Dendritic network structure and dispersal affect temporal dynamics of diversity and species persistence

Landscape connectivity structure, specifically the dendritic network structure of rivers, is expected to influence community diversity dynamics by altering dispersal patterns, and subsequently the unfolding of species interactions. However, previous comparative and experimental work on dendritic metacommunities has studied diversity mostly from an equilibrium perspective. Here we investigated the effect of dendritic versus linear network structure on local (α‐diversity), among (β‐diversity) and total (γ‐diversity) temporal species community diversity dynamics. Using a combination of microcosm experiments, which allowed for active dispersal of 14 protists and a rotifer species, and numerical analyses, we demonstrate the general importance of spatial network configuration and basic life history tradeoffs as driving factors of different diversity patterns in linear and dendritic systems. We experimentally found that community diversity patterns were shaped by the interaction of dispersal within the networks and local species interactions. Specifically, α‐diversity remained higher in dendritic networks over time, especially at highly connected sites. β‐diversity was initially greater in linear networks, due to increased dispersal limitation, but became more similar to β‐diversity in dendritic networks over time. Comparing the experimental results with a neutral metacommunity model we found that dispersal and network connectivity alone may, to a large extent, explain α‐ and β‐diversity dynamics. However, additional mechanisms, such as variation in carrying capacity and competition–colonization tradeoffs, were needed in the model to capture the detailed temporal diversity dynamics of the experiments, such as a general decline in γ‐diversity and long‐term dynamics in α‐diversity. 相似文献

The functional relevance of regulating proteins is often limited to specific binding sites such as the ends of microtubules or actin-filaments. A localization of proteins on these functional sites is of great importance. We present a quantitative theory for a diffusion and capture process, where proteins diffuse on a filament and stop diffusing when reaching the filament’s end. It is found that end-association after one-dimensional diffusion is the main source for tip-localization of such proteins. As a consequence, diffusion and capture is highly efficient in enhancing the reaction velocity of enzymatic reactions, where proteins and filament ends are to each other as enzyme and substrate. We show that the reaction velocity can effectively be described within a Michaelis-Menten framework. Together, one-dimensional diffusion and capture beats the (three-dimensional) Smoluchowski diffusion limit for the rate of protein association to filament ends. 相似文献

Axonal and dendritic transport in single Purkinje neurons of cerebellar slice cultures was quantified as single transport
distances. Examination of the cells within a vital tissue was regarded as being an approach to the in situ condition. The
Purkinje cells were organotypically integrated in the in vitro tissues and extended long axonal projections connecting synapses
to the target neurons. The tracer horseradish peroxidase (HRP) was applied via microinjection to the somata of the Purkinje
cells and the injected neurons were incubated thereafter for defined time-intervals. The tracer was transported anterogradely
into the neuron processes. The measurements on both the axonal and the dendritic transport of microinjected HRP revealed continuous
transportation with increasing times of postincubation. This transport was reduced by the use of microtubule-depolymerizing
drugs. The axonal transport of the tracer was either retarded in colchicine-treated cells or continuously reduced for up to
50% in vinblastine-treated neurons. Thus, a correlation of axonal transport to the microtubules was demonstrated. The dendrites
were filled with the tracer after 60 min of postincubation. Dendritic transport was reduced by the use of vinblastine, and
not significantly by colchicine. The results strongly support the dependence of neuronal transport on microtubules as a component
of the cytoskeleton.

We identified a short RNA fragment, complementary to the Escherichia coli 23S rRNA segment comprising nucleotides 735 to 766 (in domain II), which when expressed in vivo results in the suppression of UGA nonsense mutations in two reporter genes. Neither UAA nor UAG mutations, examined at the same codon positions, were suppressed by the expression of this antisense rRNA fragment. Our results suggest that a stable phylogenetically conserved hairpin at nucleotides 736 to 760 in 23S rRNA, which is situated close to the peptidyl transferase center, may participate in one or more specific interactions during peptide chain termination. 相似文献

OBJECTIVE: To investigate how the morphological and physiological properties of single myocytes isolated from the hypertrophied, failing left ventricles (LV) differ from those of normal or hypertrophied not failing ventricles. METHOD: Single myocytes were isolated separately from right (RV) and left ventricles (LV) of male spontaneously hypertensive rats (SHR) or Wistar-Kyoto (WKY) rats at the age of 6 and 12 months and of SHRs which developed or not developed heart failure at the age of 20-24 months. We measured cells dimensions, range and kinetics of electrically stimulated or initiated by caffeine contractions and Ca2+ transients, and investigated the response of cells to thapsigargin. RESULTS: The transversal dimensions of the LV myocytes of 6 months old SHRs showed approximately 20% increase with respect to transversal dimensions of their RV myocytes and LV and RV myocytes of WKY rats. The difference did not change with progressing age and in the heart failure. The LV myocytes of 6 or 12 months old SHRs showed slowed kinetics of the Ca2+ transients and of contraction and relaxation and decreased contractile response to 2 s superfusion with 15 mM caffeine preceded by 5 mM Ni2+ used as an index of the sarcoplasmic reticulum (SR) Ca2+ content. Despite of this the range of shortening and relative contribution of the SR to contraction (assessed by measuring of the residual contractile response to electrical stimulation in cells poisoned with thapsigargin) or relaxation (assessed by calculation of the ratio of rate constants of the electrically stimulated and stimulated by 30 s superfusion with caffeine Ca2+ transients) was not altered in the hypertrophied myocytes. Properties of the LV myocytes of the 20-24 old SHRs with or without heart failure did not differ from those of LV myocytes of younger SHRs. The contractile response to caffeine of their RV myocytes dropped to the level of that in the LV myocytes. CONCLUSION: Our results suggest that transition from the compensated hypertrophy to the heart failure in 20-24 months old SHRs did not result from the further changes in properties of the surviving myocytes. Data from literature suggest that myocyte apoptosis and remodeling of the extramyocyte space is the more likely reason. 相似文献

Proximal mouse Chromosome (Chr) 16 shows conserved synteny with human Chrs 16, 8, 22, and 3. The mouse Chr 16/human Chr 22
conserved synteny region includes the DiGeorge/Velocardiofacial syndrome region of human Chr 22q11.2. A physical map of the
entire mouse Chr 16/human Chr 22 region of conserved synteny has been constructed to provide a substrate for gene discovery,
genomic sequencing, and animal model development. A YAC contig was constructed that extends ca. 5.4 Mb from a region of conserved
synteny with human Chr 8 at Prkdc through the region conserved with human Chr 3 at DVL3. Sixty-one markers including 37 genes are mapped with average marker
spacing of 90 kb. Physical distance was determined across the 2.6-Mb region from D16Mit74 to Hira with YAC fragmentation. The central region from D16Jhu28 to Igl-C1 was converted into BAC and PAC clones, further refining the physical map and providing sequence-ready template. The gene
content and borders of three blocks of conserved linkage between human Chr 22q11.2 mouse Chr 16 are refined.
