Addition of calcium chloride to an egg homogenate of Strongylocentrotus purpuratus stimulates O2 consumption which is not inhibited by millimolar cyanide. Results strongly suggest that Ca2+-stimulated O2 consumption is at least partially the result of polyunsaturated fatty acid oxidation. First, addition of arachidonic acid (AA), or other polyunsaturated fatty acids, to the homogenate enhance Ca2+-stimulated O2 consumption; this enhancement, by AA, being coupled to its oxidation to a hydroxy fatty acid. Second, calcium stimulates a lipase activity in the homogenate that is capable of releasing free fatty acids. Third, Ca2+-stimulated O2 consumption and AA oxidation have virtually identical calcium requirements and pH optima. The sequence of events then is that upon calcium addition to the homogenate, lipase activity is increased which liberates free fatty acids. At the same time calcium also activates a polyunsaturated fatty acid oxygenase, possibly lipoxygenase, that converts the free fatty acids to hydroxy fatty acids. The possible physiological importance of this reaction is underscored by the high affinity for Ca2+ [approximately 10(-7)M], an ion known to increase above the required levels at fertilization. The pH activity profile also suggests possible physiological modulation because a pH change of 6.8 increasing to 7.2, as suggested to occur after fertilization, yields almost a twofold increase in O2 consumption. Egg homogenates from many other invertebrate species have the ability to oxidize AA in a Ca2+-dependent fashion. For the investigated species, the presence of Ca2+-stimulated O2 consumption and AA oxidation correlates with the presence of cyanide insensitive respiration in the intact egg. 相似文献

How are complex visual entities such as scenes represented in the human brain? More concretely, along what visual and semantic dimensions are scenes encoded in memory? One hypothesis is that global spatial properties provide a basis for categorizing the neural response patterns arising from scenes. In contrast, non-spatial properties, such as single objects, also account for variance in neural responses. The list of critical scene dimensions has continued to grow—sometimes in a contradictory manner—coming to encompass properties such as geometric layout, big/small, crowded/sparse, and three-dimensionality. We demonstrate that these dimensions may be better understood within the more general framework of associative properties. That is, across both the perceptual and semantic domains, features of scene representations are related to one another through learned associations. Critically, the components of such associations are consistent with the dimensions that are typically invoked to account for scene understanding and its neural bases. Using fMRI, we show that non-scene stimuli displaying novel associations across identities or locations recruit putatively scene-selective regions of the human brain (the parahippocampal/lingual region, the retrosplenial complex, and the transverse occipital sulcus/occipital place area). Moreover, we find that the voxel-wise neural patterns arising from these associations are significantly correlated with the neural patterns arising from everyday scenes providing critical evidence whether the same encoding principals underlie both types of processing. These neuroimaging results provide evidence for the hypothesis that the neural representation of scenes is better understood within the broader theoretical framework of associative processing. In addition, the results demonstrate a division of labor that arises across scene-selective regions when processing associations and scenes providing better understanding of the functional roles of each region within the cortical network that mediates scene processing. 相似文献

Objective
The level of T cell activation in untreated HIV disease is strongly and independently associated with risk of immunologic and clinical progression. The factors that influence the level of activation, however, are not fully defined. Since endogenous glucocorticoids are important in regulating inflammation, we sought to determine whether less optimal diurnal cortisol patterns are associated with greater T cell activation.Methods
We studied 128 HIV-infected adults who were not on treatment and had a CD4+ T cell count above 250 cells/µl. We assessed T cell activation by CD38 expression using flow cytometry, and diurnal cortisol was assessed with salivary measurements.Results
Lower waking cortisol levels correlated with greater T cell immune activation, measured by CD38 mean fluorescent intensity, on CD4+ T cells (r = −0.26, p = 0.006). Participants with lower waking cortisol also showed a trend toward greater activation on CD8+ T cells (r = −0.17, p = 0.08). A greater diurnal decline in cortisol, usually considered a healthy pattern, correlated with less CD4+ (r = 0.24, p = 0.018) and CD8+ (r = 0.24, p = 0.017) activation.Conclusions
These data suggest that the hypothalamic-pituitary-adrenal (HPA) axis contributes to the regulation of T cell activation in HIV. This may represent an important pathway through which psychological states and the HPA axis influence progression of HIV. 相似文献15.

