Disturbance can impact natural communities in multiple ways. However, there has been a tendency to focus on single indicators of change when examining the effects of disturbance. This is problematic as classical diversity measures, such as Shannon and Simpson indices, do not always detect the effects of disturbance. Here, we instead take a multilevel, hierarchical approach, looking for signatures of disturbance in the capacity and diversity of the community, and also in allocation and demography at the population level. Using recreational use as an example of disturbance, and the freshwater streams of Trinidad as a model ecosystem, we repeatedly sampled the fish communities and physical parameters of eight pairs of recreational and nonrecreational sites every 3 months over a 28‐month period. We also chose the Trinidadian guppy (Poecilia reticulata) as the subject of our population‐level analyses. Regression tree analysis, together with analysis of deviance, revealed that community capacity and community species richness were greater at sites with higher levels of recreational use. Interestingly, measures of community diversity that took into account the proportional abundance of each species were not significantly associated with recreational use. Neither did we find any direct association between recreational use and proportion of guppy biomass in the community. However, population‐level differences were detected in the guppy: Sex ratio was significantly more female‐biased at more disturbed sites. Our findings emphasize the importance of considering multiple levels when asking how disturbance impacts a community. We advocate the use of a multilevel approach when monitoring the effects of disturbance, and highlight gaps in our knowledge when it comes to interpreting these effects.  相似文献

Understanding how genetic variation is maintained within species is a major goal of evolutionary genetics that can shed light on the preservation of biodiversity. Here, we examined the maintenance of a regulatory single-nucleotide polymorphism (SNP) of the X-linked Drosophila melanogaster gene fezzik. The derived variant at this site is at intermediate frequency in many worldwide populations but absent in populations from the ancestral species range in sub-Saharan Africa. We collected and genotyped wild-caught individuals from a single European population biannually over a period of 5 years, which revealed an overall difference in allele frequency between the sexes and a consistent change in allele frequency across seasons in females but not in males. Modeling based on the observed allele and genotype frequencies suggested that both sexually antagonistic and temporally fluctuating selection may help maintain variation at this site. The derived variant is predicted to be female-beneficial and mostly recessive; however, there was uncertainty surrounding our dominance estimates and long-term modeling projections suggest that it is more likely to be dominant. By examining gene expression phenotypes, we found that phenotypic dominance was variable and dependent upon developmental stage and genetic background, suggesting that dominance may be variable at this locus. We further determined that fezzik expression and genotype are associated with starvation resistance in a sex-dependent manner, suggesting a potential phenotypic target of selection. By characterizing the mechanisms of selection acting on this SNP, our results improve our understanding of how selection maintains genetic and phenotypic variation in natural populations.  相似文献

We use microsatellite loci to detail the multiple paternity patterns in broods from 10 wild populations of the guppy ( Poecilia reticulata ) found in Northern Trinidad. The populations span two major drainages comprising the Caroni and the Oropouche, and include sites that are characterized by either high or low predation. Across the populations the frequency of multiple paternity is high with 95% (range: 70%–100%) of broods having multiple sires. Broods have an average of 3.5 sires (range: 1–9) and a mixed-model analysis suggests that broods from high predation sites have marginally more sires than do those from low predation sites, but this is true only in the Oropouche drainage. There is no difference in sire number between predation sites in the Caroni drainage. Brood size, but not female body length, is correlated with the number of sires and the correlation cannot be attributed solely to the stochastic process associated with sperm competition and a 'fair raffle'. Within broods there is significant skew in reproductive success among males, which may reflect variation in sperm competitiveness or female choice. There is, however, no difference in the skew among populations from different predation regimes or drainages. Finally, high predation populations were characterized by increased genetic variability at the microsatellite loci, suggesting a larger effective population size. We discuss explanations for the high degree of multiple paternity but the general lack of any major differences among broods from ecologically different populations.  相似文献

