A thorough literature review was conducted to investigate the behaviour of food waste in anaerobic digestion experiments. The main goal of this literature review was to study the effect of several operating parameters on methane yields and to develop a simplified regression equation to predict methane generation. Using a data prospection methodology, all the papers published within 2013–2015 that contained selected keywords were included in this study (a total of 613 papers). After screening, 167 papers were finally retrieved using the search engines and our methodology. From these papers, data from 231 experiments were recorded and evaluated. The parameters recorded in each paper were: operation mode (batch or continuous), temperature (mesophilic or thermophilic), moisture content (wet or dry), presence or absence of pretreatment, reactor scale (laboratory, bench, pilot, demonstration/full scale), presence or absence of co-substrates (co- or mono-digestion), organic loading rate, hydraulic retention time (HRT) and methane yield. The novelty of the work is that it employed various statistical tools to examine the effect of the above-mentioned factors on food waste methane generation. Most of the experiments were performed at mesophilic temperatures, at a wet system without substrate pretreatment. An equal number of papers described mono-digestion and co-digestion studies, and an equal number of papers described batch and continuous reactor experiments. The mean HRT for the continuous processes was 36.7 days. Statistical analysis indicated that the parameters that significantly affected methane yields were the “operation mode” and “pretreatment”. A best reduced regression model was fitted to the methane yield data to describe the above effects. As a general conclusion, with this methodology, that involved the analysis of a large number of studies (with different conditions and set-ups, heterogeneous waste, etc.), correlations between some typical operating parameters of anaerobic digestion and methane yields were not obvious.  相似文献

The ability to initiate reactions with a flash of light and to monitor reactions over a wide temperature range allows detailed analysis of reaction mechanisms in photosynthetic reaction centers (RCs) of purple bacteria. In this protein, the electron transfer from the reduced primary quinone (Q(A)(-)) to the secondary quinone (Q(B)) is rate-limited by conformational changes rather than electron tunneling. Q(B) movement from a distal to a proximal site has been proposed to be the rate-limiting change. The importance of quinone motion was examined by shortening the Q(B) tail from 50 to 5 carbons. No change in rate was found from 100 to 300 K. The temperature dependence of the rate was also measured in three L209 proline mutants. Under conditions where Q(B) is in the distal site in wild-type RCs, it is trapped in the proximal site in the Tyr L209 mutant [Kuglstatter, A., et al. (2001) Biochemistry 40, 4253-4260]. The electron transfer slows at low temperature for all three mutants as it does in wild-type protein, indicating that conformational changes still limit the reaction rate. Thus, Q(B) movement is unlikely to be the sole, rate-limiting conformational gating step. The temperature dependence of the reaction in the L209 mutants differs somewhat from wild-type RCs. Entropy-enthalpy compensation reduces the difference in rates and free energy changes at room temperature.  相似文献

The neurotransmitter serotonin is an important regulator of energy balance. In the brain, serotonergic fibres from midbrain raphe nuclei project to key feeding centres, where serotonin acts on specific receptors to modulate the activity of various downstream neuropeptide systems and autonomic pathways and thus affects ingestive behaviour and energy expenditure. Serotonin, released by intestinal enterochromaffin cells, also appears to regulate energy homeostasis through peripheral mechanisms. Serotonergic effects on energy balance lead to secondary effects on glucose homeostasis, based on a well-established link between obesity and insulin resistance. However, serotonergic pathways may also directly affect glucose homeostasis through regulation of autonomic efferents and/or action on peripheral tissues. Several serotonergic compounds have been evaluated for clinical use in the treatment of obesity and type 2 diabetes; results of these trials are discussed here. Finally, future directions in the elucidation of serotonergic metabolic regulation are discussed.  相似文献

Introduction

Granulocyte-macrophage colony-stimulating factor (GM-CSF) has been shown to be important in the development of inflammatory models of rheumatoid arthritis and there is encouraging data that its blockade may have clinical relevance in patients with rheumatoid arthritis. The aims of the current study were to determine whether GM-CSF may also be important for disease and pain development in a model of osteoarthritis.