Humoral immune responses elicited after secondary exposure to immunizing Ag are characterized by robust and elevated reactivity of memory B cells that exceed those of naive B cells during the primary response. The mechanism underlying this difference in responsiveness of naive vs memory B cells remains unclear. We have quantitated the response of naive and memory human B cells after in vitro stimulation with T cell-derived stimuli. In response to stimulation with CD40 ligand alone or with IL-10, both IgM-expressing and Ig isotype-switched memory B cells entered their first division 20-30 h earlier than did naive B cells. In contrast, the time spent traversing subsequent divisions was similar. Consistent with previous studies, only memory cells differentiated to CD38(+) blasts in a manner that increased with consecutive division number. These differentiated CD38(+) B cells divided faster than did CD38(-) memory B cell blasts. Proliferation of CD40 ligand-stimulated naive B cells as well as both CD38(+) and CD38(-) cells present in cultures of memory B cells was increased by IL-10. In contrast, IL-2 enhanced proliferation of CD38(-) and CD38(+) memory B cell blasts, but not naive cells. Thus, memory B cells possess an intrinsic advantage over naive B cells in both the time to initiate a response and in the division-based rate of effector cell development. Background
The World Health Organization, governments, and communities agree that community action is likely to reduce risky alcohol consumption and harm. Despite this agreement, there is little rigorous evidence that community action is effective: of the six randomised trials of community action published to date, all were US-based and focused on young people (rather than the whole community), and their outcomes were limited to self-report or alcohol purchase attempts. The objective of this study was to conduct the first non-US randomised controlled trial (RCT) of community action to quantify the effectiveness of this approach in reducing risky alcohol consumption and harms measured using both self-report and routinely collected data.Methods and Findings
We conducted a cluster RCT comprising 20 communities in Australia that had populations of 5,000–20,000, were at least 100 km from an urban centre (population ≥ 100,000), and were not involved in another community alcohol project. Communities were pair-matched, and one member of each pair was randomly allocated to the experimental group. Thirteen interventions were implemented in the experimental communities from 2005 to 2009: community engagement; general practitioner training in alcohol screening and brief intervention (SBI); feedback to key stakeholders; media campaign; workplace policies/practices training; school-based intervention; general practitioner feedback on their prescribing of alcohol medications; community pharmacy-based SBI; web-based SBI; Aboriginal Community Controlled Health Services support for SBI; Good Sports program for sports clubs; identifying and targeting high-risk weekends; and hospital emergency department–based SBI. Primary outcomes based on routinely collected data were alcohol-related crime, traffic crashes, and hospital inpatient admissions. Routinely collected data for the entire study period (2001–2009) were obtained in 2010. Secondary outcomes based on pre- and post-intervention surveys (n = 2,977 and 2,255, respectively) were the following: long-term risky drinking, short-term high-risk drinking, short-term risky drinking, weekly consumption, hazardous/harmful alcohol use, and experience of alcohol harm. At the 5% level of statistical significance, there was insufficient evidence to conclude that the interventions were effective in the experimental, relative to control, communities for alcohol-related crime, traffic crashes, and hospital inpatient admissions, and for rates of risky alcohol consumption and hazardous/harmful alcohol use. Although respondents in the experimental communities reported statistically significantly lower average weekly consumption (1.90 fewer standard drinks per week, 95% CI = −3.37 to −0.43, p = 0.01) and less alcohol-related verbal abuse (odds ratio = 0.58, 95% CI = 0.35 to 0.96, p = 0.04) post-intervention, the low survey response rates (40% and 24% for the pre- and post-intervention surveys, respectively) require conservative interpretation. The main limitations of this study are as follows: (1) that the study may have been under-powered to detect differences in routinely collected data outcomes as statistically significant, and (2) the low survey response rates.Conclusions
This RCT provides little evidence that community action significantly reduces risky alcohol consumption and alcohol-related harms, other than potential reductions in self-reported average weekly consumption and experience of alcohol-related verbal abuse. Complementary legislative action may be required to more effectively reduce alcohol harms.Trial registration

