A methodology for obtaining reproducible in vitro induction of the synthesis of the acute phase reactant serum amyloid P-component (SAP) by purified mouse hepatocytes was established. Optimal hepatocyte culture conditions for the induction and synthesis of SAP required certain hormones, a substratum for cell attachment, and activated macrophages. Leibowitz L15 medium had to be supplemented with dexamethasone, indomethacin, insulin, glucose, and fetal bovine serum. Purified mouse IL 1 could substitute for activated macrophages in the induction of SAP. Hepatocytes were allowed to adhere to a collagen matrix to enhance both cell viability and SAP synthesis induced by IL 1. Elicited macrophages cultured with hepatocytes were capable of augmenting SAP synthesis in the presence of IL 1.  相似文献

The natural mortality rate of trout fry and the impact of weed cutting and cleaning out of weeds, branches and twigs from the stream-bed upon the mortality rate of trout fry was studied on several reaches in small streams. Above an initial density of 3.1 fry m–2 natural mortality rate was found to be density-dependent in the first few months of life. In reaches where weed cutting and cleaning out of the stream-bed had taken place the mortality rate was higher than that found under natural conditions in undisturbed reaches. The results support the suggestion that stream management works which make the stream more uniform will be in conflict with sound fishery management.  相似文献

Binding of 1,2-epoxy-3-butene, the primary metabolite of butadiene, to hemoglobin (Hb) and excretion of its mercapturic acid in urine were studies as potential indicators of butadiene exposure. Four groups of Wistar rats were exposed to butadiene at 0, 250, 500 and 1000 ppm 6 h/day, 5 days/week, during 2 weeks. Blood was collected at the end of exposure and 17 days later for analysis of hemoglobin adducts and adduct stability. Urine was collected each day during exposure (afternoon samples) and in between exposures (morning samples). Adducts of 1,2-epoxy-3-butene to N-terminal valine in Hb were measured using the N-alkyl Edman procedure and GC/MS of the thiohydantoin derivatives. The corresponding mercapturic acid was analysed, after deacetylation, through derivatization with phthaldialdehyde and HPLC with fluorescence detection. The Hb adducts proved to be stable and are therefore useful for dosimetry of long-term exposure to butadiene. The adduct levels increased linearly with exposure dose up to 1000 ppm (3 nmol/g Hb at 1000 ppm). The increase with exposure dose of the mercapturic acid concentration in urine was also compatible with a linear does response up to 1000 ppm. The sensitivity of both analytical methods needs to be improved for their application to human samples.  相似文献

Seasonal fluctuations of zooplankton biomass (dry weight) were determined during a year in two localities of Lake Xolotlán (Managua). Biomass estimations of the most common species of rotifers, cladocerans and copepods were made. The maximal zooplankton biomass was observed in February–April (dry season) in coincidence with the period of highest phytoplankton abundance. Copepods contributed with 78% and 84% to the mean zooplankton biomass at points 1 and 7, respectively. Cladocera biomass was lowest during most of the year, and it was probably controlled by fish predation. Development of rotifer biomass was more intense during the rainy season, when detritus particles were more abundant. Daily fluctuations of zooplankton biomass were not pronounced.  相似文献

Summary Five new elements of the mitotic control in the fission yeast Schizosaccharomyces pombe were isolated from gene libraries as multicopy suppressors of the conditional lethal phenotype of win1-1 weel ts cdc25ts triple mutant strains. These genes were designated wisl + –wis5+for win suppressing, and do not correspond to winl + or any of the previously characterised mitotic control genes. None of the wis genes is capable of suppressing the cdc phenotype of cdc25 ts strains, suggesting that their effect is not simply to reverse the effect of loss of cdc25 function. wisl + has been previously reported to encode a putative serine/threonine protein kinase that acts as a dosage-dependent inducer of mitosis. wis4 + appears to be a specific suppressor of the winl-1 mutation. wis2 + and wis3 + are capable of suppressing a wide range of cdc phenotypes arising from the combination of various mutations with wee1 ts and cdc25 ts, suggesting that the wis2 + and wis3 + products may interact with elements central to the mitotic control.  相似文献

