In January-February 1991, in Prydz Bay, phytoplankton bloomwas evident in the inner shelf area with the dominant diatomsbeing represented mainly by pennate species of the Nitzschia-Fragilariopsisgroup. Dinoflagellates and naked flagellates were most abundantin the centre of the bay; however, larger heterotrophic speciesprevailed at the southern stations. Cell carbon values (average317 µg l–1; range 92-1048 µg l–1) foundin the bloom in the south were chiefly due to pennate diatomsand larger heterotrophic dinoflagellates. Much lower carbonvalues (average 51 µg l–1; range 7-147 µgl–1) in the outer shelf region were mainly contributedby large centric diatoms (70-110 mu;m) and small dinoflagellates(5-25 µm). Wide ranges of algal cell sizes were observedin both southern and northern communities; the overlapping ofsizes of diatoms and flagellates, the latter containing heterotrophs,suggested complex trophic relationships within the planktonand an enhanced heterotrophic activity in the south. iSERS (SERS=surface‐enhanced Raman scattering) microscopy is an emerging Raman‐based staining technique for the selective localization of target proteins on cells and tissues using antibody‐ SERS nanotag conjugates. In this contribution we demonstrate the feasibility of iSERS for imaging of programmed cell death‐ligand 1 (PD‐L1), an important predictive biomarker, on single SkBr‐3 breast cancer cells. Low molecular weight protein kinase from maize seedlings was isolated. The molecular weight of the enzyme was below 10 000. The enzyme preferentially utilized ATP and casein as donor and acceptor of phosphorus, respectively. Casein kinase was a cyclic nucleotide-independent, heparin-sensitive enzyme. The low molecular weight casein kinase was resolved into basic, neutral and slightly acidic peaks of activity by chromatofocussing.  相似文献

Genetic variants of serum alkaline phosphatase were studied by the method of starch gel electrophoresis in the Zlotnicka Pstra breed of pigs. Two regions of alkaline phosphatase migration were observed. A single fraction in region I and four different phenotypes: AB, B, BC and BD in region II, were found. For AB, B and BC phenotypes the genetic control by three alleles AkpA, AkpB and AkpC in suggested. The effects of chilling and freezing temperature on membrane permeability and ATP content were studied in the leaves of cucumber ( Cucumis sativus L.) and winter rape ( Brassica napus L. var. oleifera L.) leaves, grown at different temperatures. In the winter rape leaves, the endogenous ATP content was modified by application of dinitrophenol (DNP) solutions of different concentrations. The low temperature-induced changes in membrane permeability (as monitored by the conductivity method) were found to be associated with ATP decrease, both in the chilling-sensitive and chilling-resistant (subjected to freezing) plants. In tissues showing reversible injuries, changes in ATP content preceded those in membrane permeability and the adenylate energy charge was affected slightly. In tissues showing irreversible membrane damage, the ATP content was always below 0.4 μmol (g dry weight)−1 and the adenylate energy charge was near 0.5. DNP treatment increased freezing sensitivity of winter rape leaves. In the cold-hardened winter rape leaves, however, freezing and thawing did not significantly affect ATP content or the energy charge, although the specimen showed a rather large increase in membrane permeability. In these leaves ATP content recovered about 20 h after a freezing and thawing treatment. It is proposed that a decrease in ATP supply might be the primary reason for the membrane leakiness at low temperature, both in chilling-sensitive and chilling-resistant (subjected to freezing) plants. The conclusion is, however, not true for the cold-acclimated, frostadapted cells.  相似文献

Reaction of α-chymotrypsin with o-[35S]hydroxy-α-toluenesulfonic acid sultone (I) at pH 7 produces an intermediate which substantially decomposes over a period of 24 hr at pH 3 as determined by gel chromatography. Isolation of the intermediate at pH 3 followed by denaturation in urea, reduction of disulfide bonds, and SDS-urea gel electrophoresis resulted in separation of the three peptides which compose the enzyme. Radioactivity (35S) was associated with the peptide which contains serine 195 and not with the peptide containing histidine 57. These results indicate that a covalent linkage is established between sultone I and α-chymotrypsin, presumably at serine 195.  相似文献
