In order to determine the role of the plasma membrane H+-ATPase and alternative oxidase (alternative pathway of respiration) in the regulation of intracellular pH during development of the tobacco male gametophyte, we studied the changes in pH due to the inhibition of these enzymes by orthovadanate and benzhydroxamic acid, respectively. The inhibition of these enzymes decreased the intracellular pH at all three studied stages of the male gametophyte development: middle and late binuclear pollen grains and activated mature pollen grain. The data obtained suggest that H+-ATPase and alternative oxidase are involved in the regulation of intracellular pH of the pollen grain during its differentiation and activation that precede germination. At the same time, during the recovery of intracellular pH after its acidification by propionic acid, it was found that other mechanisms, not related to the above mentioned, greatly contribute to the regulation of pH. 相似文献

A simple test-system has been developed for the first time in order to detect the ability of effectors (lipoplexes) to activate the complement system in an antibody-independent manner to serve as acceptors of nascent C4b and to inhibit formation of the key enzyme of complement, C3-convertase. The effect of plasmid DNA (pCMV-SPORT-LacZ), negatively charged cardiolipin (CL), neutral phosphatidylcholine (PC) vesicles and their lipoplexes, on the complement system was studied using the method developed. It was revealed that PC vesicles did not affect the complement system, while CL vesicles manifested low activation. The influence of plasmid DNA and its lipoplex based on PC liposomes as well on the complement system was very low. PC/LacZ lipoplex (143 microg/ml) acted on the complement system like 5.36 microg/ml heat aggregated IgG (agg) (the level of no pathological ruptures), whereas CL/LacZ lipoplex (143 microg/ml) acted similar to 10.7 microg/ml IgG (agg). Thus, weak activation of the complement system with CL lipoplex, and even weaker for the PC lipoplex testified to the use of neutral and positively charged lipoplexes preferably in gene therapy protocols. Resonant Raman scattering was used as a novel, rapid, non-destructive optical technique to measure zeaxanthin levels in Flavobacterium multivorum ATCC 55238. Culture broth, after bacterial growth for 40 h, exhibited characteristic resonance Raman vibrational modes at 1159 cm–1 (C-C stretch) and 1525 cm–1 (C=C stretch) upon excitation at 488 nm. A striking correlation was observed between the carotenoid level as estimated by HPLC and by resonance Raman spectroscopy. 相似文献

The topological structure of the simplest critical fragments in biochemical systems has been characterized. The conditions are considered where the critical fragments induce oscillations of the concentrations of the system participants. To illustrate, three biochemical systems (transport of ions through a membrane, protein phosphorylation, and two-substrate reaction) are discussed. The kinetic schemes of these systems contain one of the discovered critical fragments. The hornet is an endothermic insect. Daily variations in hornet surface temperature were measured. Three peaks were found between 9:30 and 10: 30 a.m., 11 and 12 a.m. and between 2 and 3 p.m. Electrical current and voltage values were highest along the head. Electrical current along the gaster and the head flowed towards the thorax, i.e., from body parts with minimal temperature towards the body part with maximal temperature. Current and voltage values measured across the cuticle of the gaster were about 5nA and 100 mV, respectively, and these were of the same order of magnitude as the current and voltage values along the cuticle. It was found that: 1) temperature regulation most probably originates in the thorax and 2) there is a correlation between the temperature distribution along the hornet body surface and levels of the cuticular electrical signals. 相似文献

Social insects, belonging to the order Hymenoptera, maintain a fixed, optimal temperature in their nest. Thus, in social wasps and hornets, the optimal nest temperature is 29 degrees C, despite the fact that they are distributed in regions of varying climates both in the northern and southern hemispheres of the globe. Since hornets and bees are relatively small insects, determination of their own body temperature as well as that of their nest and the brood was made via thermometers or by the use of infrared (IR) rays. It has been suggested that thermoregulation in social insect colonies is effected primarily by the adult insects via muscle activation, that is, fluttering of their wings, which can raise both their own and the ambient temperature by many degrees centigrade. However, the larval brood can also contribute to the thermoregulation by acting as heat resources and thereby raising the ambient temperature by 1-2 degrees C. To this end, the adult hornets are endowed with a well-developed musculature and their larvae, too, have muscles that enable them to move about. Not so the hornet pupae which are enclosed in a silk envelope (the cocoon), with a rather thick silk cap spun by the pupating larvae, and have rather undeveloped muscles. In the latter instance, it stands to reason that the pupae benefit from the nest warming achieved primarily by the adult hornets, but how is the information regarding their thermal needs relayed from them to the adults? Previously we showed that the adult hornets are attracted to the pupae by pheromones released by the latter, but such chemical compounds can only convey information of a general nature and we are still left with the question as to how the adult hornet can gauge or ascertain the temperature of a single insulated pupa. The present study provides evidence that the hornet pupa can indeed transmit information regarding its body temperature via electrical means. 相似文献

Sufficient conditions for existence of concentration oscillations of components of complex reactions are considered on the basis of graph theory. Graphical rules were developed for detecting oscillators in kinetic schemes of complex biochemical systems. The main types of topology and principles of construction of kinetic schemes of oscillators containing two, three, and four substances are considered. The resulting oscillators might be a part of the biochemical systems of the n-th order. In the present work it is shown that the phenomenon of interphase chromosome centromeric region displacement, earlier revealed by the authors, is not realized in G0-lymphocytes with heterozygous BRCA1/2 gene mutations. The role of these genes in DNA double strand break (DSB) reparation is known. It is concluded that chromosome locus displacement is necessary for DSB repair, at least in the process of homologous recombination. In accordance with our data, some feature (pericentromeric cluster disintegration and displacement, the nucleus size increasing) characteristic for S- and G0-lymphocytes are observed in normal G0-lymphocytes treated with 3 and 10 cGy. However, the size of nucleus in G0-lymphocytes is restored through 6 hours after irradiation in opposite to the process in dividing cells. It was proposed that some typical for resting cell functions of G0-lymphocytes after inducing by adaptive doze of radiation are stopped as similarly as after stimulation of cells. Interestingly, that the process of the induced chromosome loci displacement is correlated with the decreasing of DNA reparation possibilities under UV-irradiation. The induced apoptosis level also decreases when chromosome loci are displaced. The possible mechanisms of the revealed phenomenon are discussed.
