Summary— Malate synthase (MS; EC 4.1.3.2), an enzyme specific to the glyoxylate cycle, was studied in cotyledons of dark-grown soybean (Glycine max L) seedlings with light and electron microscopy techniques. Immunogold localization confirmed biochemical evidence that MS from soybean is a glyoxysomal matrix enzyme.  相似文献

Phycobiliprotein aggregates were isolated from the prokaryote Acaryochloris marina, containing chlorophyll d as major pigment. In the electron microscope the biliprotein aggregates appear as rod-shaped structures of 26.0×11.3 nm, composed of four ring-shaped subunits 5.8 nm thick and 11.7 nm in diameter. Spectral data indicate that the aggregates contain two types of biliproteins: phycocyanin and an allophycocyanin-type pigment, with very efficient energy transfer from the phycocyanin- to allophycocyanin-type constituent. The chromophore-binding polypeptides of the pigments have apparent molecular masses of 16.2 and 17.4 kDa. They crossreact with antibodies against phycocyanin and allophycocyanin from a red alga.  相似文献

Malate dehydrogenase isoenzymes from eight cyanophycean species were investigated with polyacrylamide disc gel electrophoresis. Using 7% acrylamide the pherograms from each species showed 5–8 zones with malate dehydrogenase activity. It was demonstrated that in Anabaena flos-aquae there are 8 isoenzyme bands which include 3 forms of equal molecular weight, two of which consist of several isomers differing in their net charge. The MDH zymograms of the blue-green algae investigated can be used as “fingerprints”. The isoenzyme pattern of the MDHs of Anacystis nidulans makes its position in the order Chroococcales uncertain.

Wäßrige Extrakte aus acht Cyanophyceenarten wurden einer Polyacrylamid-Discelektrophorese unterzogen und die erhaltenen Elektropherogramme auf Malat-Dehydrogenase (MDH)-Aktivität geprüft. Dabei ergab sich, daß unter unseren Versuchsbedingungen die MDH der getesteten Cyanophyceen in 5–8 Isoenzymbanden aufspaltet. Für Anabaena flos-aquae konnte gezeigt warden, daß sich die 8 Isoenzymbanden auf wenigstens 3 Molekulargewichts-Isomere zurückführen lassen, von denen zwei noch mehrere Ladungsisomere bilden. Die erhaltenen Zymogramme zeigen “fingerprint”-Charakter. was ihre mögliche Verwendbarkeit für die Chemotaxonomie der Cyanophyta nahelegt. Die Stellung von Anacystis nidulans innerhalb der CyanophyceenOrdnungen wird diskutiert.  相似文献

The elongation rate of wheat coleoptiles, treated with IAA andABA, was already affected during the first 8 hr of culture.The most sensitive zone of the material—for hormonal treatments—wasfirst localized and then comparatively cultured both in situand in vitro. A conspicuous accumulation of taste buds occurs in the rostral part of the plica sublingualis ("frenal lamella") of Alouatta and Aotus forming taste areas (area gustatoria) superficially situated in the oral mucous membrane. They are found in close vicinity to the orifices of the sublingual salivary glands, but are lacking in the aboral part of the plica sublingualis. They do not occur in all primate species studied. A taste area does not projects above the surface of the surrounding tissue like a papilla. The taste buds open not in crypts of furrows of the oral mucosa, but directly into the spatium sublinguale of the oral cavity proper. In the anterior part of the cavum oris proprium different kinds and very differentiated qualities of sensoral information are perceived (touch, olfaction, temperature). It is conceivable that the taste areas play an important role in perceiving fresh saliva, together with the other sensorial structures in this part of the mouth. This problem can be solved experimentally and by behavioral studies, In addition to its topographical relation to the tongue, the organon sublinguale of Callicebus is structurally very similar to the plica sublingualis of Aotus and Alouatta. Since a sublingua does not occur in New World monkeys, it can be concluded that this organ represents a plica sublingualis which became adherent to the undersurface of the tongue.  相似文献

Summary While T4D phages and both T4v and T4v1x mutants are equally sensitive to photodynamic action, T4x mutants are much more sensitive. It is proposed that T4x mutants undergo an abortive repair process involving action of the nuclease coded by the v gene.Lambda redB mutants deficient in protein are much more sensitive to photodynamic action than wildtype phages and redA mutants. On the other hand lambda redA mutants, which do not produce the phage-specific exonuclease, are much more sensitive to U.V. light than wildtype phages and redB mutants.These results were presented in an abstracted version at the V. International U.V.-Colloquium Grundlagen der UV-Wirkung, Kühlungsborn, DDR, in October 1969.  相似文献

