Neural precursor cell expressed, developmentally down-regulated gene 8 (NEDD8) is a recently discovered ubiquitin-like posttranslational modifier. NEDD8 acts predominantly as a regulator of ubiquitin-protein ligases and as a decoy for proteins targeted for proteasomal degradation. It thereby controls key events in cell cycle progression and embryogenesis. Deneddylase-1 (DEN1/NEDP1/SENP8) features a selective peptidase activity converting the proNEDD8 precursor to its mature form and an isopeptidase activity deconjugating NEDD8 from substrates such as cullins and p53. In this study, we describe a high-throughput screening (HTS)-compatible time-resolved fluorescent resonance energy transfer (TR-FRET) assay measuring the peptidase activity of DEN1.  相似文献

A swampy nest site of the sweat bee Augochloropsis caerulans (Vachal, 1903) in southern Brazil is reported. The bees colonized small earth mounds scattered throughout a water-covered area. Although the substrate of these mounds has high organic matter content, the risk of microbial infestation seems not to be enhanced owing to permanent water saturation. The nests are shallow and can be found 3 cm above the water table. Each nest consists of a cluster of vertically oriented cells on pillars in an oval shaped cavity. The specific nest architecture is thought to mitigate microbial infestation of the brood and to prevent water excess. Nests of A. caerulans are not restricted to water-saturated substrates, but the specific nest architecture presumably enables the bee to propagate in such habitats.  相似文献

Trinidad and Tobago has a long history of producing high-quality cacao (Theobroma cacao L.). Cacao genotypes in Trinidad and Tobago are of a highly distinctive kind, the so-called “Trinitario” cultivar group, widely considered to be of elite quality. The origin of Trinitario cacao is unclear, although it is generally considered to be of hybrid origin. We used massive parallel sequencing to identify polymorphic plastidic single nucleotide polymorphisms (cpSNPs) and polymorphic plastidic simple sequence repeats (cpSSRs) in order to determine the origin of the Trinitario cultivar group by comparing patterns of polymorphism to a reference set of ten completely sequenced chloroplast genomes (nine T. cacao and one outgroup, T. grandiflorum (Willd. ex Spreng.) Schum). Only three cpSNP haplotypes were present in the Trinitario cultivars sampled, each highly distinctive and corresponding to reference genotypes for the Criollo (CRI), Upper Amazon Forastero (UAF) and Lower Amazon Forastero (LAF) varietal groups. These three cpSNP haplotypes likely represent the founding lineages of cacao to Trinidad and Tobago. The cpSSRs were more variable with eight haplotypes, but these clustered into three groups corresponding to the three cpSNP haplotypes. The most common haplotype found in farms of Trinidad and Tobago was LAF, followed by UAF and then CRI. We conclude that the Trinitario cultivar group is of complex hybrid origin and has derived from at least three original introduction events.  相似文献

High levels of female and male sterility were observed among the hybrids from one of the two reciprocal crosses between a wild strain of D. melanogaster known as pi2 and laboratory strains. The sterility, which is part of a common syndrome called hybrid dysgenesis, was found to be associated with the rudimentary condition of one or both of the ovaries or testes. All other tissues, including those of the reproductive system were normal, as were longevity and mating behavior. The morphological details of the sterility closely mimic the agametic condition occurring when germ cells are destroyed by irradiation or by the maternal-effect mutation, grandchildless. We suggest that sterility in hybrid dysgenesis is also caused by failure in the early development of germ cells. There is a thermo-sensitive period beginning at approximately the time of initiation of mitosis among primordial germ cells a few hours before the egg hatches and ending during the early larval stages. Our results suggest that hybrid dysgenesis, which also includes male recombination, mutation and other traits, may be limited to the germ line, and that each of the primordial germ cells develops, or fails to develop, independently of the others. This hypothesis is consistent with the observed frequencies of unilateral and bilateral sterility, with the shape of the thermosensitivity curves and with the fact that males are less often sterile than females. The features of this intraspecific hybrid sterility are found to resemble those seen in some interspecific Drosophila hybrids, especially those from the cross D. melanogaster X D. simulans.  相似文献

A model of the C5a receptor was built based on the assumption that the seven membrane-spanning helices of known inward/outward direction are in an arrangement roughly similar to that in bacteriorhodopsin. Guidelines for the positioning of the helices were cysteine pairing, 'ridges into grooves' interdigitation of side chains and aromatic cluster formation. The chain segments protruding from the membrane are too short for folding into an independent ectodomain. The only longer segment (179-202) is tied down in its centre onto the membrane by a disulphide bridge and, thereby, made into two short loops as well. Ideas of the interaction of the C5a receptor with its ligand were derived mainly from the search for accommodation of the functionally essential arginine residues 40 and 74 of C5a. Asp82 is the only charged residue in a pocket approximately 20 A below the receptor surface and is conserved in the rhodopsin superfamily. It commends itself for binding Arg74 which is the tip of the flexible C-terminal chain of C5a, and rules out Arg40 in the structurally well-defined part of the molecule. The latter may bind to Glu180 at the bottom of a more shallow pocket which happens to resemble the substrate-binding site of trypsin.  相似文献
