Abstract The primary structure of the 16S rDNA of Acidothermus cellulolyticus , a cellulolytic, asporogenous rod isolated from a hot spring, was determined by automated sequence analysis. This Gram-variable organism is not related to members of other aerobic thermophilic genera found in similar environments, such as Thermus and Thermomicrobium , but clusters within the phylogenetically defined radiation of the actinomycetes where it represents an individual subline of descent. 相似文献

The aim of this study was to investigate the microbial community thriving at two shallow hydrothermal vents off Panarea Island
(Italy). Physico-chemical characteristics of thermal waters were examined in order to establish the effect of the vents on
biodiversity of both Bacteria and Archaea. Water and adjacent sediment samples were collected at different times from two
vents, characterised by different depth and temperature, and analysed to evaluate total microbial abundances, sulphur-oxidising
and thermophilic aerobic bacteria. Total microbial abundances were on average of the order of 105 cells ml−1, expressed as picoplanktonic size fraction. Picophytoplanktonic cells accounted for 0.77–3.83% of the total picoplanktonic
cells. The contribution of bacterial and archaeal taxa to prokaryotic community diversity was investigated by PCR–DGGE fingerprinting
method. The number of bands derived from bacterial DNA was highest in the DGGE profiles of water sample from the warmest and
deepest site (site 2). In contrast, archaeal richness was highest in the water of the coldest and shallowest site (site 1).
Sulphur-oxidising bacteria were detected by both culture-dependent and -independent methods. The primary production at the
shallow hydrothermal system of Panarea is supported by a complex microbial community composed by phototrophs and chemolithotrophs. 相似文献

In the search for novel Xenorhabdus strains in a recently described nematode species, Steinernema thermophilum, three strains (strain 28(T) = DSM 17382(T), strain 42 = DSM 17383 and strain 43 = DSM 17384) were isolated from three independent isolation approaches from crushed mixture of infective juveniles. 16S rRNA gene sequence comparison of strains 28(T) and DSM 17383 indicated identity and the phylogenetic position pointed towards an individual taxon within the phylogenetic dendrogram of Xenorhabdus type strains. The nearest phylogenetic relatives of strain 28(T) were Xenorhabdus poinarii and Xenorhabdus szentirmaii (97.7% each). The three isolates were almost identical in reaction towards the API and BIOLOG substrate panels but differed in their reactions from those of the established type strains of the genus Xenorhabdus. These clear genomic and metabolic differences let us propose a new species, Xenorhabdus indica sp. nov. for the three clones. The type strain is strain 28(T), DSM 17382(T), CIP 108830(T). 相似文献

A strain EG19T of aerobic bacteria able to form pleomorphic cells was isolated from a brine spring runoff stream in the west central region
of the province of Manitoba, Canada. The pale pinkish purple strain contained bacteriochlorophyll a incorporated into light-harvesting I and reaction center complexes. Its inability to grow under anaerobic illuminated conditions
prompted designation as a member of the functional group known as aerobic anoxygenic phototrophic bacteria. Phylogenetic analysis
of the 16S rRNA gene sequence revealed that it belonged to the Gammaproteobacteria, forming a distinct branch of phototrophs distantly related to most described aerobic anoxygenic phototrophs, quite marginally
related (95.6%) both to the only other described gammaproteobacterial aerobic phototroph, Congregibacter litoralis, and also to nonphototrophs in the genus Haliea (95.1–96.1%). Physiological tests demonstrated tolerance profiles to salinity (0–18% NaCl), pH (7–12), and temperature (7–40°C)
consistent with survival in a shallow hypersaline stream on the exposed, vegetation-depleted salt playa of its native East
German Creek. Phylogenetic data and phenotypic properties such as pigment composition, morphology, and physiology support

Microbial diversity of cultivatable bacteria associated with the North Sea bryozoan Flustra foliacea
The microbial diversity of cultivatable bacteria associated with the bryozoan species Flustra foliacea from the North Sea was investigated by a molecular approach. Amplified ribosomal RNA restriction analyses (ARDRA) and 16S rDNA partial sequence analysis revealed differences in the composition of cultivatable bacteria populations from single bryozoan colonies collected from two different sampling sites in the North Sea as well from one site taken at different points in time. Whereas gamma-Proteobacteria identified as Shewanella frigidimarina, Pseudoalteromonas ssp. and Psycbrobacter ssp. were predominant on samples of Flustra I (taken near the island of Helgoland), most bacteria isolated from Flustra II, originating from the Steingrund, could be affiliated to Gram-positive taxa. Survey of the bryozoan samples from the latter site in February 2000 led to the detection of a phylogenetically mixed bacterial population, consisting of gamma-, and alpha-Proteobacteria and Gram-positive bacteria with low and high GC-content (Flustra III). As these bacteria are among the most widely isolated organisms from the marine environment, it may be concluded that the bryozoan Flustra foliacea accepts colonization of surfaces by bacteria which are common inhabitants of the marine environment and which may have been transferred into this environment from terrestrial sites. 相似文献

Abstract A 16S rDNA study was performed on at least two species, including the type strain of the type species, of each genus described to possess peptidoglycan of Group B. Analyses confirm that these actinomycetes form a phylogenetically coherent cluster within the arthrobacteria subline of descent. While members of the genera Agromyces, Clavibacter, Curtobacterium and Rathayibacter appear to be phylogenetically coherent, members of Microbacterium and Aureobacterium do not cluster according to their taxonomic affiliation but form one large cluster in which species of both genera are intermixed. Species with no taxonomic standing, i.e. "Corynebacterium aquaticum" , and "Brevibacterium helvolum" form three separate sublines of descent and can be considered nuclei of future novel genera. 相似文献

A novel type of purple sulfur bacterium was isolated from a hypersaline sulfur spring on the shore of the Dead Sea. The cells of the isolate are irregularly rod-shaped or curved, and motile by means of a tuft of polar flagella. The photosynthetic system, containing bacteriochlorophyll a and carotenoids of the spirilloxanthin series, is located on stacks of lamellar membranes in the cell cytoplasm. The organism can grow either photoautotrophically with sulfide as electron donor, which is oxidized via extracellular sulfur to sulfate, or photoheterotrophically, using acetate, succinate, fumarate, malate or pyruvate as carbon sources. The bacterium is obligately anaerobic, and requires a source of reduced sulfur for growth. The isolate is moderately halophilic, and grows optimally at NaCl concentrations between 3 and 8%, temperatures between 30 and 45°C, and neutral pH. 16S ribosomal RNA oligonucleotide cataloging suggests a close relationship to purple sulfur bacteria of the genus Ectothiorhodospira. As the isolate differs greatly from the described members of the genus Ectothiorhodospira, we describe the isolate as a new species, and propose the name Ectothiorhodospira marismortui sp. nov. 相似文献
