Divalent cations activate the lysophospholipase and transacylase reactions catalyzed by the same enzymes in the yeast Saccharomyces cerevisiae. The activation was observed at neutral pH, but not at the pH optimum of lysophospholipase/transacylase, near 3.5. Adenine nucleotides, especially AMP and ADP, are strong inhibitors of the same group of enzymes. Half maximal inhibition by AMP was found at a concentration of about 20 M. The inhibition by nucleotides in low concentrations is enhanced by divalent cations.  相似文献

Two DNA duplexes of identical sequence and 35 nt in length were synthesized by an original and a highly improved version of phosphoramidite chemistry. By base composition analysis, DNA synthesized by improved chemistry (termed DMTS-imp) contained no detectable modified bases while DNA synthesized by the original chemistry (termed DMTS-std) had a large number of modifications. Under optimal reaction conditions, HhaI and RsaI cleaved the DMTS-std duplex to 76-77% completion and the DMTS-imp duplex to 96-99% completion. Restriction analysis and piperidine treatment yielded estimates of approximately 3.0% modified nucleotides in DMTS-std and approximately 1.0% in DMTS-imp. Overall, the improvements in chemistry increased the restriction efficiency of synthetic DNA up to 10-fold.  相似文献

Interaction between a volatile anesthetic, methoxyflurane, and dipalmitoylphosphatidylcholine (DPPC) vesicle membrane was analyzed by nuclear Overhauser effect (NOE) difference spectroscopy and two-dimensional nuclear Overhauser spectroscopy (NOESY). The NOE difference spectra were obtained by selectively irradiating methoxy protons (hydrophobic end) of the anesthetic: a negative nuclear Overhauser effect of -2.94% was observed with the choline methyl protons of DPPC. The NOESY spectra revealed a cross-peak between the anesthetic methoxy protons and the choline methyl protons. A dipole-dipole interaction exists between the hydrophobic end of the anesthetic and the hydrophilic head group of DPPC. No other cross-peaks were observed. The anesthetic orients itself at the membrane/water interface by interacting with the hydrophilic surface of the DPPC membrane, leaving the hydrophilic end of the anesthetic molecule in the aqueous phase. The preferred residence site of dipolar volatile anesthetics is the membrane/water interface.  相似文献

Each of a range of pharmacological agents which function to increase intracellular levels of cAMP caused a morphological 'differentiation' of neuroblastoma x glioma hybrid, NG108-15, cells grown in tissue culture. Associated with this differentiation, increased incorporation of [32P]ADP-ribose catalysed by pertussis toxin was noted into a band of some 39-40 kDa in membranes derived from these cells. Immunoblotting using two antipeptide antisera which identify different regions of Go alpha demonstrated marked increases in the levels of this polypeptide in membranes of the differentiated cells. However, levels of the beta-subunit did not increase appreciably with differentiation.  相似文献

The crystal structure of thermitase from Thermoactinomyces vulgaris has been determined by x-ray diffraction at 2.2 A resolution. The structure was solved by a combination of single isomorphous replacement and molecular replacement methods. The structure was refined to a conventional R factor of 0.24 using restrained least square procedures CORELS and PROLSQ. The tertiary structure of thermitase is similar to that of subtilsin BPN'. The greatest differences between these structures are related to the insertions and deletions in the sequence.  相似文献

Infusion of phenobarbital and CCl4 was found to induce comuton control of mitochondrial respiration in a liver of starved rats. Comuton regulation of liver mitochondria respiration can be activated either by increase in liver activity or by damage caused by CCl4. Identification of the truncated peptides by plasma desorption mass spectrometry in C-terminal sequence determination with carboxypeptidases offers several advantages over analysis of the liberated amino acids. It is possible to perform in situ digestion of a nitrocellulose-bound sample already used for molecular weight determination and thus obtain sequence information without further sample consumption. In time-course analysis the analytical information, although not obtained in real time, is sufficiently rapid to adjust the digestion conditions. There is no need for quantitation because the identification is based on molecular weight differences. Sensitivity in the low picomole range is obtainable. The digestion of a number of peptides (900-3500 Da) with carboxypeptidase Y and MII has been monitored. It was found that successive use of the enzymes or use of a mixture of the enzymes was often advantageous. The sequence of up to 10 residues from the C-terminus has been determined for the peptides studied.  相似文献

Structural-functional organization of the lymphoid organs and functional state of the adrenals have been studied in animals, subjected to cold in early postnatal period, as well as changes of the parameters mentioned to a short and prolonged cooling in mature rats. For the animals increase in the thymus mass and in reproduction rate of lymphocytes in the thymus, spleen and lymph nodes is specific against the background of high corticosteroid secretion. When the control animals are kept in cold for a long time, after the phase of an acute stress, accompanied with hypercorticism and a pronounced lymphatic effect, during the period of an increased cold stability, the high secretion of glucocorticoid hormones is accompanied with a certain activation of thymus-dependent zones in the peripheral lymphoid organs. In the mature rats, subjected to cold at early ontogenesis both stress-reaction to cooling and rearrangement in the regulatory systems studied does not develop at adaptation to cold.  相似文献

An interaction of rabbit muscle D-glyceraldehyde-3-phosphate dehydrogenase and 3-phosphoglycerate kinase labeled with FITC was studied by following the changes in fluorescence intensity of the bound dye. The association between the two enzymes was found to be a rather slow process characterized by a second order rate constant of 1.1 +/- 0.2.10(3) M-1 s-1, the KD of the complex between apoenzymes being 3.2.10(-7) M. The stability of the complex increased upon increase of temperature and ionic strength of the medium, suggesting a hydrophobic character of association. The ligands which bind at the active centers of the two enzymes (NAD+, ATP, 3-phosphoglycerate) weakened the bienzyme association. Unlabeled 3-phosphoglycerate kinase was unable to displace the FITC-labeled enzyme from the complex. Taken together, the results indicate that interaction between D-glyceraldehyde-3-phosphate dehydrogenase and 3-phosphoglycerate kinase labeled by FITC is assisted by the dye, which may bind at nucleotide-binding sites of GPDH. No interaction was observed between the FITC-labeled 3-phosphoglycerate kinase and lactate dehydrogenase, which suggests that protein-protein interaction at specific "recognition" sites may be a prerequisite for the complex formation.  相似文献

To explore the secondary structures of lingual and pancreatic lipases, circular dichroism measurements were performed. Maximum average ellipticities were used to calculate the percentage of alpha-helices, beta-sheets, and random coils. Lingual lipase had an ellipticity of -20235 +/- 140 deg cm2/dmol (mean +/- SE) at 220 nm suggesting 60% alpha-helix, 20% beta-sheet and 20% random coil structure, but the mean ellipticity for pancreatic lipase was -14093 +/- 82 deg cm2/dmol (mean +/- SE) at 210 nm suggesting a 34.8% alpha-helical, 25% beta-sheet and 40% random coil secondary structure. An alpha-helical stretch of residues with a large hydrophobic moment ("globular" alpha-helix by hydrophobic moment plot) from amino acids 382 through 389 at the COOH-terminal end of lingual lipase was noted. This sequence, absent in pancreatic lipase, may account for the avid binding of lingual lipase to fat emulsion particles.  相似文献
