社会性动物的个性特征是对环境和社群的适应性结果,对个性的研究是探究群居动物复杂社会关系及其差异的重要内容。相对于个性特征评级法和行为编码法,自然观察法是分析判断野生动物个性特征较为理想的方法之一,最大限度地突出了野生动物的自然属性及个性特征类型。本研究以栖息于安徽黄山的野生短尾猴投食群体鱼鳞坑A1群为研究对象,采用目标动物取样法和全事件记录法,于2015年7月至2016年6月期间采集成年个体自然发生的行为数据,并采用主成分分析方法判断成年个体的个性特征及类型,并分析性别差异。结果表明:成年雄性个体具有5种个性特征,即领导型、内向型、社交型、孤僻型以及受欺凌型,成年雌性个体具有6种个性特征,即领导型、孤僻型、内向型、不安型、神经质型以及友好型。研究结果与已有结果一致,说明自然观察法可能是分析野生动物个性类型的较好 选择,同时,不同性别之间个性类型的差异性进一步显示了野生动物自然属性。本研究为深入理解动物个体水平上的差异性及适应性提供了科学依据。  相似文献

蝙蝠是很多病原微生物的自然宿主, 全球多项研究表明蝙蝠是巴尔通体(Bartonella species)的主要宿主。为了解滇西南地区蝙蝠中巴尔通体的流行特征, 我们于2015-2017年间在云南省4个地区应用网捕法捕获蝙蝠3种305只。经种类鉴定后采集肝脾组织, 提取核酸, 通过TaqMan实时荧光定量PCR方法检测巴尔通体的tmRNA基因ssrA, 并进行测序鉴定和系统发育分析。结果发现172只蝙蝠检出该基因, 总感染率为56.4%; 其中临沧、西双版纳、保山和瑞丽4个采样点的蝙蝠感染率分别为50.0% (22/44)、61.7% (29/47)、62.1% (18/29)和55.7% (103/185)。中菊头蝠(Rhinolophus affinis)、小菊头蝠(R. blythi)和棕果蝠(Rousettus leschenaultii)的感染率分别为50.0% (22/44)、62.1% (18/29)和56.9% (132/232), 差异没有统计学意义(χ2 = 1.135, P = 0.567), 表明巴尔通体在云南当地的蝙蝠种群中高度流行。定量PCR扩增产物2次扩增后测序获得37个巴尔通体ssrA序列, 属于10个系统发育分支, 其中1个为伊丽莎白巴尔通体(B. elizabethae)、特利波契巴尔通体(B. tribocorum)和克拉斯诺夫巴尔通体(B. krasnovii)的近缘种。其余序列与已知巴尔通体距离较远, 与亚洲、欧洲和美洲等其他地域来源于蝙蝠的巴尔通体近缘。遗传多样性分析显示, ssrA基因的核苷酸多样性指数(π)为0.11381 ± 0.00928, 基因型多样性指数(Hd)为0.985 ± 0.010, 形成29个基因型(单倍型), 说明云南蝙蝠巴尔通体具有丰富的遗传多样性。通过对本研究标本与全球相关序列的系统发育网络重建, 分析全球蝙蝠巴尔通体的地理和宿主分布特征, 可以看出巴尔通体与蝙蝠之间存在显著的宿主特异性关联。因此可初步确定蝙蝠-巴尔通体具有协同进化特征, 同时受到地理隔离的影响。  相似文献

Aphids are insects that cause direct damage to crops by the removal of phloem sap, but more importantly they spread devastating viruses. Aphids use their sophisticated mouthpart (i.e. stylet) to feed from the phloem sieve elements of the host plant. To identify genes that affect host plant resistance to aphids, we previously screened an Arabidopsis thaliana activation tag mutant collection. In such mutants, tagged genes are overexpressed by a strong 35S enhancer adjacent to the natural promoter, resulting in a dominant gain-of-function phenotype. We previously identified several of these mutants on which the aphid Myzus persicae showed a reduced population development compared with wild type. In the present study we show that the gene responsible for the phenotype of one of the mutants is At5g65040 and named this gene Increased Resistance to Myzus persicae 1 (IRM1). Overexpression of the cloned IRM1 gene conferred a phenotype identical to that of the original mutant. Conversely, an IRM1 knockout mutant promoted aphid population development compared to the wild type. We performed Electrical Penetration Graph analysis to investigate how probing and feeding behaviour of aphids was affected on plants that either overexpressed IRM1 or contained a knockout mutation in this gene. The EPG results indicated that the aphids encounter resistance factors while reaching for the phloem on the overexpressing line. This resistance mechanism also affected other aphid species and is suggested to be of mechanical nature. Interestingly, genetic variation for IRM1 expression in response to aphid attack was observed. Upon aphid attack the expression of IRM1 was initially (after 6 hours) induced in ecotype Wassilewskija followed by suppression. In Columbia-0, IRM1 expression was already suppressed six hours after the start of the infestation. The resistance conferred by the overexpression of IRM1 in A. thaliana trades off with plant growth.  相似文献

