On-line monitoring of the methanol concentration in Pichia pastoris cultures producing an heterologous lipase by sequential injection analysis 总被引:1,自引:0,他引:1
An automated sequential injection analysis (SIA) system using stop-flow technique was developed to determine methanol concentration by means of the enzymatic reactions of alcohol oxidase and peroxidase. Its application as an on-line device for monitoring Pichia pastoris fermentations producing an heterologous protein was demonstrated. Linear response, observed up to 2 g l–1, was reached by including a dilution chamber in the SIA manifold. The sampling frequency was 7 analyses per hour with a relative standard deviation lower than 4%. 相似文献

The breakthrough performance of stacks of dye-cellulosic fabric in affinity chromatography of lysozyme was investigated in batch and flow experiments. Breakthrough curves were significantly affected by flow rate and were not dependent on the feed solution concentration. System dispersion curves could not explain the flow-rate dependence. Breakthrough curves were analyzed by coupling the kinetic model for pore mass transfer as the only controlling resistance and a system dispersion model. From the analysis, pore film mass transfer resistance was found to be the leading rate-limiting factor when the residence time in the column is greater than 5 min. The model was used to predict the operating and design parameters needed to obtain sharp breakthrough curves. Selectivity studies using lysozyme and bovine serum albumin mixtures showed a high system selectivity for lysozyme. 相似文献

The bacterial Na+(Li+)/H+ antiporter NhaA has been expressed in the yeast Saccharomyces cerevisiae. NhaA was present in both the plasma membrane and internal membranes, and it conferred lithium but not sodium tolerance. In cells containing the yeast Ena1-4 (Na+, Li+) extrusion ATPase, the extra lithium tolerance conferred by NhaA was dependent on a functional vacuolar H+ ATPase and correlated with an increase of lithium in an intracellular pool which exhibited slow efflux of cations. In yeast mutants without (Na+, Li+) ATPase, lithium tolerance conferred by NhaA was not dependent on a functional vacuolar H+ ATPase and correlated with a decrease of intracellular lithium. NhaA was able to confer sodium tolerance and to decrease intracellular sodium accumulation in a double mutant devoid of both plasma membrane (Na+, Li+) ATPase and vacuolar H+ ATPase. These results indicate that the bacterial antiporter NhaA expressed in yeast is functional at both the plasma membrane and the vacuolar membrane. The phenotypes conferred by its expression depend on the functionality of plasma membrane (Na+, Li+) ATPase and vacuolar H+ ATPase. 相似文献

Production of a Rhizopus oryzae lipase from Pichia pastoris using alternative operational strategies

Different cultivation strategies have been compared for the production of Rhizopus oryzae lipase (ROL) from Pichia pastoris. Several drawbacks have been found using a methanol non-limited fed-batch. On the one hand, oxygen limitation appeared at early cell dry weights and, on the other hand, high cell death was observed. A temperature limited fed-batch has been proposed to solve both problems. However, in our case study a methanol non-limited fed-batch results in better productivities. Finally, a lower salt medium were used to overcome cell death problems and a temperature limited fed-batch was applied thereafter to solve oxygen transfer limitations. This combined strategy has resulted in lower productivities when compared to a methanol non-limited fed-batch. However the culture could be longer prolonged and a 1.3-fold purer final product was obtained mainly due to cell death reduction. 相似文献

The behaviour of Pseudomonas fluorescens EPS62e was investigated in apple and pear orchards under Mediterranean climatic conditions. The trials studied the influence of weather conditions, plant host species, presence of indigenous microbial community and spread from treated to nontreated trees on colonization and survival. Population dynamics were assessed by real-time PCR and CFU-counting methods. With inoculated flowers, weather conditions were optimal for colonization, and EPS62e established high and stable population levels around 10(8) CFU per organ, according to both methods of analysis. The plant host species did not influence the colonization rate, and the biocontrol agent dominated the microbial communities of blossoms, representing up to 100% of the total cultivable population. With inoculated leaves, the EPS62e population decreased to nondetectable levels 30 days after treatment according to both methods used. EPS62e spread moderately in the orchard, being detected in nontreated flowers of trees 15-35 m from the inoculation site. The combined use of real-time PCR and CFU-counting methods of analysis permitted the identification of three physiological states for EPS62e in the field, which consisted of active colonization, survival and entry into a viable but nonculturable state, and cell death. 相似文献

