Background

The diagnosis of human African trypanosomiasis (HAT) caused by Trypanosoma brucei gambiense relies mainly on the Card Agglutination Test for Trypanosomiasis (CATT). There is no immunodiagnostic for HAT caused by T. b. rhodesiense. Our principle aim was to develop a prototype lateral flow test that might be an improvement on CATT.

Methodology/Principle Findings

Pools of infection and control sera were screened against four different soluble form variant surface glycoproteins (sVSGs) by ELISA and one, sVSG117, showed particularly strong immunoreactivity to pooled infection sera. Using individual sera, sVSG117 was shown to be able to discriminate between T. b. gambiense infection and control sera by both ELISA and lateral flow test. The sVSG117 antigen was subsequently used with a previously described recombinant diagnostic antigen, rISG65, to create a dual-antigen lateral flow test prototype. The latter was used blind in a virtual field trial of 431 randomized infection and control sera from the WHO HAT Specimen Biobank.

Conclusion/Significance

In the virtual field trial, using two positive antigen bands as the criterion for infection, the sVSG117 and rISG65 dual-antigen lateral flow test prototype showed a sensitivity of 97.3% (95% CI: 93.3 to 99.2) and a specificity of 83.3% (95% CI: 76.4 to 88.9) for the detection of T. b. gambiense infections. The device was not as good for detecting T. b. rhodesiense infections using two positive antigen bands as the criterion for infection, with a sensitivity of 58.9% (95% CI: 44.9 to 71.9) and specificity of 97.3% (95% CI: 90.7 to 99.7). However, using one or both positive antigen band(s) as the criterion for T. b. rhodesiense infection improved the sensitivity to 83.9% (95% CI: 71.7 to 92.4) with a specificity of 85.3% (95% CI: 75.3 to 92.4). These results encourage further development of the dual-antigen device for clinical use.  相似文献

The males of many fig wasps fight fatally for mating opportunities. The concentration of females in space has been proposed as one factor selecting for their aggressive behaviour. We studied the temporal distribution of receptive females to obtain a clearer impression of the operational sex ratio in figs. Females of nonpollinating species emerged from figs over a protracted period of time and this resulted in an extremely male-biased operational sex ratio, conducive to extreme fighting. Since there were so few receptive females at any one time, a male could defend an eclosing female. Consequently, the largest, Otitesella longicauda male in a fig had a much higher mating success than smaller males. This suggests that larger males have a larger fitness advantage than larger females and a Trivers-Willard effect could have important implications for sex allocation. Apterous and seemingly nondispersing males routinely left their figs. Such dispersal can affect both (1) sex allocation by reducing the degree of local mate competition between brothers and (2) male dimorphism by reducing the mating opportunities of males with a dispersing morphology. We show that the wingless digitata males of the Otitesella digitata species group disperse on to leaves close to their natal fig. An extremely male-biased sex ratio resulted in almost all O. longicauda females being mated. These findings suggest that the classical concept of the fig wasp mating system is too simplistic and that important assumptions of sex allocation models are violated. We estimated selection on adult body size for two generations in two populations of Aquarius remigis, as part of a long‐term study of the adaptive significance of sexual size dimorphism (SSD). Net adult fitness was estimated from the following components: prereproductive survival, daily reproductive success (mating frequency or fecundity), and reproductive lifespan. Standardized selection gradients were estimated for total length and for thorax, abdomen, genital and mesofemur lengths. Although selection was generally weak and showed significant temporal and spatial heterogeneity, patterns were consistent with SSD. Prereproductive survival was strongly influenced by date of eclosion, but size (thorax and genital lengths in females; total and abdomen lengths in males) played a significant secondary role. Sexual selection favoured smaller males with longer external genitalia in one population. Net adult fitness was not significantly related to body size in females, but was negatively related to size (thorax and total length) in males.  相似文献

Summary Free radicals have an important role in the metabolism and development of aerobic organisms; however, their uncontrolled production leads to oxidative stress. This paper explores the possibility that free radical mediated stress has a role in tissue culture recalcitrance. In the context of this paper, recalcitrance is considered to be the inabilit of plant tissue cultures to respond to culture manipulations; in its broadest terms, this study also concerns the time-related decline (i.e. in vitro aging) and loss of morphogenetic competence and totipotent capacity. Studies on a diverse range of in vitro plant systems have shown that tissue cultures produce free radicals, lipid peroxides and toxic, aldehydic lipid peroxidation products. Levels of these compounds vary in response to different tissue culture manipulations, but their production is enhanced during dedifferentation and antioxidant profiles also vary throughout different phases of culture. A hypothesis is presented which suggests that tissue culture manipulations cause major metabolic and developmental changes, some of which may predispose in vitro cultures to increased free radical formation. If antioxidant protection is compromised, oxidative stress ensues and free radicals and their reaction products react with macromolecules such as DNA, proteins and enzymes, causing cellular dysfuction and as a result, the cultures become recalcitrant.  相似文献