Evolution of reproductive isolation is an important process, generating biodiversity and driving speciation. To better understand this process, it is necessary to investigate factors underlying reproductive isolation through various approaches but also in various taxa. Previous studies, mainly focusing on diploid animals, supported the prevalent view that reproductive barriers evolve gradually as a by-product of genetic changes accumulated by natural selection by showing a positive relationship between the degree of reproductive isolation and genetic distance. Haplodiploid animals are expected to generate additional insight into speciation, but few studies investigated the prevalent view in haplodiploid animals. In this study, we investigate whether the relationship also holds in a haplodiploid spider mite, Amphitetranychus viennensis (Zacher). We sampled seven populations of the mite in the Palaearctic region, measured their genetic distance (mtDNA) and carried out cross experiments with all combinations. We analyzed how lack of fertilization rate (as measure of prezygotic isolation) as well as hybrid inviability and hybrid sterility (as measures of postzygotic isolation) varies with genetic distance. We found that the degree of reproductive isolation varies among cross combinations, and that all three measures of reproductive isolation have a positive relationship with genetic distance. Based on the mtDNA marker, lack of fertilization rate, hybrid female inviability and hybrid female sterility were estimated to be nearly complete (99.0–99.9% barrier) at genetic distances of 0.475–0.657, 0.150–0.209 and 0.145–0.210, respectively. Besides, we found asymmetries in reproductive isolation. The prevalent view on the evolution of reproductive barriers is supported in the haplodiploid spider mite we studied here. According to the estimated minimum genetic distance for total reproductive isolation in parent population crosses in this study and previous work, a genetic distance of 0.15–0.21 in mtDNA (COI) appears required for speciation in spider mites. Variations and asymmetries in the degree of reproductive isolation highlight the importance of reinforcement of prezygotic reproductive isolation through incompatibility and the importance of cytonuclear interactions for reproductive isolation in haplodiploid spider mites.  相似文献

A series of synthetic peptide fragments derived from RANTES were designed, synthesized, and evaluated to determine the effect of N-terminal truncation on the ability of the lead compound Ac[Ala(10,11)]RANTES-(1-14)NH(2) to inhibit HIV-1 infectivity. Both the lead compound and the truncated analogue Ac[Ala(10,11)]RANTES-(3-14)NH(2) were able to significantly inhibit HIV-1 infectivity. These results suggest that a small synthetic peptide may be able to mimic RANTES and have the ability to prevent transmission of HIV-1.  相似文献

Summary CCR5 (CC-chemokine receptor 5) is a key co-receptor, in concert with CD4, for infectivity of HIV-1 (human immunodeficiency virus type-1) into healthy human cells, and RANTES, an endogenous ligand for CCR5, is a potent inhibitor of HIV-1 infectivity. In this structure-activity relationship (SAR) study, peptide fragments derived from RANTES were designed, synthesized and evaluated for their ability to inhibit HIV-1 infectivity. The goal was to determine the effect of peptide length on anti-HIV activity and to obtain an optimally sized RANTES peptide probe for further SAR studies. The analogue Ac[Ala10,11]RANTES-(1–14)NH2, AA14, was identified as an effective inhibitor of HIV-1 infectivity at 10 nM but despite the functional activity, surprisingly it did not exhibit any notable affinity for the CCR5 chemokine receptor. Further, increasing peptide size enhanced neither the inhibition of HIV-1 infectivity nor CCR5 receptor affinity. As a potent inhibitor of HIV-1 infectivity, the lead analogue most likely utilizes a different (and currently unknown) mechanism than interaction with CCR5 for anti-HIV activity.  相似文献

Synopsis Newly hatched larval Hoplosternum littorale do not exhibit bimodal respiration upon hatching but depend initially on branchial respiration. As the respiratory intestine develops, its increasing length leads to an increasing dependence on aerial respiration. The respiratory intestine first appears when the fish is 9 days old but is microscopic at this age and becomes detectable by gross morphological examination at 13 days old. Development of the gut for the remainder of the juvenile period (up to 32 days old) consists of increasing length, mucosal folding, concentration of taste buds, gastric glands, mucous glands and goblet cells. Stage 1 commenced with newly hatched larvae until the larval fish were 11 days old. During this stage the fish did not have the capacity to air-breathe and thus behaved as oxygen conformers. Stage 2 extended from day 12 to day 23 and the fish had the capacity to air-breathe although the respiratory intestine was still developing. Stage 3 began from day 24 to day 32 and at this point the respiratory intestine was well-developed and functioned efficiently. The rate of oxygen consumption of the larval fish was found to be related to dissolved oxygen concentration, age of the larval fish and various interactions of these two factors.  相似文献

In callichthyid catfishes, the posterior intestine is modified to function as an air breathing organ by being air-filled, thin-walled and highly vascularized. These modifications make it unsuitable for digestive functions and digesta has to be transported quickly through this region to minimize disruption of vital respiratory functions. However, the weak muscles of the wall of the respiratory intestine make this problematic. It is hypothesized that the unidirectional ventilatory air current within the respiratory intestine is responsible for the quick transport of digesta through the respiratory intestine. To verify this, movement of digesta through the alimentary tract was examined in Hoplosternum littorale and Corydoras aeneus that were either allowed to breathe air or prevented from air breathing. When air breathing was prevented, digesta was not transported to the rectum in H. littorale and there was a 94% reduction in the amount of digesta in the rectum of C. aeneus. This study suggests that the anterior digestive intestine facilitates the passage of air although it is filled with digesta. The anterior digestive intestine packages digesta into a string of slightly compressed boluses, creating an air channel in the digestive intestine thus allowing air to pass unimpeded.  相似文献