Seasonal changes of the amylolytic activity in the gut content of roach and rudd in four Tyrolean lakes and the adaptation of the amylase to four acclimation temperatures and to different natural foods were studied. In roach amylolytic activity varied with the environmental temperature. Between September and October, however, enzyme activity shifted to a higher level where it remained for the entire cold season. In rudd, which has almost the same seasonal pattern of amylolytic activity as the roach, the activity of the enzyme is independent of environmental temperature. This suggests an endogenous rhythm of enzyme activity. Under natural conditions the roach, but not the rudd, has a higher amylolytic activity when feeding on animals and a lower activity when feeding on detritus. The laboratory experiments revealed no significant differences in amylase activity between fish feeding on animals and those feeding on plants, although in both species the enzymatic activity was lowest when Chara served as food.  相似文献

6-Hydroxydopamine (6-OH-DA) has been shown to produce degenerative changes in noradrenergic nerve terminals and preterminals in the CNS following intracisternal, intraventricular or direct injection into the brain parenchyma. Systemic injection of 6-OH-DA is known to result in degenerative changes in noradrenergic terminals in the peripheral nervous system. However, only a few studies have been carried out on the effects of systemic injections of 6-OH-DA on noradrenergic terminals in the CNS. In the present study cynomolgus and squirrel monkeys were injected intravenously on two successive days with total doses of 350 mg/kg and 150 mg/kg of 6-OH-DA, respectively, and sacrificed at 2 and 24 h following the second injection. Degenerative changes in the area postrema (AP) neurons in all injected animals were characterized by a generalized increase in electron density of cytoplasmic elements in axonal terminals and preterminals. Multilamellar bodies, clusters of clear and dense core vesicles, increased numbers of secondary lysosomes, and an increase in the number of glycogen increased markedly in injected animals, but no other glial alterations were observed. The number of mast cells in the AP was greater in injected than in noninjected animals, both in the perivascular spaces (PVS) and in parenchymal locations. Cell processes in the PVS were occasionally observed to contain electron dense bodies, and degenerative changes were seen in supraependymal processes in some injected animals.  相似文献

Cultures of the autotrophic bacteriumMethanobacterium thermoautotrophicum were shown to assimilate acetate when grown on CO2 and H2 in the presence of acetate. At 1 mM acetate 10% of the cell carbon came from acetate, the rest from CO2. At higher concentrations the percentage increased to reach a maximum of 65%at acetate concentrations higher than 20 mM. The data suggest that acetate may be an important carbon source under physiological conditions.The incorporation of acetate into alanine, aspartate and glutamate was studied in more detail. The cells were grown on CO2 and H2 in the presence of 1 mM U-14C-acetate. The three amino acids were isolated from the labelled cells by a simplified procedure. Alanine, aspartate and glutamate were found to have the same specific radioactivity. Degradation studies showed that C1 of alanine C1 and C4 of aspartate, and C1 and C5 of glutamate were exclusively derived from CO2, whereas C2 and C3 alamine and aspartate, and C3 and C4 of glutamate were partially derived from acetate. These findings and the presence of pyruvate synthase, phosphoenolpyruvate carboxylase and -ketoglutarate synthase inM. thermoautotrophicum indicate that CO2 is assimilated into the three amino acids via acetyl CoA carboxylation to pyruvate, phosphoenolpyruvate carboxylation to oxaloacetate, and succinyl CoA carboxylation to -ketoglutarate.  相似文献

Stoichiometry, kinetics, and optical properties of rabbit muscle pyruvate kinase activated with Co(II), Ni(II), Mg(II), and Mn(II) were studied. The stoichiometry of metal binding to enzyme was found to be 4 metal ions per tetrameric enzyme for Co(II) and Ni(II) by carrying out circular dichroic titrations. Cu(II) and Fe(II) were inactive. Ca(II) and Zn(II) were not activating, and were inhibitory with respect to all of the active cations. The temperature dependence of the optimal velocity is similar for all activating metals. The pH rate profiles suggest that there are two classes of enzyme activation by metal ions. Mg(II) and Mn(II) are quite similar to each other while Co(II) and Ni(II) are different from them but similar to each other. Absorption, natural, and magnetic CD in the visible region were used to probe the environment of the activating divalent cation in Ni(II)- and Co(II)-activated pyruvate kinase and their complexes with substrates and inhibitors...  相似文献
