The chronic oral administration of phenformin (20 mg/kg daily during 8 days) to the normal concious dog provokes a decrease in the fasting thiamine levels of plasma and erythrocytes. During intragastric loading tests with thiamine hydrochloride (30 mg/kg), carried out during the chronic treatment with the biguanide, the curve of plasma hyperthiaminemia is increased. Objective: Our goal was to test any association between human plasma circulating levels of monocyte chemoattractant protein‐1 (cMCP‐1) and insulin resistance and to compare monocyte chemoattractant protein‐1 (MCP‐1) adipose tissue gene expression and cMCP‐1 in relation with inflammatory markers. Research Methods and Procedures: cMCP‐1 was measured in n = 116 consecutive control male subjects to whom an insulin sensitivity (Si) test was performed. Circulating levels of soluble CD14, soluble tumor necrosis factor receptor type 2 (sTNFR2), soluble interleukin‐6 (sIL‐6), and adiponectin also were measured. Subcutaneous adipose tissue samples were obtained from n = 107 non‐diabetic and type 2 diabetic subjects with different degrees of obesity. Real‐time polymerase chain reaction was used to measure gene expression of MCP‐1, CD68, tumor necrosis factor‐α (TNF‐α), and its receptor TNFR2. Results: In the Si study, no independent effect of cMCP‐1 levels on insulin sensitivity was observed. In the expression study, in non‐diabetic subjects, MCP‐1 mRNA had a positive correlation with BMI (r = 0.407, p = 0.003), TNF‐α mRNA (r = 0.419, p = 0.002), and TNFR2 mRNA (r = 0.410, p = 0.003). In these subjects, cMCP‐1 was found to correlate with waist‐to‐hip ratio (r = 0.322, p = 0.048). In patients with type 2 diabetes, MCP‐1 mRNA was up‐regulated compared with non‐diabetic subjects. TNF‐α mRNA was found to independently contribute to MCP‐1 mRNA expression. In this group, CD68 mRNA was found to correlate with BMI (r = 0.455, p = 0.001). Discussion: cMCP‐1 is not associated with insulin sensitivity in apparently healthy men. TNF‐α is the inflammatory cytokine associated with MCP‐1 expression in subcutaneous adipose tissue. 相似文献

Field experiments and wild population monitoring have been performed to study the population biology of the rare long-lived Kosteletzkya pentacarpos (Malvaceae) in the Llobregat delta (Catalonia, NE Spain). Field experiments explored the fate of seeds in soil at different depths, seedling emergence, and seedling survival, growth and flowering with and without canopy cover during the first 2 years of life. They also were used to ascertain the size-related pattern of seedling survivorship and flowering. Field data concerning mortality, growth and fecundity of adult plants were collected yearly in three wild populations for 7–9 years. In old adults (reproducing long before the beginning of the study), ANOVAR tests were performed to compare maximum diameter, total and fertile shoots, and viable seeds per plant between years and populations. New adults (starting flowering the first year of study or in subsequent years) were used to explore, using linear and polynomial regressions, the association of RGR and both total and fertile shoot production with (i) plant size (maximum basal diameter or its logarithm); (ii) plant age (years in adult stage); and (iii) plant age after removing the effect of size and year-to-year fluctuations. In this case, we examined the age-related pattern of the residuals obtained from the regressions with size and year. The study identified the following main demographic features of K. pentacarpos: (i) transient, shallow soil seed bank; (ii) shade tolerance of seedling emergence; (iii) canopy-facilitation of seedling survival and bolting during the first two years of life; (iv) size-related pattern for seedling survivorship but not flowering; (v) exclusive dependence on a fluctuating seed output for reproduction; (vi) rapid adult growth; and (vii) high adult longevity but (viii) rapid depletion of fecundity with age. Seed output was highly constrained by mining insects. The changing size-structure and the decreasing reproductive success of old adults in several populations suggest that K. pentacarpos might undergo a dynamics of population establishment and extinction in the Ricarda marshes. Because of fluctuating reproduction and the lack of a persistent seed bank, the conservation of standing adult populations appears to be a key factor to ensure the persistence of the species. 相似文献

Interspecies transmission of prions is a well-established phenomenon, both experimentally and under field conditions. Upon passage through new hosts, prion strains have proven their capacity to change their properties and this is a source of strain diversity which needs to be considered when assessing the potential risks associated with consumption of prion contaminated protein sources. Rabbits were considered for decades to be a prion resistant species until proven otherwise recently. To determine the extent of rabbit susceptibility to prions and to assess the effects of passage of different prion strains through this species a transgenic mouse model overexpressing rabbit PrPC was developed (TgRab). Intracerebral challenges with prion strains originating from a variety of species including field isolates (ovine SSBP/1 scrapie, Nor98- scrapie; cattle BSE, BSE-L and cervid CWD), experimental murine strains (ME7 and RML) and experimentally obtained ruminant (sheepBSE) and rabbit (de novo NZW) strains were performed. On first passage TgRab were susceptible to the majority of prions (Cattle BSE, SheepBSE, BSE-L, de novo NZW, ME7 and RML) tested with the exception of SSBP/1 scrapie, CWD and Nor98 scrapie. Furthermore, TgRab were capable of propagating strain-specific features such as differences in incubation periods, histological brain lesions, abnormal prion (PrPd) deposition profiles and proteinase-K (PK) resistant western blotting band patterns. Our results confirm previous studies proving that rabbits are not resistant to prion infection and show for the first time that rabbits are susceptible to PrPd originating in a number of other species. Although retinoic acid (RA) has been implicated as one of the diffusible signals regulating forebrain development, patterning of the forebrain has not been analyzed in detail in knockout mouse mutants deficient in embryonic RA synthesis. We show that the retinaldehyde dehydrogenase 2 (RALDH2) enzyme is responsible for RA synthesis in the mouse craniofacial region and forebrain between the 8- and 15-somite stages. Raldh2-/- knockout embryos exhibit defective morphogenesis of various forebrain derivatives, including the ventral diencephalon, the optic and telencephalic vesicles. These defects are preceded by regionally decreased cell proliferation in the neuroepithelium, correlating with abnormally low D-cyclin gene expression. Increases in cell death also contribute to the morphological deficiencies at later stages. Molecular analyses reveal abnormally low levels of FGF signaling in the craniofacial region, and impaired sonic hedgehog signaling in the ventral diencephalon. Expression levels of several regulators of diencephalic, telencephalic and optic development therefore cannot be maintained. These results unveil crucial roles of RA during early mouse forebrain development, which may involve the regulation of the expansion of neural progenitor cells through a crosstalk with FGF and sonic hedgehog signaling pathways. 相似文献

Regulatory approvals for deliberate release of GM maize events into the environment have lead to real situations of coexistence between GM and non-GM, with some fields being cultivated with GM and conventional varieties in successive seasons. Given the common presence of volunteer plants in maize fields in temperate areas, we investigated the real impact of GM volunteers on the yield of 12 non-GM agricultural fields. Volunteer density varied from residual to around 10% of plants in the field and was largely reduced using certain cultural practices. Plant vigour was low, they rarely had cobs and produced pollen that cross-fertilized neighbour plants only at low—but variable—levels. In the worst-case scenario, the estimated content of GMO was 0.16%. The influence of GM volunteers was not enough to reach the 0.9% adventitious GM threshold but it could potentially contribute to adventitious GM levels, especially at high initial densities (i.e. above 1,000 volunteers/ha). 相似文献
