When far red light preincubated cells of Euglena gracilis are transferred to dark or light, chlorophyll fluorescence (F0 and Fm) decreases. Non-photochemical quenching in the dark is suggested to be induced partly by chlororespiration and partly by changes in the distribution of excitation energy between the photosystems. Depending on the light intensities it was possible to resolve the non-photochemical quenching into at least three different components. The slowest relaxation phase of non-photochemical quenching occurred only after exposure to high light and was assigned to photoinhibition. The other two components were an energy-dependent quenching (qE), and the one which we attribute to a spill over mechanism. We suggest that both photosystems use a common antenna system consisting of LHC I and LHC II proteins. In contrast to higher plants, qE in Euglena gracilis is independent of the xanthophyll cycle and an aggregation of LHC II. The Pax gene Pox meso (Poxm) was the first and so far only gene whose initial expression was shown to occur specifically in the anlage of the somatic mesoderm, yet its role in somatic myogenesis remained unknown. Here we show that it is one of the crucial genes regulating the development of the larval body wall muscles in Drosophila. It has two distinct functions expressed during different phases of myogenesis. The early function, partially redundant with the function of lethal of scute [l(1)sc], demarcates the ;Poxm competence domain', a domain of competence for ventral and lateral muscle development and for the determination of at least some adult muscle precursor cells. The late function is a muscle identity function, required for the specification of muscles DT1, VA1, VA2 and VA3. Our results led us to reinterpret the roles of l(1)sc and twist in myogenesis and to propose a solution of the 'l(1)sc conundrum'.  相似文献

Hantaviruses predominantly infect human endothelial cells and, in the absence of cell lysis, cause two diseases resulting from increased vascular permeability. Andes virus (ANDV) causes a highly lethal acute pulmonary edema termed hantavirus pulmonary syndrome (HPS). ANDV infection enhances the permeability of endothelial cells in response to vascular endothelial growth factor (VEGF) by increasing signaling responses directed by the VEGFR2-Src-VE-cadherin pathway, which directs adherens junction (AJ) disassembly. Here we demonstrate that inhibiting pathway-specific VEGFR2 and Src family kinases (SFKs) blocks ANDV-induced endothelial cell permeability. Small interfering RNA (siRNA) knockdown of Src within ANDV-infected endothelial cells resulted in an ~70% decrease in endothelial cell permeability compared to that for siRNA controls. This finding suggested that existing FDA-approved small-molecule kinase inhibitors might similarly block ANDV-induced permeability. The VEGFR2 kinase inhibitor pazopanib as well as SFK inhibitors dasatinib, PP1, bosutinib, and Src inhibitor 1 dramatically inhibited ANDV-induced endothelial cell permeability. Consistent with their kinase-inhibitory concentrations, dasatinib, PP1, and pazopanib inhibited ANDV-induced permeability at 1, 10, and 100 nanomolar 50% inhibitory concentrations (IC(50)s), respectively. We further demonstrated that dasatinib and pazopanib blocked VE-cadherin dissociation from the AJs of ANDV-infected endothelial cells by >90%. These findings indicate that VEGFR2 and Src kinases are potential targets for therapeutically reducing ANDV-induced endothelial cell permeability and, as a result, capillary permeability during HPS. Since the functions of VEGFR2 and SFK inhibitors are already well defined and FDA approved for clinical use, these findings rationalize their therapeutic evaluation for efficacy in reducing HPS disease. Endothelial cell barrier functions are disrupted by a number of viruses that cause hemorrhagic, edematous, or neurologic disease, and as a result, our findings suggest that VEGFR2 and SFK inhibitors should be considered for regulating endothelial cell barrier functions altered by additional viral pathogens.  相似文献

We focus on reconstructing a spatiotemporal scenario of diversification of a widespread South American species, the Pectoral Sparrow Arremon taciturnus (Aves: Passerellidae). This species is widely distributed in both the humid and the dry forests of South America and therefore provides an interesting model for understanding the connection between different biomes of South America. We examined nucleotide sequences of the mitochondrial genes Cytochrome b (cyt-b) and NADH subunit 2 (ND2) from 107 specimens, and one nuclear marker (intron 7 of the β-fibrinogen gene) from a subset of samples collected across the distribution ranges of A. t. taciturnus and A. t. nigrirostris. Six major lineages were recovered in the phylogenies that displayed high levels of variance of allele frequencies and corresponded to distinct geographical locations. The estimation of divergence times provided evidence that diversification of the six lineages of the Pectoral Sparrow occurred throughout the Late Pleistocene across major cis-Andean biomes and Amazonian interfluves. Our dataset for A. taciturnus provides further evidence that rivers in Amazonia constitute barriers promoting allopatric speciation, with occasional sharing of alleles among lineages, particularly those with adjacent distributions.  相似文献

