A river classification framework is needed to make good management and planning decisions about river health and biodiversity. We developed a multi-attribute ecological river typology to address this need in the Australian State of New South Wales (801,428 km2). Multivariate patterns in data collected from 322 reference sites were used to define river types for each attribute: abiotic features (10 types), fish assemblages (6 types) and macroinvertebrate assemblages from river edges (8 types) and riffle zones (5 types). We used classification tree analysis to define broad regions for each attribute and then to construct identification keys for river types within each region using slope, elevation, maximum distance from source, latitude and mean annual rainfall. These keys allow the mapping of the likely spatial extent of river types and the assignment of a multi-attribute river-type identity to a river reach anywhere in the State. We used the average dissimilarity distances among the river types and the rates of misclassification of reference sites to assess the reliability of the assignments for different attributes in different regions. This approach to river classification can be applied anywhere in the world, resulting in simple to highly complex typologies depending on data availability. In data-poor areas it may result in a single attribute typology based on remotely derived variables and a coarsely defined reference condition. Previous studies have shown that the hydrogen atom transfer (HAT) reactions of tert-butoxyl radical from the Parkinsonian proneurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) occur with low selectivity at the allylic and non-allylic alpha-C-H positions. In this paper, we report a more comprehensive regiochemical study on the reactivity of the tert-butoxyl radical as well as on the associated primary kinetic deuterium isotope effects for the various hydrogen atom abstractions of MPTP. In addition, the results of a computational study to estimate the various C-H bond dissociation energies of MPTP are presented. The results of the present study show the allylic/non-allylic selectivity is approximately 73:21. The behavior of the tert-butoxyl radical mediated oxidation of MPTP contrasts with this reaction as catalyzed by monoamine oxidase B (MAO-B) that occurs selectively at the allylic alpha-carbon. These observations lead to the conclusion that the tert-butoxyl radical is not a good chemical model for the MAO-B-catalyzed bioactivation of MPTP.  相似文献

This study aimed to investigate the effects of the 5-HT7 receptor agonist (LP44) and antagonist (SB269970) on LPS-induced in vivo tissue damage and cell culture by molecular methods. This study was conducted in two steps. For in vivo studies, 24 female rats were divided into four groups. Group I: healthy; II (2nd h): LPS 5 mg/kg administered intraperitoneally (i.p.); III (4th h): LPS 5 mg/kg administered i.p.; IV (8th h): LPS 5 mg/kg administered i.p. For in vitro studies, we used the A549 cell line. Groups: I control (healthy) (2–4 h); II LPS: 1 µg/ml E. Coli O55:B5 strain (2–4 h); III agonist (LP44) 10?9 M (2–4 h); IV antagonist (SB269970) 10?9 M (2–4 h); V LPS+agonist 10?9 M (LP44 1 µg/ml) (2–4 h); VI LPS+antagonist 10?9 M (2–4 h). In molecular analyses, we determined increased TNF-α, IL-1β, NF-κB, and 5-HT7 mRNA expressions in rat lung tissues and increased TNF-α, iNOS, and 5-HT7 mRNA expressions in the A549 cell line. In in vitro parameters, LP44 agonist administration-related decrease was observed. Our study showed that lung 5-HT7 receptor expression is increased in LPS-induced endotoxemia. All this data suggest that 5-HT7 receptor overexpression is an important protective mechanism during LPS-induced sepsis-related cell damage.  相似文献

Low levels of histone covalent modifications are associated with gene silencing at telomeres and other regions in the yeast S. cerevisiae. Although the histone deacetylase Sir2 maintains low acetylation, mechanisms responsible for low H2B ubiquitylation and low H3 methylation are unknown. Here, we show that the ubiquitin protease Ubp10 targets H2B for deubiquitylation, helping to localize Sir2 to the telomere. Ubp10 exhibits reciprocal Sir2-dependent preferential localization proximal to telomeres, where Ubp10 serves to maintain low H2B Lys123 ubiquitylation in this region and, through previously characterized crosstalk, maintains low H3 Lys4 and Lys79 methylation in a slightly broader region. Ubp10 is also localized to the rDNA locus, a second silenced domain, where it similarly maintains low histone methylation. We compare Ubp10 to Ubp8, the SAGA-associated H2B deubiquitylase involved in gene activation, and show that telomeric and gene-silencing functions are specific to Ubp10. Our results suggest that these H2B-deubiquitylating enzymes have distinct genomic functions.  相似文献

