the formation site of the experimentally induced lung granuloma were quantitatively and qualitatively determined during their development by estimation of alkaline phosphatase active free cells. These represent 32 per cent the 2nd day and 27 per cent the 4th, of all the lung granuloma cells, and rapidly decrease to 8 per cent the 8th day, to 4 per cent the 11th and to only 1.5 per cent the 15th day. The participation of alkaline phosphatase circulating cells appears to be important only the first days, the process becoming by proliferation and differentiation a histiocytic, epithelioidic, and lympho-plasmocytic structure under the influence of antigenic and non-antigenic components of the used immunologic adjuvant.  相似文献

A systemic approach to the pulmonary lobule in health and disease is attempted. There are emphasized the lobule definition, structure and functions (respiratory, metabolic, defensive), the program of lobular structures, their relation ways, their capacity of growth and regeneration, all contributing to the relative independence and autonomy of pulmonary lobules and to their behaviour as wholes. Taking into account the characters of pulmonary lobules in health, their pathological involvement is analysed on human and experimental materials, pointing out the importance of the aerogenic component of pathogenesis in determining the first phases of lung diseases, and of the true pulmonary characters of lesions, in comparison with those induced by the parenteral penetration of agents.  相似文献
