Savage score statistics are employed to develop a test for comparing survival distributions with right-hand singly censored data. The procedure is motivated by the interest in developing a powerful method for determining differences when true survival distributions cross. Examination of small-sample characteristics under the null hypothesis indicate that asymptotic critical values yield a slightly conservative test. Power of the test compares favorably with other criteria, including the modified Smirnov procedure, particularly if there is a single crossing of the survival curves.  相似文献

Production of platelet-activating factor by washed rabbit platelets under stimulation with the ionophore A23187 was investigated utilizing two groups of platelet preparations. The first platelet preparation contained 0.03 +/- 0.02% contaminating white cells, while the second preparation contained 0.48 +/- 0.27% white cells. The latter preparation produced platelet-activating factor, mainly 1-hexadecyl-2-acetyl-sn-glycero-3-phosphocholine, 8.3 +/- 6.3 pmol (mean +/- standard deviation) with a range of 2.6 to 21.4 pmol (n = 9), followed by small quantities of 1-octadecenyl- and 1-octadecyl-2-acetyl-sn-glycero-3-phosphocholine. In contrast, there was no production of 1-alkyl-2-acetyl-sn-glycero-3-phosphocholine by the former platelet preparation having 0.03% leukocytes. These quantitative analyses were carried out by the selected ion monitoring technique and it was concluded that it is necessary to consider the presence of contaminating white cells in studies on the production of platelet-activating factor by platelets.  相似文献

Cultured and propagated smooth muscle cells contain adenylate cyclase (AC) responsive to catecholamines and their analogues. Isoproterenol and zinterol were the most effective stimulants of AC activity with EC50 = 8.5 X 10(-8)M. They were followed by epinephrine, phenylephrine and norepinephrine (EC50 = 7.5 X 10(-7)M, 6.5 X 10(-6)M and 4 X 10(-6)M, respectively). When the selective antagonists for beta 1 and beta 2 receptors (beta 1-type practolol and atenolol, beta 1/beta 2-type propranolol and beta 2-type butoxamine) were tested against isoproterenol, epinephrine and norepinephrine stimulation of AC activity, the beta 1 in contrast to beta 2 antagonists were found ineffective. The alpha-blockers (phentolamine alpha 1/alpha 2-type antagonists) and yohimbine (alpha 2-type antagonist) alone or in the presence of propranolol did not significantly inhibit the catecholamine-induced enhancement of cAMP formation. On the other hand, prazosine (alpha 1-type antagonist) blocked the stimulatory effect of epinephrine and norepinephrine on AC system. Similarly, the alpha 2-agonist, clonidine, did not affect the catecholamines' stimulated AC activity while alpha 1 agonist, phenylephrine, induced an additive enhancement of norepinephrine production of cAMP. The findings of beta-2- and alpha-1-type adrenergic receptors in the cultured cerebrovascular smooth muscle provide additional support for the implicated involvement of adrenergic innervation in the regulation of cerebral blood flow and/or systemic blood pressure.  相似文献

Hydrocortisone is a modulator of cell division and has been shown to prolong the replicative in vitro life span of human embryonic lung fibroblasts. Time lapse cinematography was used to analyze the proliferative behavior of individual cells in populations of fibroblasts exposed to hydrocortisone in young cultures during a single growth cycle and in aged cultures that had been continuously exposed to hydrocortisone. Results indicate that hydrocortisone causes a decrease in the interdivision time (IDT) of a portion of the cells in the population and this effect is augmented after continuous exposure to hydrocortisone. Hydrocortisone does not appear to increase the number of initial dividers in the population but increases growth rate in the early stages of the culture period. Analysis of mother-daughter IDT pairs further suggests that hydrocortisone exerts its effects on IDT independently for a given cell.  相似文献

Previous studies with probabilistic neural nets in which the neural connections are set up by means of chemical markers, revealed the existence of multiple memory domains. We generalized these studies by considering the intrinsic noise of the systems, caused by the spontaneous release of synaptic transmitter substance. A simple mathematical model is developed, which yields characteristics of multiple memory domains analogous to those occurring in noiseless nets.  相似文献