Cortices of unfertilized sea urchin eggs can be isolated in suspension and will discharge the attached cortical vesicles (CVs) in response to calcium. We describe a simple turbidometric assay for monitoring the Ca2+-induced discharge of these vesicles and also compare the discharge of vesicles isolated in a high salt medium (primarily KCl) with a medium more closely simulating the internal milieu of the cell (primarily potassium gluconate and glycine). Discharge in response to calcium is similar in both media, requiring approximately 6 μM calcium for one-half maximal discharge. There are, however, significant differences in morphology and protein composition of the two types of preparations (more proteins present in the glycine cortices) and also in the rate of discharge of the vesicles in response to calcium (KCl cortices with 6 sec as opposed to 30 sec in the glycine cortices). The glycine cortices gradually lose their ability to respond to calcium but retention of calcium sensitivity is considerably aided by inclusion of ATP in the media; ATP has no apparent effect on discharge of the KCl cortices. The glycine cortices, as opposed to the KCl cortices, exhibited variation in calcium sensitivity during the breeding season and in the number of vesicles which would not break down in response to added calcium (referred to as refractory vesicles). The question of which type of cortex preparation most closely simulates the in vivo situation is discussed, and the view is presented that the glycine cortices most closely resemble the in vivo situation. 相似文献

Biodiversity in southern Africa is globally extraordinary but threatened by human activities. Although there are considerable
biodiversity conservation initiatives within the region, no one has yet assessed the potential use of large carnivores in
such actions. Surrogate approaches have often been suggested as one such way of capitalizing on large carnivores. Here we
review the suitability of the large carnivore guild (i.e., brown hyaena Hyaena hyaena, spotted hyaena Crocuta crocutta, cheetah Acinonyx jubatus, leopard Panthera pardus, lion Panthea leo and African wild dog Lycaon pictus) to act as surrogate species for biodiversity conservation in southern Africa. We suggest that the guild must be complete
for the large carnivores to fully provide their role as ecological keystones. The potential for large carnivores to act as
umbrella and indicator species seems limited. However, self-sustaining populations of large carnivores may be useful indicators
of unfragmented landscapes. Moreover, diversity within the large carnivore guild may reflect overall biodiversity. Although
the global appeal of the large African carnivores makes them important international flagships, we stress that international
conservation funding must be linked to local communities for them to be important also locally. In summary, we suggest that
the flagship value of these large carnivores should be used to promote biodiversity conservation in the region, and that the
suggested relationship between large carnivore diversity and overall biodiversity is empirically tested. Finally we suggest
that direct conservation activities should focus on enhancing the keystone values of large carnivores through complete guild

Autism spectrum disorders (ASD) are a group of related neurodevelopmental syndromes with complex genetic etiology. We identified a de novo chromosome 7q inversion disrupting Autism susceptibility candidate 2 (AUTS2) and Contactin Associated Protein-Like 2 (CNTNAP2) in a child with cognitive and social delay. We focused our initial analysis on CNTNAP2 based on our demonstration of disruption of Contactin 4 (CNTN4) in a patient with ASD; the recent finding of rare homozygous mutations in CNTNAP2 leading to intractable seizures and autism; and in situ and biochemical analyses reported herein that confirm expression in relevant brain regions and demonstrate the presence of CNTNAP2 in the synaptic plasma membrane fraction of rat forebrain lysates. We comprehensively resequenced CNTNAP2 in 635 patients and 942 controls. Among patients, we identified a total of 27 nonsynonymous changes; 13 were rare and unique to patients and 8 of these were predicted to be deleterious by bioinformatic approaches and/or altered residues conserved across all species. One variant at a highly conserved position, I869T, was inherited by four affected children in three unrelated families, but was not found in 4010 control chromosomes (p = 0.014). Overall, this resequencing data demonstrated a modest nonsignificant increase in the burden of rare variants in cases versus controls. Nonetheless, when viewed in light of two independent studies published in this issue of AJHG showing a relationship between ASD and common CNTNAP2 alleles, the cytogenetic and mutation screening data suggest that rare variants may also contribute to the pathophysiology of ASD, but place limits on the magnitude of this contribution. 相似文献
