Endogenous stages of Eimeria sigmodontis were studied in experimentally infected cotton rats, Sigmodon hispidus. The parasites were located in mucosal epithelial cells of the cecum and colon. E. sigmodontis had a typical coccidian endogenous cycle that consisted of three asexual schizogonous stages and sexual stages composed of the macrogametes and microgametes. The first-, second-, and third-generation schizonts contained 10–17, 5–8 and 10–21 merozoites, respectively. Only one type of wall-forming body was present in the macrogamete. The microgametocyte had a monocentric type of development.  相似文献

Sublethal doses of gamma radiation are known to inhibit total RNA synthesis in the ciliate protozoan Tetrahymena. To determine if the synthesis of a particular class of RNA is preferentially inhibited, pulse-labeled RNA was isolated from normal exponentially growing cells, irradiated cells, and cells in which total RNA synthesis had recovered to the pre-irradiation level. The RNAs were analyzed by SDS-polyacrylamide gel electrophoresis and oligo(dT)-cellulose column chromatography. Inhibition of RNA synthesis primarily involves ribosomal RNA. However, radiation does not cause a delay in the processing of precursor rRNA or a preferential loss of either of the mature rRNAs. Following irradiation, poly(A)-containing RNA [poly(A+)RNA] is synthesized at a rate up to three times greater than the control rate. The elevated poly(A+)RNA synthesis occurs during the period of depressed rRNA synthesis and even after rRNA synthesis has recovered to its pre-irradiation rate. While the sizes of the total cellular ribonucleoside triphosphate pools are depressed in the irradiated cells, these pools probably do not represent the actual compartments containing the precursors for RNA synthesis, and the observed changes cannot explain the modifications in macromolecular synthesis in irradiated Tetrahymena.  相似文献

A nicotinamide adenine dinucleotide (NAD)-dependent coniferyl alcohol dehydrogenase was enriched 1,200-fold from crude extracts ofRhodococcus erythropolis. The purification procedure involved ion exchange chromotography, gel filtration on Biogel A 1,5 and Sephadex G-200, and hydroxyapatite treatment. The enzyme had a molecular weight of approximately 200,000 and displayed maximal activity at pH 9.0. The apparentK m values for NAD and coniferyl alcohol were, respectively, 0.22 and 0.645 mM. Nicotinamide adenine dinucleotide phosphate (NADP) could only partially replace NAD. The enzyme was active with vanillyl alcohol and aromatic alcohols bearing the ,-unsaturated side chain of coniferyl alcohols. These aromatic alcohols included the dilignols dehydrodiconiferyl alcohol and guaiacylglycerol--coniferyl ether.  相似文献

(1-14C) Eicosatetraenoic (Arachidonic) acid was incubated wiht microsomes from rabbit renal cortex and NADPH (1 mM) for 15 min at 37°C. The products were extracted and purified by high pressure liquid chromatography. Some of the most polar metabolites were identified by gas chromatography mass spectrometry. They were 11, 12, 19- and 11, 12,20-trihydroxy-5,8-14-eicosatrienoic acid, 14,15,19- and 14,15,20- trihydroxy-5,8,11-eicosatrienoic acid, and 11,12-dihydroxy-19-oxo- 5,8,14-eicosatrienoic acid. These products were likely formed by ω- and (ω−1)-hydroxylation of 11,12-dihydroxy-5,8,14-eicosatrienoic aic and 14,15-dihydroxy-5,8,11-eicosatrienoic acid, two recently identified metabolites of arachidonic acid in fortified rabbit kidney microsomes.  相似文献

The composition of tryptic peptides was determined for Hb (major or fast electrophoretic component) from Clethrionomys rutilus and from Ondatra zibethicus. Analysis of the separated and β-Hb chains made possible the definition of theT9 and βT12 peptides and the resolution of uncertainties in earlier Microtus data for peptides T9,T12b,βT9,βT12 and βT13. The previous postulate of two deletions in peptide βT5 is supported by the present data, and the independent dipeptide βT6 appears in the Ondatra Hb. A Microtus stem sequence is inferred and used for comparison with the new data, and cladograms are given to accomodate the extended phylogenetic progression.  相似文献