In this study the effects of 23 carbohydrates belonging to various groups upon the survival egg production and egg of female adult Pimpla turionellae L. were investigated. The best results among the carbohydrates tested was obtained with sucrose which was also employed as control. Glucose, maltose, trehalose and melezitose on the other hand showed no significant effect. The egg production was observed to be unaffected by glucose and maltose although it showed significant increase with trehalose and significant decrease with melezitose. Fructose and sorbitol caused a significant decrease in the survival of the insect. Fructose and sorbitol did not have any significant effect upon egg production whereas galactose caused a significant decrease. With the exception of galactose, no carbohydrate caused any significant effect upon egg hatching. Although they did not produce any eggs the female insects survived for 13.17, 15.13, 11.58 and 15.83 days in mannose, melibiose, raffinose and mannitol, respectively. The shortest life span was observed in arabinose followed by α-methyl- D -glucoside, dulcitol, rhamnose, cellobiose, xylose, starch, lactose, sarbose, ribose and glycogen.  相似文献

The N-terminally myristoylated matrix (MA) domain of the HIV-1 Gag polyprotein promotes virus assembly by targeting Gag to the inner leaflet of the plasma membrane. Recent studies indicate that, prior to membrane binding, MA associates with cytoplasmic tRNAs (including tRNALys3), and in vitro studies of tRNA-dependent MA interactions with model membranes have led to proposals that competitive tRNA interactions contribute to membrane discrimination. We have characterized interactions between native, mutant, and unmyristylated (myr-) MA proteins and recombinant tRNALys3 by NMR spectroscopy and isothermal titration calorimetry. NMR experiments confirm that tRNALys3 interacts with a patch of basic residues that are also important for binding to the plasma membrane marker, phosphatidylinositol-4,5-bisphosphate [PI(4,5)P2]. Unexpectedly, the affinity of MA for tRNALys3 (Kd = 0.63 ± 0.03 μM) is approximately 1 order of magnitude greater than its affinity for PI(4,5)P2-enriched liposomes (Kd(apparent) = 10.2 ± 2.1 μM), and NMR studies indicate that tRNALys3 binding blocks MA association with liposomes, including those enriched with PI(4,5)P2, phosphatidylserine, and cholesterol. However, the affinity of MA for tRNALys3 is diminished by mutations or sample conditions that promote myristate exposure. Since Gag–Gag interactions are known to promote myristate exposure, our findings support virus assembly models in which membrane targeting and genome binding are mechanistically coupled.  相似文献

Neuroscience—a branch of biology seemingly distant from nature/wildlife conservation is revolutionised by the ability to visualise the brain activity of humans. Using positron emission tomography (PET), functional magnetic resonance imaging (fMRI) and magnetoencephalograhy (MEG), neuroscience is revealing how humans are wired in ways that have bearing on any problem that involves values; and nature/wildlife conservation is surely one of those. Understanding how the human brain represents and processes morality and sacred values, and responds to conflicts could shed a powerful light on nature/wildlife conservation tactics. Conservation polices typically involve utilitarian considerations. However, research shows that conservation policies based solely on utilitarian considerations are likely to fail as the neurological process of rights and wrongs grates against the process of cost and benefits.  相似文献

Over 500 million people interact daily with Facebook. Yet, whether Facebook use influences subjective well-being over time is unknown. We addressed this issue using experience-sampling, the most reliable method for measuring in-vivo behavior and psychological experience. We text-messaged people five times per day for two-weeks to examine how Facebook use influences the two components of subjective well-being: how people feel moment-to-moment and how satisfied they are with their lives. Our results indicate that Facebook use predicts negative shifts on both of these variables over time. The more people used Facebook at one time point, the worse they felt the next time we text-messaged them; the more they used Facebook over two-weeks, the more their life satisfaction levels declined over time. Interacting with other people “directly” did not predict these negative outcomes. They were also not moderated by the size of people''s Facebook networks, their perceived supportiveness, motivation for using Facebook, gender, loneliness, self-esteem, or depression. On the surface, Facebook provides an invaluable resource for fulfilling the basic human need for social connection. Rather than enhancing well-being, however, these findings suggest that Facebook may undermine it.  相似文献
