Explants of fetal mouse spinal cord and cerebral cortex generate organotypic slow-wave and repetitive-spike discharges in vitro which can be abolished by agents which reduce the concentration of Ca++ available to the tissue. Synaptically mediated discharges are rapidly blocked in Ca++-free balanced salt solution (BSS), or in regular BSS after addition of 10?3 M EGTA, 5–10 × 10?3 M Mg++, or 10?4 M xylocaine, but simple spike potentials can still propagate. When low concentrations of cyclic AMP or dibutyryl cyclic AMP (2 × 10?6 M) are added to the Ca ++-free BSS or Ca++-antagonist-BSS, a temporary (1–20 min) restoration of characteristic complex bioelectric activities occurs (or the onset of depression is delayed if cyclic AMP is initially added). Phosphodiesterase inhibitors, e.g. 10?3 M caffeine, are also effective in restoring these blockades, whereas 5′AMP and ATP are not. Application of 10?6 M cyclic AMP or 10?3 M caffeine in regular BSS greatly enhances excitability of some CNS explants, resembling convulsive effects observed in CNS in situ. The data suggest that cyclic AMP can mobilize Ca++ from membranebound Ca pools within neurons in CNS explants so as to permit Ca++-dependent release of neurotransmitter during Ca++ deficits. Thus, it may also be that under normal conditions, cyclic AMP can regulate the availability of Ca++ for synaptic transmission in the central nervous system, thereby modulating the efficacy of synaptic functions. 相似文献

Previous experiments in anesthetized or decerebrate cats showed that the vestibular system participates in adjusting blood pressure during postural changes. The present experiments tested the hypothesis that removal of vestibular inputs in awake cats would affect orthostatic tolerance. Before the lesion, blood pressure typically remained within 10 mmHg of baseline values during nose-up-pitch body rotations of up to 60 degrees in amplitude. In contrast, bilateral peripheral vestibular lesions altered the pattern of orthostatic responses in all animals, and blood pressure fluctuated >10 mmHg from baseline values during most 60 degrees nose-up tilts in five of six animals. The deficit in correcting blood pressure was particularly large when the animal also was deprived of visual cues indicating position in space. During this testing condition, either a decrease or increase in blood pressure >10 mmHg in magnitude occurred in >80% of tilts. The deficit in adjusting blood pressure after vestibular lesions persisted for only 1 wk, after which time blood pressure remained stable during tilt. These data show that removal of vestibular inputs alters orthostatic responses and are consistent with the hypothesis that vestibular signals are one of several inputs that are integrated to elicit compensatory changes in blood pressure during movement. 相似文献

The aim of the present study was to ascertain whether cell-mediated immunity (CMI) is induced in animals by vaccination with Candida albicans ribosomes. Delayed type hypersensitivity (DTH) was detected in vivo in ribosome-vaccinated mice and guinea pigs by the footpad swelling and skin tests, respectively. The observed DTH was similar to that induced by live C. albicans organisms. A lymphocyte transformation assay was used for in vitro detection of CMI. The tritiated thymidine incorporation assays revealed that spleen lymphocytes from mice immunized with C. albicans ribosomes were stimulated by the ribosomal antigen. The findings establish that C. albicans ribosomes are able to induce CMI in experimental animals. 相似文献
