GMP-PNP, a non-hydrolyzable analog of GTP binds tightly to G-protein in the presence of Mg2+, so that the binding is stable even after exhaustive washings. This property was exploited to prepare membrane samples of rat brain where G-protein GTP-binding sites were saturated with GMP-PNP. Experiments carried out with these membranes showed that GTP, GMP-PNP, GDP-S and GMP (1 mM) inhibit the sodium-independent [3H]glutamate binding by 30–40% [F(4,40) = 5.9; p < .001], whereas only GMP-PNP activates adenylate cyclase activity [F(6,42) = 3.56; p < .01]. The inhibition of sodium-independent [3H]glutamate binding occurred in the absence of Mg2+. These findings suggest that guanine nucleotides may inhibit glutamate binding and activate adenylate cyclase through distinct mechanisms by acting on different sites.  相似文献

When Trypanosoma gambiense was exposed in vitro to rat macrophages, the addition of homologous antiserum resulted in immediate adherence of the parasites to the surface of macrophages.The antigenic components of the parasite were fractionated by Sephadex G-200, DEAE-cellulose, and block electrophoresis. Each fraction was used for immunizing rats, and the antisera obtained were each treated for agglutinating antibodies and for their effect on in vitro phagocytosis by rat macrophages. Only the antiserum capable of agglutination could enhance phagocytosis. A strong possibility that the agglutination antigen is mostly responsible for phagocytosis, hence protection, is indicated.Scanning electron micrographs showed that adherence to the macrophage is mostly by the anterior part of the parasite.  相似文献

Pretracheal abscess due to endotracheal intubation has not been reported in literature. We present a case of a female patient who was admitted with acute hypercapnic respiratory failure. Patient was initially managed with noninvasive ventilation but eventually was intubated after sustaining a cardiac arrest. She could not be extubated because of poor weaning parameters, so a tracheostomy was planned. During surgery, a pretracheal abscess was found with destruction of the second, third, and fourth tracheal rings and intact posterior tracheal wall. The possible risk factors, mechanism of injury, and preventive strategy of tracheal complication of intubation are discussed.  相似文献

Mitochondria are important organelles not only as efficient ATP generators but also in controlling and regulating many cellular processes. Mitochondria are dynamic compartments that rearrange under stress response and changes in food availability or oxygen concentrations. The mitochondrial electron transport chain parallels these rearrangements to achieve an optimum performance and therefore requires a plastic organization within the inner mitochondrial membrane. This consists in a balanced distribution between free respiratory complexes and supercomplexes. The mechanisms by which the distribution and organization of supercomplexes can be adjusted to the needs of the cells are still poorly understood. The aim of this review is to focus on the functional role of the respiratory supercomplexes and its relevance in physiology. 1. We have analyzed the behavior of two types of asymmetric molecular forms (A forms) of acetylcholinesterase (AChE) during development of chick hindlimb muscle, in vivo and in cell culture, and upon irreversible inactivation of peroneal muscle AChE with diisopropylfluorophosphate (DFP) in vivo. 2. In agreement with previous developmental studies on chick muscle, globular forms of AChE (G forms) are predominant in chick hindlimb at early embryonic ages, being gradually replaced by A forms as hatching (and, therefore, onset of locomotion) approaches. Of the two A-form types, AI appears and accumulates significantly earlier than AII, so that A/G and II/I ratios higher than 1 are attained only at about hatching time. 3. Cultures prepared from 11-day chick embryo hindlimb myoblasts express both types of A forms, with a combined activity of 27% of total AChE after 12 days in culture. AI forms appear again earlier and are much more abundant than type II asymmetric species through the life span of cultures. 4. All AChE activity in the peroneal muscle is irreversibly inactivated by injection of DFP in vivo. The recovery of A forms follows the same sequence described for normal development, with a delayed and slower recovery of AII forms as compared with AI. 5. Several hypotheses involving tail polypeptides or tissue target molecules, or posttranslational interconversion, are proposed to help explain the earlier appearance and accumulation of AI forms in chick muscle.  相似文献

State‐of‐the‐art Cu(In,Ga)Se2 (CIGS) solar cells are grown with considerably substoichiometric Cu concentrations. The resulting defects, as well as potential improvements through increasing the Cu concentration, have been known in the field for many years. However, so far, cells with high Cu concentrations show decreased photovoltaic parameters. In this work, it is shown that RbF postdeposition treatment of CuInSe2 solar cells allows for capturing the benefits from the improved absorber quality with increasing Cu content. A reduced defect density and an increased doping level for cells with high Cu concentrations close to stoichiometry are demonstrated. Implementing a high mobility front transparent conductive oxide (TCO), the improved absorbers with 1.00 eV bandgap yield a solar cell efficiency of 19.2%, and combined with a perovskite top cell a 4‐terminal tandem efficiency of 25.0% are demonstrated, surpassing the record efficiency of both subcell technologies.  相似文献

In recent decades, seed banks have spread out worldwide as essential institutions for biodiversity preservation, like new Noah’s arks. However, little is known about the diversity of practices that are involved in them. The aim of this study is to reconstruct the dynamics of operation of the different seed banks, developing a typology of them worth providing. As sources for that aim, in-depth interviews to seed banks referents, documents and other materials related to seed banks have been used. First, we describe three stages which seed conservation has undergone until it became modern seed banks. The impact of the Convention on Biological Diversity and its debate context are considered. We analyze the knowledge involved in seed banks which turn them into more than just seeds reservoirs. Afterwards, we study how seed banks are used. From the functioning of seed banks and their objectives, we have identified three bank profiles: assistentialist, productivist and preservationist profiles. Finally, we analyze a series of cases that allow us to show the type of seed banks we have proposed. Policy implications are discussed.  相似文献

SUMMARY. As part of an extended series of experiments, various combinations of primaquine-pyrimethamine were tested against pigeons infected with Plasmodium relictum.
(a) Rapid clearance of blood forms was observed, although each component had been reduced to one-half of its previously established MED (minimal effective dose) for such clearance when administered alone.
(b) Primaquine-pyrimethamine was followed by unprecedented delays in relapse after drug withdrawal. Such delays had not been elicited in any previous experiments in the series, employing each component alone, or chloroquine alone, or primaquine-chloroquine combinations, or pamaquine-plasmocid-chloroquine (Premaline N).  相似文献
