The pro-inflammatory cytokine macrophage migration inhibitory factor (MIF) is induced by glucocorticoids (GCs), but it was not previously known if MIF regulates cellular sensitivity to GC. Here we show in GC and LPS-treated peritoneal macrophages derived from MIF-/- and wt mice that the absence of endogenous MIF is associated with increased sensitivity to GC of TNF release. This is associated with increased expression of mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1), concomitant decreased phosphorylation of p38 MAPK, but no effect of MIF on nuclear factor kappaB (NF-kappaB). These results demonstrate that MIF regulates GC sensitivity by phosphorylation of p38, and provides a cellular mechanism for this observation, indicating that MKP-1 is a central target of this regulation. 相似文献

In West Africa, lineage splitting between the M and S molecular forms of the major Afro-tropical malaria mosquito, Anopheles gambiae (Diptera: Culicidae), is thought to be driven by ecological divergence, occurring mainly at the larval stage. Here, we present evidence for habitat segregation between the two molecular forms in and around irrigated rice fields located within the humid savannahs of western Burkina Faso. Longitudinal sampling of adult mosquitoes emerging from a range of breeding sites distributed along a transect extending from the heart of the rice field area into the surrounding savannah was conducted from June to November 2009. Analysis revealed that the two molecular forms and their sibling species Anopheles arabiensis are not randomly distributed in the area. A major ecological gradient was extracted in relation to the perimeter of the rice fields. The M form was associated with larger breeding sites mostly consisting of rice paddies, whereas the S form and An. arabiensis were found to depend upon temporary, rain-filled breeding sites. These results support hypotheses about larval habitat segregation and confirm the suggestion that the forms have different larval habitat requirements. A morphological phylogenetic hypothesis of the relationships among the species of Africana is provided. The phylogenetic analysis suggests that Squamata or Amphibia were ancestral hosts for species of Africana, with dispersal events to Testudinidae; a second transfer to amphibians is also suggested. On the basis of biological data for these parasites and the paleogeographic reconstructions, a South American origin for Africana spp. is indicated, with subsequent dispersal events to Africa. Aim To investigate the phylogeographical patterns of two poorly dispersing but widely distributed monogenean species, Haliotrema aurigae and Euryhaliotrematoides grandis, gill parasites of coral reef fishes from the family Chaetodontidae. Location South Pacific Ocean (SPO). Methods Sequence data from the mitochondrial cytochrome oxidase subunit I (COI) gene were obtained from samples from five localities of the SPO (Heron Island, Lizard Island, Moorea, Palau and Wallis) for the two parasite species. Phylogenetic and genetic diversity analyses were used to reconstruct phylogeographical patterns, and dates of cladogenetic events were estimated. Results Overall, 50 individuals of 17 Haliotrema aurigae and 33 of Euryhaliotrematoides grandis were sequenced from five localities of the SPO for COI mtDNA (798 bp). Our results revealed a deep phylogeographical structure in the species Euryhaliotrematoides grandis. The molecular divergence between individuals from Moorea and individuals from the remaining localities (7.7%) may be related to Pleistocene sea‐level fluctuations. In contrast, Haliotrema aurigae shows no phylogeographical patterns with the presence of most of the mitochondrial haplotypes in every locality sampled. Main conclusions Our study shows contrasting phylogeographical patterns of the two monogenean parasite species studied, despite many shared characteristics. Both parasites are found on the same host family, share the same geographical range and ecology, and are phylogenetically close. We propose two hypotheses that may help explain the diparity: the hypotheses involve differences in the evolutionary age of the parasite species and their dispersal capabilities. Additionally, the lack of phylogeographical structure in Haliotrema aurigae contrasts with its apparently restricted dispersion, which is likely to occur during the egg stage of the life cycle, inducing a passive dispersal mechanism in butterflyfish monogeneans. 相似文献

Patterns of species richness, prevalence and abundance of ectoparasites have rarely been investigated at both the levels of populations and species of hosts. Here, we investigated the effects in changes in small mammal density on species richness, abundance and prevalence of ectoparasitic fleas. The comparative analyses were conducted for different small mammal species and among several populations during a long-term survey. We tested the hypothesis that an increase in host density should be linked with an increase in parasite species richness both among host species and among populations within host species, as predicted by epidemiological models. We also used host species density data from literature. We found that host density has a major influence on the species richness of ectoparasite communities of small mammals among host populations. We found no relationship between data of host density from the literature and parasite species richness. In contrast with epidemiological hypotheses, we found no relationships between abundance, or prevalence, and host density, either among host species or among host populations. Moreover, a decrease in abundance of fleas in relation with an increase in host density was observed for two mammal species (Apodemus agrarius and A. flavicollis). The decrease or the lack of increase in flea abundance in relation with an increase in host density suggests anti-parasitic behavioural activities such as grooming. 相似文献

Abstract. The reasons why some species are resistant to extinction or are better invaders of islands than others remain unexplained. In this study, we test the hypothesis that mammals living on the mainland at higher density than predicted by the density/body mass relationship have a much greater chance to colonize a small island successfully, and/or that they are less likely to become extinct when living on small islands. For this, we used data compiled on mammals from a number of Mediterranean islands. We show a nested pattern for mammals on western Mediterranean islands, which suggests that the distribution of mammals on these islands is not the result of a random process. Using two comparative methods, we show that mammal density on the mainland, corrected for body mass, is negatively correlated with island size. Mammals with a high density compared to the density/mass relationship are the best invaders and/or probably have less chance of going extinct on small islands when population size is small. 相似文献

We investigate the patterns of parasite species richness of small mammals of the Western Palaearctic, and the effects of insularity. We compiled 28 studies dealing with 16 species of mammals. There was a significant effect of area size on the total parasite species richness, controlling for host sample size. Four mammal species were sufficiently sampled. Two are associated with humans: Mus musculus, and Rattus rattus , and two are not associated with humans, Apodemus sylvaticus and Clethrionomys glaerolus. We show that there is an effect of insularity on parasite species richness of A. sylvaticus . Rodents associated with humans show no effect of insularity on parasite species richness. These rodents appear to reflect a more complex pattern of parasite species richness, probably because of their close association with humans. Host specificity of parasites significantly decreases on islands, with the exception of Rattus rattus . We use parasitological data in order to explore the geographical connections between host populations. We show that parasites may help to resolve these connections but we emphasise that parasites are better geographic than phylogenetic markers. 相似文献