A 125-member library of synthetic linear undecapeptides was prepared based on a previously described peptide H-K1KLFKKILKF10L-NH2 (BP76) that inhibited in vitro growth of the plant pathogenic bacteria Erwinia amylovora, Xanthomonas axonopodis pv. vesicatoria, and Pseudomonas syringae pv. syringae at low micromolar concentrations. Peptides were designed using a combinatorial chemistry approach by incorporating amino acids possessing various degrees of hydrophobicity and hydrophilicity at positions 1 and 10 and by varying the N-terminus. Library screening for in vitro growth inhibition identified 27, 40 and 113 sequences with MIC values below 7.5 μM against E. amylovora, P. syringae and X. axonopodis, respectively. Cytotoxicity, bactericidal activity and stability towards protease degradation of the most active peptides were also determined. Seven peptides with a good balance between antibacterial and hemolytic activities were identified. Several analogues displayed a bactericidal effect and low susceptibility to protease degradation. The most promising peptides were tested in vivo by evaluating their preventive effect of inhibition of E. amylovora infection in detached apple and pear flowers. The peptide H-KKLFKKILKYL-NH2 (BP100) showed efficacies in flowers of 63–76% at 100 μM, being more potent than BP76 and only less effective than streptomycin, currently used for fire blight control. 相似文献

Several diseases caused by viruses, bacteria and fungi affect plant crops, resulting in losses and decreasing the quality and safety of agricultural products. Plant disease control relies mainly on chemical pesticides that are currently subject to strong restrictions and regulatory requirements. Antimicrobial peptides are interesting compounds in plant health because there is a need for new products in plant protection that fit into the new regulations. Living organisms secrete a wide range of antimicrobial peptides produced through ribosomal (defensins and small bacteriocins) or non-ribosomal synthesis (peptaibols, cyclopeptides and pseudopeptides). Several antimicrobial peptides are the basis for the design of new synthetic analogues, have been expressed in transgenic plants to confer disease protection or are secreted by microorganisms that are active ingredients of commercial biopesticides. 相似文献

Planarians are highly regenerative organisms with the ability to remake all their cell types, including the germ cells. The
germ cells have been suggested to arise from totipotent neoblasts through epigenetic mechanisms. Nanos is a zinc-finger protein
with a widely conserved role in the maintenance of germ cell identity. In this work, we describe the expression of a planarian
nanos-like gene Smednos in two kinds of precursor cells namely, primordial germ cells and eye precursor cells, during both development and regeneration
of the planarian Schmidtea mediterranea. In sexual planarians, Smednos is expressed in presumptive male primordial germ cells of embryos from stage 8 of embryogenesis and throughout development
of the male gonads and in the female primordial germ cells of the ovary. Thus, upon hatching, juvenile planarians do possess
primordial germ cells. In the asexual strain, Smednos is expressed in presumptive male and female primordial germ cells. During regeneration, Smednos expression is maintained in the primordial germ cells, and new clusters of Smednos-positive cells appear in the regenerated tissue. Remarkably, during the final stages of development (stage 8 of embryogenesis)
and during regeneration of the planarian eye, Smednos is expressed in cells surrounding the differentiating eye cells, possibly corresponding to eye precursor cells. Our results
suggest that similar genetic mechanisms might be used to control the differentiation of precursor cells during development
and regeneration in planarians.

Animal behaviour is of fundamental importance but is often overlooked in biological invasion research. A problem with such studies is that they may add pressure to already threatened species and subject vulnerable individuals to increased risk. One solution is to obtain the maximum possible information from the generated data using a variety of statistical techniques, instead of solely using simple versions of linear regression or generalized linear models as is customary. Here, we exemplify and compare the use of modern regression techniques which have very different conceptual backgrounds and aims (negative binomial models, zero-inflated regression, and expectile regression), and which have rarely been applied to behavioural data in biological invasion studies. We show that our data display overdispersion, which is frequent in ecological and behavioural data, and that conventional statistical methods such as Poisson generalized linear models are inadequate in this case. Expectile regression is similar to quantile regression and allows the estimation of functional relationships between variables for all portions of a probability distribution and is thus well suited for modelling boundaries in polygonal relationships or cases with heterogeneous variances which are frequent in behavioural data. We applied various statistical techniques to aggression in invasive mosquitofish, Gambusia holbrooki, and the concomitant vulnerability of native toothcarp, Aphanius iberus, in relation to individual size and sex. We found that medium sized male G. holbrooki carry out the majority of aggressive acts and that smaller and medium size A. iberus are most vulnerable.