The spatio-temporal distribution of Ceutorhynchus assimilis Payk. (Coleoptera: Curculionidae) adults caught in a rectangular grid of flight traps in a crop of winter oilseed rape (Brassica napus L.) was mapped and was analysed using Spatial Analysis by Distance IndicEs (SADIE). Their distribution was compared to that of their larvae and that of their parasitoid Trichomalus perfectus (Walker) (Hymenoptera: Pteromalidae) in pods. The distribution of immigrating C. assimilis adults was consistent with their arrival at the crop boundaries and movement within the crop towards its centre. Adult C. assimilis were aggregated at all times, invasion being on two fronts, leading to the formation of two major clusters within the crop. Large areas of the crop remained relatively unpopulated. During the emigration phase, numbers declined simultaneously in all parts of the crop. The distributions of adult and larval C. assimilis and of larval T. perfectus were spatially associated. The distribution of the parasitoid did not show a density dependent relationship with that of its host. We discuss the movements of insects which underlie their population distributions, the value of integrating spatial information into improved management strategies for C. assimilis and the potential for the spatial targeting of insecticides to reduce the amount applied and to conserve T. perfectus.  相似文献

Life-history theory allows predictions of how changes in environmental selection pressures along a species' geographic distribution result in discrete shifts in life-history traits. We tested for spatial patterns of 24 populations of brown bears Ursus arctos across North America that grouped according to the following environmental and population parameters: evapotranspiration as a correlate of primary productivity of vegetation, coefficient of variation of monthly evapotranspiration values as a measure of seasonality. population density, and adult female weight. Cluster analysis grouped brown bear populations into two regions: Pacific-coastal populations characterized by high population density and large females that lived in areas of high primary productivity and low seasonality. and inland and barren-ground populations characterized by relatively low density and small bears that lived in areas of low productivity and high seasonality. For each region, we tested whether life-history traits (age at maturity and interbirth interval) related to primary productivity or seasonality. High altitude (interior: > 1000 m) and high latitude (barren-ground; >65°N) populations respond to extremes in seasonality with risk-spreading adaptations. For example, age at maturity and interbirth interval increased with greater seasonality. In contrast, Pacific-coastal populations living on the western edge of brown bear geographic range respond to intraspecific competition at high densities by maximizing offspring competitive ability. For example, age at maturity increased with greater primary productivity and high population density. In each region, the female parent decided on the life-history trade-offs required to reduce the risks of offspring mortality depending on the environmental pattern.  相似文献

A fecal analysis survey was undertaken to quantify animal inputs of pathogenic and indicator microorganisms in the temperate watersheds of Sydney, Australia. The feces from a range of domestic animals and wildlife were analyzed for the indicator bacteria fecal coliforms and Clostridium perfringens spores, the pathogenic protozoa Cryptosporidium and Giardia, and the enteric viruses adenovirus, enterovirus, and reovirus. Pathogen and fecal indicator concentrations were generally higher in domestic animal feces than in wildlife feces. Future studies to quantify potential pathogen risks in drinking-water watersheds should thus focus on quantifying pathogen loads from domestic animals and livestock rather than wildlife.  相似文献

Exonuclease I was originally identified as a 5' --> 3' deoxyribonuclease present in fractionated extracts of Schizosaccharomyces pombe and Saccharomyces cerevisiae. Genetic analysis of exo1 mutants of both yeasts revealed no major defect in meiosis, suggesting that exonuclease I is unlikely to be the primary activity that processes meiosis-specific double-strand breaks (DSBs). We report here that exo1 mutants of S. cerevisiae exhibit subtle but complex defects in meiosis. Diploids containing a homozygous deletion of EXO1 show decreased spore viability associated with an increase in meiosis I nondisjunction, while intergenic recombination is reduced about twofold. Exo1p functions in the same pathway as Msh5p for intergenic recombination. The length of heteroduplex tracts within the HIS4 gene is unaffected by the exo1 mutation. These results suggest that Exo1p is unlikely to play a major role in processing DSBs to form single-stranded tails at HIS4, but instead appears to promote crossing over to ensure disjunction of homologous chromosomes. In addition, our data indicate that exonuclease I may have a minor role in the correction of large DNA mismatches that occur in heteroduplex DNA during meiotic recombination at the HIS4 locus.  相似文献

Oxidative damage is associated with Alzheimer's disease and mild cognitive impairment, but its relationship to the development of neuropathological lesions involving accumulation of amyloid-beta (Abeta) peptides and hyperphosphorylated tau protein remains poorly understood. We show that inducing oxidative stress in primary chick brain neurons by exposure to sublethal doses of H(2)O(2 )increases levels of total secreted endogenous Abeta by 2.4-fold after 20 h. This occurs in the absence of changes to intracellular amyloid precursor protein or tau protein levels, while heat-shock protein 90 is elevated 2.5-fold. These results are consistent with the hypothesis that aging-associated oxidative stress contributes to increasing Abeta generation and up-regulation of molecular chaperones in Alzheimer's disease.  相似文献