The presence of a membrane receptor for C-reactive protein (CRP-R) on the human monocytic cell line U-937 was the basis for determining the metabolic fate of the receptor-bound ligand and the functional response of the cells to CRP. Internalized [125I]CRP was measured by removing cell surface-bound [125I]CRP with pronase. Warming cells to 37 degrees C resulted in the internalization of approx. 50% of the receptor-bound [125I]CRP or receptor-bound [125I]CRP-PC-KLH complexes. U-937 cells degraded about 25% of the internalized [125I]CRP into TCA-soluble radiolabeled products. The lysosomotrophic agents (chloroquine, NH4Cl) greatly decreased the extent of CRP degradation without altering binding or internalization. In addition, a pH less than 4.0 resulted in dissociation of receptor-bound [125I]CRP. Treatment of U-937 cell with monensin, a carboxylic ionophore which prevents receptor recycling, resulted in accumulation of internalized [125I]CRP. Therefore, it appears that the CRP-R complex is internalized into an endosomal compartment where the CRP is uncoupled from its receptor and subsequently degraded. CRP initiated the differentiation of the U-937 cells so that they acquired the ability to produce H2O2 and also display in vitro tumoricidal activity. The results support the concept that internalization and degradation of CRP leads to the activation of monocytes during inflammation.  相似文献

Synopsis The numbers of trout,Salmo trutta, in Granslev », Denmark, were estimated by the removal method on 18 dates from March 1974 to March 1976. Populations density varied from 0.39 to 0.74 trout m–2 in 1974–1975 and from 0.36 to 0.59 m–2 in 1975–1976 and at all times four or five year classes were present. The age structure of the population was unstable and the variable natural survival, immigration into and emigration from the study site could not be separated. An annual growth cycle with the most rapid growth for all year classes taking place from May to early August was found. Statistically significant differences between different years occurred in the growth of the 0,I and II age groups, but no evidence of density-dependent growth was found. The biomass ranged from 35.4 to 9.5 g m–2. The total mean annual biomass was 22.8 and 14.7 g m–2 in the two years and the II group made the greatest contribution, 44 and 48%, respectively. During 1975–1976 the mean annual biomass of each year class only was about two-thirds of that in 1974–1975. Annual production in the two years was 25.7 (range 24.7–28.5) and 12.6 g m–2 (range 11.7–15.0) and the II group accounted for about 46 and 38% of the production. In addition eel,Anguilla anguilla, produced about 0.5 g m–2 yr–2. The unstable age structure of the trout population was compared with trout populations from other streams. The importance of immigration as a recruitment process in middle and lower reaches of streams and of migrations as a mechanism to optimise utilization of the total stream habitat, as well as temperature as a factor controlling the growth rate are discussed.  相似文献

D‐type cyclins predominantly regulate progression through the cell cycle by their interactions with cyclin‐dependent kinases (cdks). Here, we show that stimulating mitogenesis of Swiss 3T3 cells with phorbol esters or forskolin can induce divergent responses in the expression levels, localization and activation state of cyclin D1 and cyclin D3. Phorbol ester‐mediated protein kinase C stimulation induces S phase entry which is dependent on MAPK activation and increases the levels and activation of cyclin D1, whereas forskolin‐mediated cAMP‐dependent protein kinase A stimulation induces mitogenesis that is independent of MAPK, but dependent upon mTor and specifically increases the level and activation of cyclin D3. These findings uncover additional levels of complexity in the regulation of the cell cycle at the level of the D‐type cyclins and thus may have important therapeutic implications in cancers where specific D‐cyclins are overexpressed. Isozyme analysis of young leaf tissue was performed on progeny produced from cultured ovules resulting from crosses of seedless × seedless grapes. Two polymorphic loci, Idh and Gpi-c, were found to segregate in a simple Mendelian fashion in the populations examined. Analyses of 70 seedlings indicated that most seedlings produced by this method were zygotic. Isozyme data of 11 plants from 5 polyembryonic ovules, along with data previously reported, suggest that several mechanisms may be operative in grape that give rise to polyembryony: 1) fertilization and development of more than one cell in the embryo sac, 2) adventive embryogenesis from the zygote, or 3) embryogenic development of gametic cells in addition to the zygote in the embryo sac.Fl. Plant growth can be limited by resource acquisition and defence against consumers, leading to contrasting trade‐off possibilities. The competition‐defence hypothesis posits a trade‐off between competitive ability and defence against enemies (e.g. herbivores and pathogens). The growth‐defence hypothesis suggests that strong competitors for nutrients are also defended against enemies, at a cost to growth rate. We tested these hypotheses using observations of 706 plant populations of over 500 species before and following identical fertilisation and fencing treatments at 39 grassland sites worldwide. Strong positive covariance in species responses to both treatments provided support for a growth‐defence trade‐off: populations that increased with the removal of nutrient limitation (poor competitors) also increased following removal of consumers. This result held globally across 4 years within plant life‐history groups and within the majority of individual sites. Thus, a growth‐defence trade‐off appears to be the norm, and mechanisms maintaining grassland biodiversity may operate within this constraint.  相似文献