[3H]dGMP-3'-labelled, activated salmon testis DNA and [32P]dGMP-5'-labelled open circular M13 DNA were reacted with cis-diamminedichloroplatinum(II), cis-diamminechloroaquaplatinum(II), cis-diamminediaquaplatinum(II) or trans-diamminechloroaquaplatinum(II). The reaction was arrested after arbitrary times by adjustment to slightly alkaline solution conditions. The platinum-containing DNA was digested with Escherichia coli DNA polymerase I. The progress of nucleotide release was measured by acid precipitation of undigested DNA. Solubilized nucleotides and adducts were analyzed by HPLC. The 3'-5'-exonuclease activity liberated single-coordinated dGMP-platinum(II) adducts from both cis- and trans-platinum(II) treated salmon testis DNA and a small fraction of adducts of cis-platinum(II) that coordinated two molecules of dGMP. The bisadduct was derived from non-neighboring guanine residues probably located at or close to 3'-termini. This nuclease activity neither cut between nor after neighboring guanine residues crosslinked by cis-platinum(II). No bisadduct was liberated for trans-platinum(II). The 5'-3'-exonuclease activity did not liberate any nucleotide adducts from cis-platinum(II)-treated DNa. However, it removed single-coordinated guanine adducts of trans-diamminedichloroplatinum(II). From the kinetics of the appearance of dGMP monoadducts and the inhibition of digestion, a reaction scheme is formulated for the reaction of platinum(II) complexes with DNA that confirms and extends the previously published one [W. Schaller, H. Reisner & E. Holler (1987) Biochemistry 26, 943-950]. The longevity of the dGMP monoadduct intermediate is discussed in the context of the efficiency of cis-diamminedichloroplatinum(II) as an antitumor drug.  相似文献

Water relation parameters of leaf cells of the aquatic plant Elodea densa have been measured using the pressure probe. For cells in both the upper and lower epidermis it was found that the elastic modulus () and the hydraulic conductivity (Lp) were dependent on cell turgor (P). Lp was (7.8±5.5)·10-7 cm s-1 bar-1 (mean±SD; n=22 cells) for P>4 bar in cells of the upper epidermis and was increasing by a factor of up to three for P0 bar. No polarity of water movement or concentration dependence of Lp was observed. For cells of the lower epidermis the Lp-values were similar and the hydraulic conductivity also showed a similar dependence on turgor. No wall ingrowth or wall labyrinths (as in transfer cells) could be found in the cells of the lower epidermis. The elastic modulus () of cells of the upper epidermis could be measured over the whole pressure range (P=0–7 bar) by changing the osmotic pressure of the medium. increased linearly with increasing turgor and ranged between 10 and 150 bar. For cells of the lower epidermis the dependence of on P was similar, although the pressure dependence could not be measured on single cells. The Lp-values are compared with literature data obtained for Elodea by a nuclear magnetic resonance (NMR)-technique. DNA topoisomerase II (topo II) is the target of many anticancer drugs and is often altered in drug-resistant cell lines. In some tumor cell lines truncated isoforms of topo IIalpha are localized to the cytoplasm. To study the localization and function of individual enzyme domains, we have epitope-tagged several fragments of human topo IIalpha and expressed them by retroviral infection of rodent and human cells. We find that fusion of the topo II fragments to the hydrophobic tail of human liver cytochrome b5 anchors the fusion protein to the outer face of cytoplasmic membranes, as determined by colocalization with calnexin and selective detergent permeabilization. Moreover, whereas the minimal ATPase domain (aa 1-266) is weakly and diffusely expressed, addition of the cytb5 anchor (1-266-b5) increases its steady-state level 16-fold with no apparent toxicity. Similar results are obtained with the complete ATPase domain (aa 1-426). A C-terminal domain (aa 1030-1504) of human topo IIalpha containing an intact dimerization motif is stably expressed and accumulates in the nucleus. Fusion to the cytb5 anchor counteracts the nuclear localization signal and relocalizes the protein to cytoplasmic membranes. In conclusion, we describe a technique that stabilizes and targets retrovirally expressed proteins such that they are exposed on the cytoplasmic surface of cellular membranes. This approach may be of general use for regulating the nuclear accumulation of drugs or proteins in living cells.  相似文献
