1. Osborne-Mendel (O-M) rats displayed differences in brain and systemic tryptophan metabolism. O-M rats had decreased brainstem tryptophan-5-hydroxylase activity and decreased serotonin (5-HT) levels as compared to Sprague-Dawley rats. However, brain tryptophan levels were actually increased in O-M rats. Norepinephrine, dopamine and 5-hydroxyindole-3-acetic acid levels were not different between strains. 2. Pineal serotonin levels were increased in O-M rats. 3. Liver tryptophan 2,3-dioxygenase activity was increased in O-M rats while tyrosine aminotransferase activity was not different between strains. 4. Total blood cholesterol was decreased in O-M rats while triglycerides, free fatty acids and albumin was not different between strains. Arterial networks in 50 pairs of human fetal hands, made visible by perfusion with radiopaque media, were compared between right and left hands. The imaged primary arterial networks in the fetal hands were also compared with those in adult human and rhesus monkey hands. It was found that superficial arch configurations and their primary ramifications are bilaterally similar in human fetal hands. The configurations of the primary arterial networks are established very early in prenatal growth and may be maintained into adulthood. The similarities in the arterial network arrangements between fetal human and rhesus monkey hands suggest that the rhesus monkey hand could provide an appropriate model for studies of surgical neurovascular anastomosis.  相似文献

The unambiguous identification of ovine chromsomes has become essential for the mapping of the sheep genome, which predominantly consists of telocentric chromosomes of gradually decreasing size. Nucleolus organizer regions (NORs) and Robertsonian fusions have been used here as the cytological and morphological markers, respectively, to define the banding pattern of eight paris of sheep telocentric chromosomes that have an ambiguous identification status. Five Robertsonian chromosomes involving most of the ambiguous chromosomes as well as normal prometaphase chromosomes were stained sequentially and separately by QFQ, GTG, and Ag-NOR methodologies. The prometaphase banding patterns of the ambiguous chromosomes 4, 6, 8, 9, 21, 24, 25 and 26 are represented schematically. For providing an accurate image of the banding pattern, a system of shading has been employed to show the relative intensity of bands in a given chromosome. The results presented here will facilitate the regional mapping of the sheep genome, extend the information on cytogenetic homology with other bovids, and substantially accelerate the comparative mapping studies in Bovidae.  相似文献

To investigate the role of DNA damage in the induction of homologous recombination in mammalian cells, a series of structurally related, polycyclic aromatic carcinogens, i.e., 1-nitrosopyrene (1-NOP), N-acetoxy-2-acetylaminofluorene (N-AcO-AAF), and 4-nitroquinoline 1-oxide (4-NQO), were compared for their ability to cause intrachromosomal homologous recombination between two herpes simplex virus thymidine kinase (Htk) genes stably integrated in the genome of a tk- mouse L cell strain 333 M. Each Htk gene contains an 8-bp XhoI linker inserted at a unique site so that expression of a functional Htk enzyme requires a productive recombinational event between the two nonfunctional genes. Each carcinogen caused a dose-dependent increase in the frequency of recombination. The results were compared to what had been found previously for a structurally related carcinogen, (+/-)-7 beta, 8 alpha-dihydroxy-9 alpha, 10 alpha-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene (BPDE). As a function of concentration, BPDE was the most active agent, followed by 4-NQO, and 1-NOP, and then N-AcO-AAF. When compared on the basis of equal cell killing, the most efficient carcinogen was 1-NOP, followed by N-AcO-AAF and BPDE, and then 4-NQO. Use of tritium-labeled compounds to determine the frequency of recombination as a function of the number of adducts initially bound to DNA showed that the most effective agent was BPDE, followed by 1-NOP and 4-NQO, and then N-AcO-AAF (ratio, 6.6:2.5:1.8:1.0). To determine if these differences in recombinagenic effectiveness reflected different rates of removal of the adducts from DNA, we measured the percentage of DNA adducts removed during the 24-h period post treatment and found that 1-NOP, 4-NQO and N-AcO-AAF residues were removed at approximately the same rate, i.e., 25%-30% off. Cellular analysis of a series of independent recombinants indicated that approximately 82% of the recombinational events induced by each agent were consistent with gene conversion. DNA-DNA hybridization analysis confirmed this, and showed that each recombinant tested contained an XhoI-resistant (wild-type) Htk gene; with the majority retaining the Htk gene duplication, consistent with nonreciprocal transfer of wild-type genetic information. In the rest, only a single copy of the Htk gene remained, reflecting a single reciprocal exchange within a chromatid or a single unequal exchange between sister chromatids.  相似文献

Six loci, apoliproprotein B (including Ag(x) antigen), immunoglobulin kappa constant region (IGKC), luteinizing hormone/choriogonadotrophin receptor, avian myelocytomatosis viral related oncogene, neuroblastoma derived, ornithine decarboxylase, and proopiomelanocortin (adrenocorticotropin/beta-lipotropin) (POMC), were newly assigned to sheep chromosome 3p using a chromosomally characterized minipanel of sheep-hamster cell hybrids. Isotopic in situ hybridization of IGKC to sheep chromosome 3p22–p17 is reported, confirming the cell hybrid assignment. As these loci are all known to map to human chromosome 2p, this study demonstrates that this chromosomal segment is extensively conserved in sheep. Only POMC has been previously assigned to cattle chromosome 11, which is the equivalent of sheep chromosome 3p. Therefore, we predict that the other loci assigned in this study to sheep 3p are likely to be located on cattle 11. The provisional assignment of an additional locus, annexin-like to sheep chromosome 3p is also reported.  相似文献
