The mechanism of luminol chemiluminescence is a special case of nucleophilic addition to carbonyl compounds. The breakdown of the key intermediate, an alpha hydroxy hydroperoxide, produces a peracid ortho to an acyl diazene group. After intramolecular addition of the peracid, the energy from nitrogen expulsion is utilized in the formation of an anti-aromatic endoperoxide. Rupture along the O,O bond leaves a substantial part of the ensuing phthalate in its excited state. The emitter is shown to be a mono-protonated phthalate unaccessible by photoexcitation. The dark reaction is a concerted decomposion of the alpha hydroxy hydroperodixe to yield ground-state phthalate.  相似文献

Spring harvest rates of male wild turkeys (Meleagris gallapavo) influence the number and proportion of adult males in the population and turkey population models have treated harvest as additive to other sources of mortality. Therefore, hunting regulations and their effect on spring harvest rates have direct implications for hunter satisfaction. We used tag recovery models to estimate survival rates, investigate spatial, temporal, and demographic variability in harvest rates, and assess how harvest rates may be related to management strategies and landscape characteristics. We banded 3,266 male wild turkeys throughout New York, Ohio, and Pennsylvania during 2006–2009. We found little evidence that harvest rates varied by year or management zone. The proportion of the landscape that was forested within 6.5 km of the capture location was negatively related to harvest rates; however, even though the proportion forested ranged from 0.008 to 0.96 across our study area, this corresponded to differences in harvest rates of only 2–5%. Annual survival was approximately twice as high for juveniles as adults . In turn, spring harvest rates for adult turkeys were greater for adults than juveniles . We estimated the population of male turkeys in New York and Pennsylvania ranged from 104,000 to 132,000 in all years and ranged from 63,000 to 75,000 in Ohio. Because of greater harvest rates for adult males, the proportion of adult males in the population was less than in the harvest and ranged from 0.40 to 0.81 among all states and years. The high harvest rates observed for adults may be offset by greater recruitment of juveniles into the adult age class the following year such that these states can sustain high harvest rates yet still maintain a relative high proportion of adult males in the harvest and population. © 2011 The Wildlife Society.  相似文献

The production of acid phosphatases (E.C.3.1.3.2, ACPs) by Aspergillus niger N402A is regulated by specific growth rate, as well as phosphate availability and pH, as demonstrated by studies in continuous flow culture. Specific ACP activity was highest when A. niger was grown at pH 6.3 (64±8 U g−1) or pH 2.8 (99±11 U g−1), at a dilution rate of 0.07 h−1 and phosphate concentrations below 0.46 mM. ACP production was growth correlated for specific growth rates between 0.07 and 0.13 h−1. Four different ACPs, including two phytases, were produced by A. niger N402A. The ACP and the phytase with maximal activities at pH 5.5 were differentially expressed at different culture pH values, with greater production at low pH.  相似文献

Aims:  To characterize the secretion of proteolytic activities against keratin, collagen and elastin in liquid cultures of Bacillus cereus IZ-06b and IZ-06r isolated from wool.
Methods and Results:  Growth of B. cereus IZ-06b and IZ-06r were characterized in batch culture. Both strains needed an organic nitrogen source, were able to grow on wool or peptone as sole carbon and nitrogen sources, and metabolized glucose, maltose and other simple sugars. Proteolytic activities were investigated in batch cultures grown in peptide-restricted, carbon-sufficient medium. Secretion of proteases was induced by peptide limitation while different proteolytic activities appeared sequentially in the growth medium. When the most available components of the peptone were depleted, collagenolytic and elastolytic proteases were produced. These were later replaced by the production of keratinolytic protease.
Conclusions:  B. cereus can adjust its proteolytic affinity profile in response to the supply of organic nitrogen and sequentially secrete proteases with activities targeted against increasingly inaccessible proteinous substrates as the nutritional availability in the environment deteriorates.
Significance and Impact of the Study:  Peptide-limited, carbon-sufficient growth media containing no proteinous substrates are well suited for protease production in B. cereus while growth conditions can be adjusted to optimize the proteolytic affinity profiles.  相似文献

Epidemiological evidence suggests that chronic use of non-steroidal anti-inflammatory drugs (NSAIDs) reduces the risk of Alzheimer's disease. Recently, NSAIDs have been shown to decrease amyloid pathology in a transgenic mouse model of Alzheimer's disease. This benefit may be partially attributable to the ability of NSAIDs to selectively reduce production of the amyloidogenic A beta 42 peptide in both cultured cells and transgenic mice. Although this activity does not appear to require the action of cyclooxygenases in cultured cells, it is not known whether other NSAID-sensitive targets contribute to this A beta 42 effect. In this study, we have used both pharmacological and genetic means to determine if other known cellular targets of NSAIDs could mediate the reduction in A beta 42 secretion from cultured cells. We find that altered arachidonic acid metabolism via NSAID action on cyclooxygenases and lipoxygenases does not alter A beta 42 production. Furthermore, we demonstrate that alterations in activity of peroxisome proliferator-activated receptors, I kappa B kinase beta or nuclear factor kappa B do not affect A beta 42 production. Thus, NSAIDs do not appear to alter A beta 42 production indirectly through previously identified cellular targets and may interact directly with the gamma-secretase complex itself to affect amyloid production.  相似文献

Via cytoplasmic signal transduction pathways, cytokines induce a variety of biological responses and modulate the outcome of inflammatory diseases and malignancies. Crohn's disease is a chronic inflammatory bowel disease of unknown etiology. Perturbation of the intestinal cytokine homeostasis is believed to play a pivotal role, but the pathogenesis of Crohn's disease is not fully understood. Here, we study intestinal T cells from Crohn's disease and healthy volunteers. We show that STAT3 and STAT4 are constitutively activated in Crohn's patients but not in healthy volunteers. The activation is specific, because other STAT proteins are not constitutively activated. Furthermore, the STAT3 regulated protein, SOCS3, is also constitutively expressed in Crohn's patients but not in healthy volunteers. Taken together, these data provide evidence of abnormal STAT/SOCS signaling in Crohn's disease. This aberrant activation, so far noted only in malignant cells, establish a new critical approach for better understanding the immunopathogenesis of Crohn's disease.  相似文献

Photosynthesis in ozone-exposed duckweed (Lemna gibba)   总被引:2,自引:0,他引:2  
The photosynthetic light saturation curve in duckweed was lowered by 20–25% after ozone exposure (300 nmol mol−1, 1 h). The light flux and oxygen concentration during ozone-exposure had no effect on reduction of net photosynthesis. Net photosynthesis and photorespiration were both depressed by about 40% after exposure for 1 h to 360 nmol mol−1 ozone. We could not find any change in dark respiration after ozone exposure below 300 nmol mol−1. When the concentration of ozone was doubled from 150 nmol mol−1 to 300 nmol mol−1, the uptake of ozone in duckweed changed from 100 nmol m−2 s−1 to 170 nmol m−2 s−1. We found no differences in fluorescence (pattern) between ozone treated plants and the control plants during a period of 150 min after ozone treatment, but there was an increase in synthesis of the Dl-protein and a significant reduction in degradation after ozone treatment (300 nmol mol−1, 1 h). These results, together with fluorescence measurements, indicate that photochemical electron transport was not responsible for the ozone-induced reduction in net photosynthesis.  相似文献
