Myosin light chain phosphatase with its regulatory subunit, myosin phosphatase target subunit 1 (MYPT1) modulates Ca2+-dependent phosphorylation of myosin light chain by myosin light chain kinase, which is essential for smooth muscle contraction. The role of MYPT1 in vascular smooth muscle was investigated in adult MYPT1 smooth muscle specific knock-out mice. MYPT1 deletion enhanced phosphorylation of myosin regulatory light chain and contractile force in isolated mesenteric arteries treated with KCl and various vascular agonists. The contractile responses of arteries from knock-out mice to norepinephrine were inhibited by Rho-associated kinase (ROCK) and protein kinase C inhibitors and were associated with inhibition of phosphorylation of the myosin light chain phosphatase inhibitor CPI-17. Additionally, stimulation of the NO/cGMP/protein kinase G (PKG) signaling pathway still resulted in relaxation of MYPT1-deficient mesenteric arteries, indicating phosphorylation of MYPT1 by PKG is not a major contributor to the relaxation response. Thus, MYPT1 enhances myosin light chain phosphatase activity sufficient for blood pressure maintenance. Rho-associated kinase phosphorylation of CPI-17 plays a significant role in enhancing vascular contractile responses, whereas phosphorylation of MYPT1 in the NO/cGMP/PKG signaling module is not necessary for relaxation.  相似文献

目的:了解西安市三甲综合医院耐甲氧西林金黄色葡萄球菌(MRSA)qacA/B基因的携带情况,以便掌握其对消毒剂的耐药情况。方法:收集临床分离的152株耐甲氧西林金黄色葡萄球菌,采用聚合酶链反应检测qacA/B基因,并将qacA/B基因扩增产物进行测序,测序结果进行比对。结果:152株耐甲氧西林金黄色葡萄球菌中有11株检出qacA/B阳性,qacA/B携带率7.2%;测序结果比对发现,4株qacA的基因编码序列同一位点均有一个碱基发生突变(C→T),苏氨酸突变为异亮氨酸。结论:该地区临床分离的耐甲氧西林金黄色葡萄球菌中qacA/B携带率相对较低,但也存在抵抗消毒剂的风险,临床应加强消毒剂使用的管理,阻止MRSA的传播,减少感染的发生。  相似文献

Ca2+在植物生长发育和环境适应过程中发挥着中心调控作用,钙信号是植物生长发育和逆境响应的主要调控因子,钙结合蛋白是植物钙信号传导途径的最重要组分之一,然而植物钙结合蛋白在体内和体外与Ca2+结合的技术体系还有待完善和发展。为了系统总结植物钙结合蛋白的鉴定方法与技术,本文从定性结合、定量结合和结合方式等角度,综述了植物钙结合蛋白在体内和体外条件下与Ca2+结合的原理、方法、特点和应用前景,详细阐述了近年来的主要检测方法,并对其今后的发展趋势作了展望。本文将为植物钙结合蛋白的分离、功能鉴定和作用机制的研究提供技术支撑。  相似文献

Small ubiquitin-like modifier (SUMO1–3) conjugation plays a critical role in embryogenesis. Embryos deficient in the SUMO-conjugating enzyme Ubc9 die at the early postimplantation stage. Sumo1−/− mice are viable, as SUMO2/3 can compensate for most SUMO1 functions. To uncover the role of SUMO2/3 in embryogenesis, we generated Sumo2- and Sumo3-null mutant mice. Here, we report that Sumo3−/− mice were viable, while Sumo2−/− embryos exhibited severe developmental delay and died at approximately embryonic day 10.5 (E10.5). We also provide evidence that SUMO2 is the predominantly expressed SUMO isoform. Furthermore, although Sumo2+/− and Sumo2+/−;Sumo3+/− mice lacked any overt phenotype, only 2 Sumo2+/−;Sumo3−/− mice were found at birth in 35 litters after crossing Sumo2+/−;Sumo3+/− with Sumo3−/− mice, and these rare mice were considerably smaller than littermates of the other genotypes. Thus, our findings suggest that expression levels and not functional differences between SUMO2 and SUMO3 are critical for normal embryogenesis.  相似文献

This study aims to compare the different latent infections of herpes simplex virus type I in a murine model. One hundred and twenty BALB/c mice were randomly assigned into either of three groups: intravenous inoculation group, ocular abrasion group, and intranasal inoculation group. Six weeks later, the trigeminal ganglia (TG) were removed to detect the expression of HSV-I antigen. HSV DNA in TG was also detected by polymerase chain reaction to confirm latent infection. The rate of HSV DNA in TG detected in the intravenous inoculation group was 18/22 and 22/26 in the ocular abrasion group, both of which were higher than the rate detected in the intranasal inoculation group (18/30). The expression of HSV antigen in TG in these three groups was all negative. Mortality rate in the intravenous inoculation group was 8/30, which was much higher than those of the two other groups. Intranasal virus dripping, cornea abrasion, and intravenous injection can detect latent HSV-I infection in a murine model. Compared to two other groups, the cornea abrasion group showed less severe signs, a quicker recovery rate in acute infection, and higher incidence rate of latent infection. Therefore, it is an ideal method in the presence of latent HSV-I infection.  相似文献

A gene encoding the thermostable raw starch digesting α-amylase in Thermobifida fusca NTU22 was amplified by PCR, sequenced and cloned into Pichia pastoris X-33 host strain using the vector pGAPZαA, allowing constitutive expression and secretion of the protein. Recombinant expression resulted in high levels of extracellular amylase production, as high as 510 U/l in the Hinton flask culture broth. The purified amylase showed a single band at about 65 kDa by SDS-polyacrylamide gel electrophoresis after being treated with endo-β-N-acetylglycosaminidase H, and this agrees with the predicted size based on the nucleotide sequence. About 75% of the original activity remained after heat treatment at 60°C for 3 h. The optimal pH and temperature of the purified amylase were 7.0 and 60°C, respectively. The purified amylase exhibited a high level of activity with raw sago starch. After 48-h treatment, the DPw of raw sago starch obviously decreased from 830,945 to 378,732. The surface of starch granules was rough, and some granules displayed deep cavities.  相似文献

Regeneration of pulmonary epithelial cells plays an important role in the recovery of acute lung injury (ALI), which is defined by pulmonary epithelial cell death. However, the mechanism of the regenerative capacity of alveolar epithelial cells is unknown. Using a lung injury mouse model induced by hemorrhagic shock and lipopolysaccharide, a protein mass spectrometry‐based high‐throughput screening and linage tracing technology to mark alveolar epithelial type 2 cells (AEC2s), we analyzed the mechanism of alveolar epithelial cells proliferation. We demonstrated that the expression of Hippo‐yes‐associated protein 1 (YAP1) key proteins were highly consistent with the regularity of the proliferation of alveolar epithelial type 2 cells after ALI. Furthermore, the results showed that YAP1+ cells in lung tissue after ALI were mainly Sftpc lineage‐labeled AEC2s. An in vitro proliferation assay of AEC2s demonstrated that AEC2 proliferation was significantly inhibited by both YAP1 small interfering RNA and Hippo inhibitor. These findings revealed that YAP functioned as a key regulator to promote AEC2s proliferation, with the Hippo signaling pathway playing a pivotal role in this process.  相似文献