Methods

The role of GM-CSF was investigated using the collagenase-induced instability model of osteoarthritis. We studied both GM-CSF-/- mice and wild-type (C57BL/6) mice treated prophylactically or therapeutically with a monoclonal antibody to GM-CSF. Disease development (both early and late) was evaluated by histology and knee pain development was measured by assessment of weight distribution.

Results

In the absence of GM-CSF, there was less synovitis and matrix metalloproteinase-mediated neoepitope expression at week 2 post disease induction, and less cartilage damage at week 6. GM-CSF was absolutely required for pain development. Therapeutic neutralization of GM-CSF not only abolished the pain within 3 days but also led to significantly reduced cartilage damage.

Conclusions

GM-CSF is key to the development of experimental osteoarthritis and its associated pain. Importantly, GM-CSF neutralization by a therapeutic monoclonal antibody-based protocol rapidly and completely abolished existing arthritic pain and suppressed the degree of arthritis development. Our results suggest that it would be worth exploring the importance of GM-CSF for pain and disease in other osteoarthritis models and perhaps clinically for this form of arthritis.  相似文献

A cDNA clone encoding a putative cell wall protein (Qid3) was isolated from a library prepared from chitin-induced mRNA in cultures of the mycoparasitic fungus Trichoderma harzianum. The predicted 14 kDa protein shows a potential signal peptide, several hydrophobic domains and certain motifs that are structurally similar to proline-rich and glycine-rich plant cell wall proteins. Expression of the qid3 gene is derepressed in the absence of glucose. When introduced in yeast, qid3 expression causes cell division arrest into cytokinesis and cell separation, probably due to its cell wall localization.  相似文献

Antibody LiCO 16.88 is a human IgM recognizing a 30- to 45-kDa intracytoplasmic antigen present in human adenocarcinoma cells. An 8-mg sample of antibody labeled with 5 mCi131I was co-administered i. v. with 120 mg (three patients), 240 mg (three patients) or 480 mg (four patients) unlabeled antibody as a 4-h infusion. The plasma half-life was 24±1.2 h and the immediate apparent volume of distribution was 5.2±0.2 l at the 28-mg dose level. The plasma half-lives and the cumulative urinary excretion of radiolabel did not seem to vary significantly with increasing doses of unlabeled antibody. However, both the volume of distribution and the clearance rate from plasma increased significantly with increasing antibody dose. Uptake of antibody into tumor tissues obtained during laparotomy 8–9 days after administration varied between 0.00002% ID/g and 0.00127% ID/g. In five of seven patients, the tumor content of antibody was higher than that in adjacent normal tissue. Tumor-to-normal tissue ratios ranged from 0.8 to 10 ( =3.8±1.0). In general, the higher radioactivity(cpm)/g tumor was confirmed by both immunoperoxidase and autoradiography. Seedling performance is often a limiting factor in ecological restoration. Changes in the soil microbial community generated by invasive plants contribute to seedling failure. A method to remediate invasive species‐induced changes to the soil microbial community that results in increased native species seedling performance and decreased invasive species seedling performance could have a large impact on the success of many restoration efforts. In a greenhouse experiment, we first examined the changes in the soil microbial community created by invasive compared to native grasses. Then, we investigated four microbial treatments (bacterial inoculant, fungal inoculant, fungicide, and bactericide/fungicide) to remediate microbial plant–soil feedbacks (PSFs) created by invasive species Bromus inermis and Poa pratensis and increase the performance of natives Andropogon gerardii, Elymus canadensis, Pascopyrum smithii, and Schizachyrium scoparium. We found that the PSF mitigation treatments had some context‐dependent utility for restoration. For example, all of the treatments decreased the performance of B. inermis and fungal inoculant decreased the performance of P. pratensis. However, no single treatment increased the performance of all natives. Fungicide increased the performance of A. gerardii and E. canadensis in soil previously occupied by B. inermis and the performance of S. scoparium in soil previously occupied by P. pratensis. If validated in the field, PSF mitigation treatments may have utility for restoration practitioners.  相似文献
