Boldenone is an androgenic steroid that improves the growth and food conversion in food producing animals. In most countries worldwide, this anabolic steroid is forbidden for meat production. Until recently, the control of its illegal use was based either on 17beta-boldenone or 17alpha-boldenone (its main metabolite in cattle) identification in edible tissues, hair, faeces or urine. Recent observations and data tend to demonstrate the natural occurrence (but not ubiquitous) in cattle of these steroids, making the analytical strategy of the control more complicated. We investigated the metabolism of boldenone in cattle after intramuscular and oral treatment of boldenone, boldenone esters and boldione. The central objective was to elucidate the structures of the main metabolites (phase I and phase II) in urine, with main objective to be further in position to compare boldenone urinary profiles of treated and non-treated animals. Nine metabolites have been identified, only four were present whatever the treatment and the administered boldenone source. Nevertheless, all of them have been detected at least once in non-treated animals which did not permit us to use them as biomarkers of an illegal treatment. At last, but not at least, all metabolites were found mainly glucuro-conjugated, and rarely sulfo-conjugated, with the only exception of 17beta-boldenone. Current investigations are showing the absence of 17beta-boldenone sulfoconjugate in non-treated animals; that would permit to distinguish non-treated from treated animals with boldione, boldenone and boldenone esters.  相似文献

BackgroundIncreases in the proportion of the population with increased likelihood of cesarean section (CS) have been postulated as a driving force behind the rise in CS rates worldwide. The aim of the study was to assess if changes in selected maternal risk factors for CS are associated with changes in CS births from 1999 to 2016 in Norway.Methods and findingsThis national population-based registry study utilizes data from 1,055,006 births registered in the Norwegian Medical Birth Registry from 1999 to 2016. The following maternal risk factors for CS were included: nulliparous/≥35 years, multiparous/≥35 years, pregestational diabetes, gestational diabetes, hypertensive disorders, previous CS, assisted reproductive technology, and multiple births. The proportion of CS births in 1999 was used to predict the number of CS births in 2016. The observed and predicted numbers of CS births were compared to determine the number of excess CS births, before and after considering the selected risk factors, for all births, and for births stratified by 0, 1, or >1 of the selected risk factors. The proportion of CS births increased from 12.9% to 16.1% (+24.8%) during the study period. The proportion of births with 1 selected risk factor increased from 21.3% to 26.3% (+23.5%), while the proportion with >1 risk factor increased from 4.5% to 8.8% (+95.6%). Stratification by the presence of selected risk factors reduced the number of excess CS births observed in 2016 compared to 1999 by 67.9%. Study limitations include lack of access to other important maternal risk factors and only comparing the first and the last year of the study period.ConclusionsIn this study, we observed that after an initial increase, proportions of CS births remained stable from 2005 to 2016. Instead, both the size of the risk population and the mean number of risk factors per birth continued to increase. We observed a possible association between the increase in size of risk population and the additional CS births observed in 2016 compared to 1999. The increase in size of risk population and the stable CS rate from 2005 and onward may indicate consistent adherence to obstetric evidence-based practice in Norway.

Purpose

Along with climate change-related issues, improved water management is recognized as one of the major challenges to sustainability. However, there are still no commonly accepted methods for measuring sustainability of water uses, resulting in a recent proliferation of water footprint methodologies. The Water Impact Index presented in this paper aims to integrate the issues of volume, scarcity and quality into a single indicator to assess the reduction of available water for the environment induced by freshwater uses for human activities.

Methods

The Water Impact Index follows life cycle thinking principles. For each unit process, a volumetric water balance is performed; water flows crossing the boundaries between the techno-sphere and environment are multiplied by a water quality index and a water scarcity index. The methodology is illustrated on the current municipal wastewater management system of Milan (Italy). The Water Impact Index is combined with carbon footprint to introduce multi-impact thinking to decision makers. The Water Impact Index is further compared to results obtained using a set of three life cycle impact indicators related to water, from the ReCiPe life cycle impact assessment (LCIA) methodology.

Results and discussion

Onsite water use is the main contribution to the Water Impact Index for both wastewater management schemes. The release of better quality water is the main driver in favour of the scenario including a wastewater treatment plant, while the energy and chemicals consumed for the treatment increase the indirect water footprint and carbon footprint. Results obtained with the three midpoint indicators depict similar tendencies to the Water Impact Index.

Conclusions

This paper presents a simplified single-indicator approach for water footprinting, integrating volume, scarcity and quality issues, representing an initial step toward a better understanding and assessment of the environmental impacts of human activities on water resources. The wastewater treatment plant reduces the Water Impact Index of the wastewater management system. These results are consistent with the profile of the three midpoint indicators related to water from ReCiPe.  相似文献

Extracellular ATP (released by endothelial and immune cells) and its metabolite ADP are important pro-inflammatory mediators via the activation of purinergic P2 receptors (P2Y and P2X), which represent potential new targets for anti-inflammatory therapy. Endothelial P2Y1 receptor (P2Y1R) induces endothelial cell activation triggering leukocyte adhesion. A number of data have implicated melatonin as a modulator of immunity, inflammation, and endothelial cell function, but to date no studies have investigated whether melatonin modulates endothelial P2YR signaling. Here, we evaluated the putative effect of melatonin on P2Y1R-mediated leukocyte adhesion to endothelial cells and TNF-α production, using mesenteric endothelial cells and fresh peripheral blood mononuclear cells isolated from rats. Endothelial cells were treated with the P2Y1R agonist 2MeSATP, alone or in combination with melatonin, and then exposed to mononuclear cells. 2MeSATP increased leukocyte adhesion to endothelial cells and TNF-α production in vitro, and melatonin inhibited both effects without altering P2Y1R protein expression. In addition, assays with the Ca2+ chelator BAPTA-AM indicate that the effect of melatonin on 2MeSATP-stimulated leukocyte adhesion depends on intracellular Ca2+ modulation. P2Y1R is considered a potential target to control chronic inflammation. Therefore, our data unveiled a new endothelial cell modulator of purinergic P2Y1 receptor signaling.  相似文献

AMP-activated protein kinase (AMPK) is a heterotrimer of catalytic (α) and regulatory (β and γ) subunits with at least two isoforms for each subunit. AMPK β1 is widely expressed whilst AMPK β2 is highly expressed in muscle and both β isoforms contain a mid-molecule carbohydrate-binding module (β-CBM). Here we show that β2-CBM has evolved to contain a Thr insertion and increased affinity for glycogen mimetics with a preference for oligosaccharides containing a single α-1,6 branched residue. Deletion of Thr-101 reduces affinity for single α-1,6 branched oligosaccharides by 3-fold, while insertion of this residue into the equivalent position in the β1-CBM sequence increases affinity by 3-fold, confirming the functional importance of this residue.  相似文献
