Sugar-beet plants ( Beta vulgaris L. cv. Monohill) were cultivated for 4 weeks in a complete nutrient solution. Indirect effects of cadmium were studied by adding 5, 10 or 20 μ M CdCl2 to the culture medium while direct effects were determined by adding 1, 5, 20, 50 or 2 000 μ M CdCl2 to the assay media. The photosynthetic properties were characterized by measurement of CO2 fixation in intact plants, fluorescence emission by intact leaves and isolated chloroplasts, photosystem (PS) I and PSII mediated electron transport of isolated chloroplasts, and CO2-dependent O2 evolution by protoplasts. When directly applied to isolated leaves, protoplasts and chloroplasts. Cd2+ impeded CO2 fixation without affecting the rates of electron transport of PSI or PSII or the rate of dark respiration. When Cd2+ was applied through the culture medium the capacity for, and the maximal quantum yield of CO2 assimilation by intact plants both decreased. This was associated with: (1) decreased total as well as effective chlorophyll content (PSII antennae size), (2) decreased coupling of electron transport in isolated chloroplasts, (3) perturbed carbon reduction cycle as indicated by fluorescence measurements. Also, protoplasts isolated from leaves of Cd2+-cultivated plants showed an increased rate of dark respiration.  相似文献

Optimal research and management of species with age structure often depends on estimates of age-specific population parameters, which in turn depends on reliable methods for age determination. By counting annuli in the cementum of incisor root tips from 51 known-age moose (Alces alces) between 1 and 12 years old, we examined the variation in accuracy and repeatability of age estimates provided by three research technicians with different experiences of aging moose. The most experienced technician estimated the moose age correctly in up to 90% of the cases, while the technician with no prior experience estimated age correctly in up to 73% of the cases. The medium-experienced technician achieved a lower accuracy (up to 53%), indicating that experience alone is not sufficient if the basic training or lack of routine checks against other colleagues or a known-age material are not undertaken. The percentage moose aged within ±1 year from correct age was significantly higher in all technicians (94–98%). After the generally high accuracy, we also found high repeatability (0.980–0.994) within technicians. We conclude that this method of age-determination provides reliable estimates that can be used by management and research to gain information about age-specific patterns in moose populations. However, to obtain estimates of high accuracy the technicians should be well trained, have gained the necessary experience, and most preferably, have access to a sample of teeth from known-age moose.  相似文献

Methylating agents introduce cytotoxic 1-methyladenine (1-meA) and 3-methylcytosine (3-meC) residues into nucleic acids, and it was recently demonstrated that the Escherichia coli AlkB protein and two human homologues, hABH2 and hABH3, can remove these lesions from DNA by oxidative demethylation. Moreover, AlkB and hABH3 were also found to remove 1-meA and 3-meC from RNA, suggesting that cellular RNA repair can occur. We have here studied the preference of AlkB, hABH2 and hABH3 for single-stranded DNA (ssDNA) or double-stranded DNA (dsDNA), and show that AlkB and hABH3 prefer ssDNA, while hABH2 prefers dsDNA. This was consistently observed with three different oligonucleotide substrates, implying that the specificity for single-stranded versus double-stranded DNA is sequence independent. The dsDNA preference of hABH2 was observed only in the presence of magnesium. The activity of the enzymes on single-stranded RNA (ssRNA), double-stranded RNA (dsRNA) and DNA/RNA hybrids was also investigated, and the results generally confirm the notion that while AlkB and hABH3 tend to prefer single-stranded nucleic acids, hABH2 is more active on double-stranded substrates. These results may contribute to identifying the main substrates of bacterial and human AlkB proteins in vivo.  相似文献

The fluorescence kinetics of both intact needles and isolated chloroplasts of summer active and winter stressed Pinus sylvestris were measured at both room temperature and 77 K. It was confirmed that winter stress inhibited the photochemical capacity of photosystem II but also that winter stress caused the strongest inhibition of the electron transport at the site where the plastoquinone pool is reduced. Parallel analyses of the fluorescence characteristics of photosystem II (F693) and photosystem I (F729) during photosystem II trap closure furthermore revealed that the yield of spillover of excitation energy from photosystem II to photosystem I decreased upon winter stress. We suggest that this is because of an increased radiationless decay of excitation energy both at the reaction center and antennae levels of photosystem II. There is, however, also a possibility that the decreased yield of spill-over is accentuated by a partial detachment of the light harvesting chlorophyll a/b complex from photosystem II upon winter stress.Paper presented at the FESPP meeting in Strasbourg (1984).  相似文献

Photoinhibition was examined in naturally exposed willow leaves in the field. In the afternoon on clear and warm days, the quantum yield of electron transport, derived from gas exchange data, was decreased by 28%. Besides this photoinhibition, decreases in the photosynthetic capacity and in the stomatal conductance were also observed. Of these three limitations of carbon assimilation, photoinhibition was the major one at limiting light only.To investigate the generality of photoinhibition, shade- and sun-acclimated leaves of fourteen different species were compared in a laboratory study. Photoinhibition was quantified by fluorescence measurements following exposure to moderate and high light for 30 min. The extent of photoinhibition was inversely related to the photochemical quenching, qp, during exposure (the proportion of open PS II traps). This relationship was the same independent of the species, the light-acclimation state of the leaf and the light intensity. However, sun- and shade-acclimated leaves occupied opposite sides of the relationship: the sun-leaves showed lower photoinhibition and higher qp. The peripheral light-harvesting antenna complex (LH2) of purple photosynthetic bacteria is an ideal testing ground for models of structure–function relationships due to its well-determined molecular structure and ultrafast energy deactivation. It has been the target for numerous studies in both theory and ultrafast spectroscopy; nevertheless, certain aspects of the convoluted relaxation network of LH2 lack a satisfactory explanation by conventional theories. For example, the initial carotenoid-to-bacteriochlorophyll energy transfer step necessary on visible light excitation was long considered to follow the Förster mechanism, even though transfer times as short as 40 femtoseconds (fs) have been observed. Such transfer times are hard to accommodate by Förster theory, as the moderate coupling strengths found in LH2 suggest much slower transfer within this framework. In this study, we investigate LH2 from Phaeospirillum (Ph.) molischianum in two types of transient absorption experiments—with narrowband pump and white-light probe resulting in 100 fs time resolution, and with degenerate broadband 10 fs pump and probe pulses. With regard to the split Qx band in this system, we show that vibronically mediated transfer explains both the ultrafast carotenoid-to-B850 transfer, and the almost complete lack of transfer to B800. These results are beyond Förster theory, which predicts an almost equal partition between the two channels.  相似文献

Empirical evidence strongly indicates that human exploitation has frequently led to rapid evolutionary changes in wild populations, yet the mechanisms involved are often poorly understood. Here, we applied a recently developed demographic framework for analyzing selection to data from a 20‐year study of a wild population of moose, Alces alces. In this population, a genetic pedigree has been established all the way back to founders. We demonstrate harvest‐induced directional selection for delayed birth dates in males and reduced body mass as calf in females. During the study period, birth date was delayed by 0.81 days per year for both sexes, whereas no significant changes occurred in calf body mass. Quantitative genetic analyses indicated that both traits harbored significant additive genetic variance. These results show that selective harvesting can induce strong selection that oppose natural selection. This may cause evolution of less favorable phenotypes that become maladaptive once harvesting ceases.  相似文献
