The neuropeptide galanin is widely expressed in the periphery and the central nervous system and mediates diverse physiological processes and behaviors including alcohol abuse, depression and anxiety. Four genes encoding galanin and its receptors have been identified (GAL, GALR1, GALR2 and GALR3). Recently we found that GAL haplotypes were associated with alcoholism, raising the possibility that genetic variation in GALR1, GALR2 and GALR3 might also alter alcoholism risk. Tag single nucleotide polymorphisms (SNPs) were identified by genotyping SNP panels in controls from five populations. For the association study with alcoholism, six GALR1, four GALR2 and four GALR3 SNPs were genotyped in a large cohort of Finnish alcoholics and non-alcoholics. GALR3 showed a significant association with alcoholism that was driven by one SNP (rs3,091,367). Moreover, the combination of the GALR3 rs3,091,367 risk allele and GAL risk haplotypes led to a modestly increased odds ratio (OR) for alcoholism (2.4) as compared with the effect of either GAL (1.9) or GALR3 alone (1.4). Likewise, the combination of the GALR3 and GAL risk diplotypes led to an increased OR for alcoholism (4.6) as compared with the effect of either GAL (2.0) or GALR3 alone (1.6). There was no effect of GALR1 or GALR2 on alcoholism risk. This evidence suggests that GALR3 mediates the alcoholism-related actions of galanin.  相似文献

The parameters investigated in this study with the objective of evaluating growth, lactation and reproductive performances, included birth weight, litter size, 0-90 days gain and average daily gain of kids as well as the milk yield and progesterone profile of Red Sokoto does supplemented with crop-residue based rations during the long-dry period of the subhumid zone in Nigeria. A total of 7 treatments of 4 goats each was utilised. All treatment groups had a basal diet of Digitaria smutsii hay and natural pasture ad libitum. Ration A supplemented with the conventional concentrate was used as the positive control; rations B and C were supplemented with crop residues; and ration D without supplement was used as the negative control. Supplementation with concentrate and crop residues significantly increased (P < 0.05) the birth weight and liveweight gains of kids, but littersize was unaffected. The heaviest kids at birth (1.3-1.4 kg) were from does in treatments 1A, 2A and 2C, while does in treatments 1B, 2B, 1C and D had the lightest kids (1.07-1.18 kg). The highest gains of 53.9 g x day(-1) were recorded in treatment 2A and the least (32.4 g x day(-1)) in treatment 1B. Supplementation also significantly influenced (P < 0.01) the daily milk yield of dams over the 90-day period of the dry season. All the does had similar progesterone profiles from late gestation through parturition to early lactation irrespective of their treatment group. It was concluded that ration C fed at the 2% level is a good and affordable supplementary feed package for increased birth weight and preweaning gains in kids for meat production.  相似文献

L(+)-lactic acid production was investigated using an enzymatic hydrolysate of waste office automation (OA) paper in a culture of the filamentous fungus Rhizopus oryzae. In 4 d culture, 82.8 g/l glucose, 7 g/l xylose, and 3.4 g/l cellobiose contained in the hydrolysate were consumed to produce 49.1 g/l of lactic acid. The lactic acid yield and production rate were only 0.59 g/g and 16.3 g/l/d, respectively, only 75% and 61% of the results from the glucose medium. The low production rate from waste OA hydrolysate was elucidated by trials using xylose as the sole carbon source; in those trials, the lactic acid production rate was 7.3 g/l/d, only 28% that of glucose or cellobiose. The low lactic acid yield from waste OA hydrolysate was clarified by trials using artificial hydrolysates comprised of 7:2:1 or 7:1:2 ratios of glucose:cellobiose:xylose. For both, the lactic acid production rate of 17.4 g/l/d matched that of waste OA paper, while the lactic acid yield was similar to that of the glucose medium. This indicates that the production rate may be inhibited by xylose derived from hemicellulose, and the yield may be inhibited by unknown compounds derived from paper pulp.  相似文献

Active beta1,3-N-acetylglucosaminyltransferase 2 (beta3GnT2) was produced in the baculovirus expression system (BES) and in stably transformed insect Tn-5B1-4 cells. beta3GnT2 was expressed as a secreted fusion protein with GFP(UV) with three different types of signal sequence to enhance the secretion of the fusion protein. In the stably transformed cells, the maximal beta3GnT2 activity differed between isolates, but their secretion efficiencies were similar. The difference between the maximal beta3GnT activities of the isolates studied was considered to be due to the presence of a copy number of the fusion gene, as determined on the basis of the results of Southern blot analysis. The beta3GnT activities of the culture supernatant in BES (Tn-5B1-4 cells) without or with the addition of the protease inhibitor, leupeptin, were 0.68 and 2.01 mU/ml, respectively. The stably transformed Tn-5B1-4 cells (Tn-pXme11) exhibited a beta3GnT activity of 6.83 mU/ml, which was 3.4-fold higher than that observed for BES with the leupeptin addition. The purity of fusion protein purified from the culture supernatant of the Tn-pXme11 was higher than 95% on SDS-PAGE, in contrast with that purified from the culture supernatant of the baculovirus-infected cells which contained low-molecular-weight fragments of the fusion protein. The stably transformed cell line is more suitable than BES for the efficient production of the secretory protein, beta3GnT2.  相似文献

Drug resistance in pathogenic trypanosomes threatens successful control of fatal sleeping sickness in man and hinders economic livestock production in sub-Saharan Africa. We report on the occurrence and development of drug resistance, and discuss the genetic basis of such resistance in Trypanosoma brucei. Understanding these mechanisms at the molecular level will enable improved management of existing drugs and provide valuable clues to the development of new trypanocides.  相似文献

Enzymatic hydrolysis of waste office paper was evaluated using three commercial cellulases, Acremonium cellulase, Meicelase, and Cellulosin T2. Varying the enzyme loading from 1 to 10% (w/w) conversion of waste office paper to reducing sugar was investigated. The conversion increased with the increase in the enzyme loading: in the case of enzyme loading of 10% (w/w), Acremonium cellulase yielded 79% conversion of waste office paper, which was 17% higher compared to Meicelase, 13% higher than that of Cellulosin T2. Waste activated bleaching earth (ABE) that contained 40% rapeseed oil and was discharged by an oil refinery plant, was used for riboflavin production in a culture of Ashbya gossypii. When 125 g/L waste ABE that contained 50 g/L rapeseed oil was added into the culture, the riboflavin concentration was 1.12 g/L, which was almost 1.6-fold as high as that of pure rapeseed oil. However, in waste ABE concentration higher than 125 g/L, the produced riboflavin concentration decreased, which was due to the difficulty in mixing due to the presence of a high amount of solid material in the culture. The surface of the waste ABE was smooth without a hitch, because of being covered with rapeseed oil. However, after the culture, the surface of the waste ABE seemed like that of new one, and the oil content was nearly zero grams per liter. The waste ABE, oily clay, and its black color gradually fade and yellow little by little, and finally the waste ABE changed to yellow powder. Of the riboflavin produced during the culture, 70% was adsorbed in the oil free waste ABE. With diluted alkali solution, extraction only two times yielded 90% recovery of riboflavin adsorbed in the waste ABE. The waste ABE containing waste vegetable oil was suitable for raw material for production of the value-added useful bioproducts, which might be a good model for reuse of the waste resource.  相似文献