Scytonemin is an ultraviolet radiation (UVR)-screening compound synthesized by some sheathed cyanobacteria exposed to high solar and sky radiation. It is primarily produced in response to UVA radiation, but certain environmental stresses can enhance synthesis. This study focuses on the effects of periodic desiccation on scytonemin synthesis in three desiccation-tolerant cyanobacterial strains, Nostoc punctiforme PCC 73102, Chroococcidiopsis CCMEE 5056 and Chroococcidiopsis CCMEE 246. Nostoc punctiforme and Chroococcidiopsis CCMEE 5056 exposed to UVA radiation produced more concentrated scytonemin screens when experiencing periodic desiccation (i.e. 1 day desiccated for every 2 days hydrated) than when continuously hydrated. A more concentrated scytonemin screen would reduce the amount of UVR damage accrued when cells are desiccated and metabolically inactive. This might allow the cyanobacteria to allocate more energy to systems other than UVR damage repair during rehydration, which would facilitate recovery. The scytonemin screen is extremely stable, remaining largely intact in the sheaths of desiccated N. punctiforme even when continuously exposed to UVA radiation for about 2 months. In contrast to the above findings, scytonemin synthesis in Chroococcidiopsis CCMEE 246, a strain that produces scytonemin constitutively under low visible light (no UVA), was partially inhibited by periodic desiccation.  相似文献

Computational and experimental research has revealed that auditory sensory predictions are derived from regularities of the current environment by using internal generative models. However, so far, what has not been addressed is how the auditory system handles situations giving rise to redundant or even contradictory predictions derived from different sources of information. To this end, we measured error signals in the event-related brain potentials (ERPs) in response to violations of auditory predictions. Sounds could be predicted on the basis of overall probability, i.e., one sound was presented frequently and another sound rarely. Furthermore, each sound was predicted by an informative visual cue. Participants’ task was to use the cue and to discriminate the two sounds as fast as possible. Violations of the probability based prediction (i.e., a rare sound) as well as violations of the visual-auditory prediction (i.e., an incongruent sound) elicited error signals in the ERPs (Mismatch Negativity [MMN] and Incongruency Response [IR]). Particular error signals were observed even in case the overall probability and the visual symbol predicted different sounds. That is, the auditory system concurrently maintains and tests contradictory predictions. Moreover, if the same sound was predicted, we observed an additive error signal (scalp potential and primary current density) equaling the sum of the specific error signals. Thus, the auditory system maintains and tolerates functionally independently represented redundant and contradictory predictions. We argue that the auditory system exploits all currently active regularities in order to optimally prepare for future events.  相似文献

Der gesamte freie Teil des Fruchtblattes scheidet Nektar ab; die Narbenflüssigkeit enthält außer Schleim auch Zucker und dient, wie die Beobachtung blumenbesuchender Bienen zeigte, ebenfalls als Nektar. Die Sekretion erfolgt als Diffusion durch die Epidermisaußenwände und durch die nicht abgehobene, dünne Kutikula. Da im Fruchtblattgewebe weder Lokalisation von Stärke und Zucker entsprechend den Stellen der Abscheidung noch eine histologische Differenzierung eines Nektariumgewebes vorhanden ist, kann das Blütennektarium der untersuchten Arten als relativ ursprünglich angesehen werden.Die Futterkörper, welche sich an den Innenstaminodien, den fertilen Staubblättern und andeutungsweise an der Blütenhülle vonCalycanthus florida befinden, wurden hinsichtlich ihres Baues und ihrer Inhaltsstoffe beschrieben. Als vorherrschender Reservestoff ist Stärke vorhanden. Da nachDiels die Futterkörper des nahe verwandtenCalycanthus occidentalis fettes Öl und Proteine enthalten, ist zugleich ein Fall gegeben, wo entsprechende Blütenteile nahe verwandter Arten verschiedene Stoffe specichern.An den Futterkörpern der Innenstaminodien wurde zu Anthesebeginn eine schwache Nektarabscheidung nachgewiesen. Durch diese für die Ökologie der Futterkörper wichtige Tatsache ließen sich auch bezüglich der bei der Rückbildung fertiler Staubblätter auftretenden Nektarabscheidung und der Beziehung von Futterkörpern zu Nektarien neue Gesichtspunkte finden.Mit 9 Textabbildungen.  相似文献

Flavonoids accumulate in plant vacuoles usually as O-glycosylated derivatives, but several species can also synthesize flavonoid C-glycosides. Recently, we demonstrated that a flavanone 2-hydroxylase (ZmF2H1, CYP93G5) converts flavanones to the corresponding 2-hydroxy derivatives, which are expected to serve as substrates for C-glycosylation. Here, we isolated a cDNA encoding a UDP-dependent glycosyltransferase (UGT708A6), and its activity was characterized by in vitro and in vivo bioconversion assays. In vitro assays using 2-hydroxyflavanones as substrates and in vivo activity assays in yeast co-expressing ZmF2H1 and UGT708A6 show the formation of the flavones C-glycosides. UGT708A6 can also O-glycosylate flavanones in bioconversion assays in Escherichia coli as well as by in vitro assays with the purified recombinant protein. Thus, UGT708A6 is a bifunctional glycosyltransferase that can produce both C- and O-glycosidated flavonoids, a property not previously described for any other glycosyltransferase.  相似文献