Batch experiments were conducted to examine the effects of dissolved oxygen concentration on the degradation of 2,4-dichlorophenoxyacetic acid (2,4-D) by an enrichment culture of 2,4-D-utilizing bacteria. A modified Monod equation was found to describe the relationship between the specific growth rate and the concentrations of both the organic substrate and dissolved oxygen. Values for the maximum specific growth rate, yield, and Monod coefficient for growth on 2,4-D were 0.09 h-1, 0.14 g/g, and 0.6 mg/liter, respectively. The half-saturation constant for dissolved oxygen was estimated to be 1.2 mg/liter. These results suggest that dissolved oxygen concentrations below 1 mg/liter may be rate limiting for the biodegradation of chlorinated aromatic compounds such as 2,4-D, which have a requirement for molecular oxygen as a cosubstrate for metabolism.  相似文献

Recent investigations from this laboratory have identified a metallothionein-like protein in the rat brain with an elution volume (ve/vo) of 2.08 and a molecular weight of smaller than 10,000. The synthesis of this protein was stimulated following intracerebroventricular (icv, 0.20 μmol zinc/μL/h, 48 h), but not intraperitoneal (ip) administration of ZnSO4. Furthermore, chronic ip administration of ZnSO4 (5.0 mg/kg/d/10 d) did not alter the level of the metallothionein-like protein in the brain. However, the hepatic metallothionein was induced following icv administration of ZnSO4. The chromatofocusing of metallothionein-like protein isolated by gel permeation chromatography on Sephadex G-75 exhibits three zinc-binding peaks, which focus on pH 6.8, 6.2, and 5.3, respectively. It is expected that the protein peak focusing at 5.3 is a metallothionein-like protein. Purification of the zinc stimulated metallothionein-like protein on ion exchange chromatography on DEAE-Sephadex A-25 columns, using a linear gradient elution procedure produced two isoforms, eluting, respectively, at 75 and 137 mM of Tris-acetate buffer, pH 7.5. The comparative high-performance liquid chromatographic (HPLC) profiles of the zinc-induced hepatic metallothionein isoforms I and II (retention times 17.39 and 18.73 min) and that of the zinc-stimulated metallothionein-like protein isoforms I and II (retention times 17.32 and 18.64 min) are very similar. The function(s) of the metallothionein-like protein isoforms in the brain remains to be elucidated.  相似文献

Previous studies revealed that whistled songs of starlings can be separated into different categories, among which five themes are found in the repertoires of all (or most) individuals. These “specific” themes show local variations but the different themes have different sizes of dialect areas. This study investigates the temporal evolution of this complex dialectal system. Observations were made in an area where dialects were mapped three years earlier. They reveal an obvious temporal evolution of dialects, although the degree of variation differs according to the theme. No structural change was observed for two themes (CC and ST), but some temporal dynamics in their dialect boundaries did appear. There were changes in the structures and boundaries of two other themes (IT and HT). However, they showed:
  • — a relationship between new and old structures, either with persistence of the old variants or with an apparent drift. Almost no really new structures were observed.
  • — all the 1980 variants recorded in 1983 were found at sites not far from the original one. The main changes consisted of extensions or regressions of variants along boundaries.
Lastly, the temporal evolution of RT dialects is unusual in that the changes are more sudden. The variants have been replaced by different ones in some cases, but in others they have remained stable. The complexity of the dialectal system is persistent on a temporal level, varying from stability for ST to sudden changes for RT, with IT and HT intermediate.  相似文献

After reviewing basic technical considerations, we discuss some applications of flow cytometry in French laboratories. This methodology is used in several areas: oncology, cellular pharmacotoxicology, molecular biology and genetics, immunology, as well as cellular biology and physiology. We also examine the evolution of this technique in two directions: on the one hand, the appearance of increasingly sophisticated instruments; on the other, the development of less expensive and less complicated apparatuses principally directed at clinical applications.  相似文献