Circadian rhythms in cardiac function are apparent in e.g., blood pressure, heart rate, and acute adverse cardiac events. A circadian clock in heart tissue has been identified, but entrainment pathways of this clock are still unclear. We cultured tissues of mice carrying bioluminescence reporters of the core clock genes, period 1 or 2 (per1luc or PER2LUC) and compared in vitro responses of atrium to treatment with medium and a synthetic glucocorticoid (dexamethasone [DEX]) to that of the suprachiasmatic nucleus (SCN) and liver. We observed that PER2LUC, but not per1luc is rhythmic in atrial tissue, while both per1luc and PER2LUC exhibit rhythmicity in other cultured tissues. In contrast to the SCN and liver, both per1luc and PER2LUC bioluminescence amplitudes were increased in response to DEX treatment, and the PER2LUC amplitude response was dependent on the time of treatment. Large phase-shift responses to both medium and DEX treatments were observed in the atrium, and phase responses to medium treatment were not attributed to serum content but the treatment procedure itself. The phase-response curves of atrium to both DEX and medium treatments were found to be different to the liver. Moreover, the time of day of the culturing procedure itself influenced the phase of the circadian clock in each of the cultured tissues, but the magnitude of this response was uniquely large in atrial tissue. The current data describe novel entrainment signals for the atrial circadian clock and specifically highlight entrainment by mechanical treatment, an intriguing observation considering the mechanical nature of cardiac tissue.  相似文献

We previously showed that mice deficient in the Inducible Costimulator ligand (ICOSL-KO) develop more severe disease and lung pathology with delayed bacterial clearance upon respiratory infection of Chlamydia muridarum. Importantly, the exacerbation of disease in ICOSL-KO mice was seen despite heightened IFN-γ/Th1 responses, the major defense mechanisms against Chlamydia. To gain insight into the mechanism of ICOS function in this model, we presently analyzed anti-Chlamydia immune responses in mice lacking the entire ICOS (ICOS-KO) versus knock-in mice expressing a mutant ICOS (ICOS-Y181F) that has selectively lost the ability to activate phosphoinositide 3-kinase (PI3K). Like ICOSL-KO mice, ICOS-KO mice showed worse disease with elevated IFN-γ/Th1 responses compared to wild-type (WT) mice. ICOS-Y181F mice developed much milder disease compared to ICOS-KO mice, yet they were still not fully protected to the WT level. This partial protection in ICOS-Y181F mice could not be explained by the magnitude of IFN-γ/Th1 responses since these mice developed a similar level of IFN-γ response compared to WT mice. It was rather IL-17/Th17 responses that reflected disease severity: IL-17/Th17 response was partially impaired in ICOS-Y181F mice compared to WT, but was substantially stronger than that of ICOS-KO mice. Consistently, we found that both polarization and expansion of Th17 cells were partially impaired in ICOS-Y181F CD4 T cells, and was further reduced in ICOS-KO CD4 T cells in vitro. Our results indicate that once the IFN-γ/Th1 response is above a threshold level, the IL-17/Th17 response becomes a limiting factor in controlling Chlamydia lung infection, and that ICOS plays an important role in promoting Th17 responses in part through the activation of PI3K.  相似文献

HSSP是用大豆密码子改造的10 kD玉米醇溶蛋白基因。在前期研究中,从获得的转基因大豆中筛选到1份单拷贝转基因材料GSDH5。该研究采用染色体步移法获取转基因大豆GSDH5的T-DNA插入位点的左边界旁侧序列,对获得的左边界旁侧序列进行分析,设计特异性引物,建立转基因大豆GSDH5特异性检测方法;采用Real-time PCR检测外源基因在转基因大豆不同组织部位(根、茎、叶、花和种子)中的表达量,采用RT-PCR和Western blot检测外源基因在转录和翻译水平上的遗传稳定性,并对转基因大豆GSDH5中的粗蛋白、含硫氨基酸含量及主要农艺性状进行测定分析,为培育高含硫氨基酸转基因大豆新品种奠定基础。结果表明:(1)分子鉴定显示,外源基因HSSP和筛选标记基因Bar成功整合到受体大豆‘东农50’基因组中,且以单拷贝的形式整合到大豆基因组中。(2)HSSP基因成功插入到大豆基因组1号染色体非编码区52 873 883 bp处。(3)HSSP基因在转基因大豆GSDH5的种子中特异性表达,且在T_2~T_4代转基因大豆中能够稳定遗传并表达。(4)‘东农50’粗蛋白含量在41.53%~43.32%之间,GSDH5粗蛋白含量在40.18%~43.03%之间,两者相比无显著差异;GSDH5种子中硫氨基酸占种子干样的比例为1.35%,占种子蛋白的比例为3.14%,与转基因受体品种‘东农50’相比,占比显著升高,分别增加了11%和16%。(5)转基因大豆GSDH5植株与受体品种‘东农50’在单株荚数、百粒重、株高、结荚习性、花色、叶形等方面均无显著差异,证明HSSP基因的插入对大豆植株的生长发育无不良影响。研究认为,转基因大豆GSDH5材料具备进一步培育成高含硫氨基酸大豆新品种的潜力。  相似文献
