Antibiotic resistance is a global current threat of increasing importance. Moreover, biofilms represent a medical challenge since the inherent antibiotic resistance of their producers demands the use of high doses of antibiotics over prolonged periods. Frequently, these therapeutic measures fail, contributing to bacterial persistence, therefore demanding the development of novel antimicrobials. Esters of bicyclic amines (EBAs), which are strong inhibitors of Streptococcus pneumoniae growth, were initially designed as inhibitors of pneumococcal choline-binding proteins on the basis of their structural analogy to the choline residues in the cell wall. However, instead of mimicking the characteristic cell chaining phenotype caused by exogenously added choline on planktonic cultures of pneumococcal cells, EBAs showed an unexpected lytic activity. In this work we demonstrate that EBAs display a second, and even more important, function as cell membrane destabilizers. We then assayed the inhibitory and disintegrating activity of these molecules on pneumococcal biofilms. The selected compound (EBA 31) produced the highest effect on S. pneumoniae (encapsulated and non-encapsulated) biofilms at very low concentrations. EBA 31 was also effective on mixed biofilms of non-encapsulated S. pneumoniae plus non-typeable Haemophilus influenzae, two pathogens frequently forming a self-produced biofilm in the human nasopharynx. These results support the role of EBAs as a promising alternative for the development of novel, broad-range antimicrobial drugs encompassing both Gram-positive and Gram-negative pathogens. 相似文献

In the frugivory networks of many arid and semi‐arid Mesoamerican ecosystems, columnar cacti act as keystone species that produce fruits with a high content of water and nutrients attractive to numerous vertebrates. The aim of this investigation was to assess the fruit removal patterns of two guilds of frugivores on the fruits of the woolly torch Pilosocereus leucocephalus. We assessed fruit pulp removal in two ways: by estimating the consumption of seeds given the amount of pulp removed per visit and by estimating the percentage of pulp removal over time. We put exclosures on unripe, intact fruits to keep frugivores from removing material. Once ripe, we removed the exclosures and tracked animal visitation of 69 fruits using camera traps. We obtained a total of 2,162 hr of footage (14:47 hours of them with effective pulp removal). The highest number of visitors is that of diurnal species (n = 12, all birds) versus only four nocturnal (three bats, one rodent). The most effective species in pulp removal are birds. Bats play a modest role in frugivory of this cactus. The significance of this work is twofold: (a) birds and bats consume the fruit pulp of this cactus and likely disperse its seeds, and (b) although bats rank high in pulp removal effectiveness, birds as a guild far outweigh their importance in this system, as they are not only more frequent but also remove more pulp and seeds. Both groups are known to be important in cacti seed dispersal, and our findings are essential in understanding the population dynamics of the woolly torch and in elucidating its seed dispersal ecology. 相似文献

The Magellanic Penguin (Spheniscus magellanicus) is the most abundant and widely distributed seabird breeding on the Patagonian Coast of Argentina. We combined conventional stomach content and stable isotope analysis to assess Magellanic Penguin diet during the chick rearing stage at the two northernmost colonies in an area subject to fisheries. In 2011 and 2013, Thornfish (Bovichtus argentinus) was the main prey by mass at Complejo Islote Lobos (63.0% and 32.3%, respectively) and Argentine Anchovy (Engraulis anchoita) at Estancia San Lorenzo (85.2 and 63.3%, respectively). Magellanic Penguins from both colonies showed low isotopic niche overlap in 2011 (36%) and no overlap in 2013, suggesting a different use of prey species and/or foraging areas. Stable isotope mixing models showed that Argentine Anchovy (52.8%) and Thornfish (65.9%) were the main prey at Complejo Islote Lobos in 2011 and 2013, respectively, while Patagonian Redfish (Sebastes oculatus) (46.4%) and Squat Lobster (Munida gregaria) (50%) were the main prey at Estancia San Lorenzo in 2011 and 2013, respectively. Results show that in addition to Argentine Anchovy, previously recognized as main prey for breeding Magellanic Penguins in northern Patagonia, other juvenile or small sized fish are important diet items. Diet results suggest different scenarios of food conditions for each colony, despite the relative short distance between breeding locations. The low contribution of Argentine Hake (Merluccius hubbsi) and Argentine Shortfin Squid (Illex argentinus) suggests a low trophic overlap with commercial fisheries. The information provided is key to understand changes in the marine ecosystem and potential penguin-fishery interactions. 相似文献

The Notothenia coriiceps liver is commonly infected with parasites, reducing the hepatic mass and inducing the regeneration. In order to better understand
the effect of nutrient influx on hepatic regeneration at 0°C, a usual mammal hepatotrophic factor (HF) solution was injected
into ten fish (HF group), while ten fish were injected with saline solution (control), twice a day, for 15 days. The liver
and carcass weight were measured, and samples were obtained for histological studies. The HF group presented a higher liver/carcass
weight (62.5%) than control group. This increase in liver mass was due mainly to hepatocytes hypertrophy, including nuclear
size increase and cytoplasmic inclusions of glycogen. Hyperplasia is also observed, although to a lesser extent. The hepatic
reaction to HF in Antarctic fish was here demonstrated for the first time, helping to understand the liver response to seasonal

Ceragenin CSA-13, a cationic steroid, is here reported to show a concentration-dependent bactericidal/bacteriolytic activity against pathogenic streptococci, including multidrug-resistant Streptococcus pneumoniae. The autolysis promoted by CSA-13 in pneumococcal cultures appears to be due to the triggering of the major S. pneumoniae autolysin LytA, an N-acetylmuramoyl-L-alanine amidase. CSA-13 also disintegrated pneumococcal biofilms in a very efficient manner, although at concentrations slightly higher than those required for bactericidal activity on planktonic bacteria. CSA-13 has little hemolytic activity which should allow testing its antibacterial efficacy in animal models. 相似文献

Aphid salivation in sieve elements and phloem sap ingestion were linked to waveforms in the Electrical Penetration Graph (EPG).
Non-viruliferousRhopalosiphum padi (L.) (Hemiptera, Aphididae) on barley yellow dwarf virus (BYDV) infected wheat could acquire the virus, which was used as
an indication for phloem sap ingestion, whereas virus inoculation by viruliferous aphids on healthy plants was associated
with salivation in sieve elements or other phloem cells. Probing was monitored and the waveforms recorded were related to
ELISA results of test plants. The EPG patterns A, B, and C are indicative of the stylet pathway phase, whereas patterns E1
and E2 reflect the phloem (sieve element) phase with an unknown activity (E1) or with ingestion and concurrent salivation
(E2). Aphids showing pathway and E1 rarely acquired virus, suggesting that little or no phloem sap ingestion can occur during
these patterns, whereas those showing additionally pattern E2 did so substantially, indicating phloem sap ingestion. The main
pattern related to virus inoculation was E1, although some aphids were able to inoculate plants during pathway. Pattern E1
clearly reflects the most important salivation into sieve elements. Pattern E2 had no clear contribution to virus inoculation,
supporting the present hypothesis that during this pattern the saliva is mixed with the phloem sap in the single canal at
the stylet tips and ingested immediately, without reaching the plant tissue. Sustained sap ingestion did not affect virus
inoculation. So, BYDV inoculation mainly occurs during the first period of a sieve element puncture which is always formed

As approaches to ecological restoration become increasingly large scale and collaborative, there is a need to better understand social aspects of restoration and how they influence land management. In this article, we examine social perspectives that influence the determination of ecological reference conditions in restoration. Our analysis is based on in‐depth interviews with diverse stakeholders involved in collaborative restoration of fire‐adapted forest landscapes. We conducted interviews with 86 respondents from six forest collaboratives that are part of the U.S. Forest Service's Collaborative Forest Landscape Restoration Program. Collaboratives use a variety of approaches to develop reference conditions, including historic, contemporary, and future scenarios. Historical conditions prior to European settlement (nineteenth century or “pre‐settlement” conditions), or prior to more recent grazing, logging, and exclusion of fire, were the predominant type of reference used in all sites. Stakeholders described benefits and limitations of reference conditions. Primary benefits include (1) providing a science‐based framework for bringing stakeholders together around a common vision; (2) gaining social understanding and acceptance of the underlying need for restoration; and (3) serving to neutralize otherwise value‐laden discussions about multiple, sometimes competing, resource objectives. Limitations stem from (1) concerns over social conflict when reference conditions are perceived to contradict other stakeholder values and interests, (2) differing interpretations of reference condition science, (3) inappropriate application or over‐generalization of reference information, and (4) limited relevance of historical references for current and future conditions in some ecosystems.
