Despite high taxonomic diversity elsewhere in North America during the Pleistocene, vertebrate faunas are exceedingly rare in the region of northern Mexico. Térapa, a unique fossil site located in the present-day desert of Northcentral Sonora, Mexico (29°41′N, 109°39′W, 605 m elevation), contributes to our understanding of the paleoecology and paleoclimate of the region during the Late Pleistocene, ca. 43,000-40,000 cal. yr BP. At least 60 vertebrate taxa, including amphibians, turtles, a crocodilian, snakes, birds and many mammals, have been recovered from an 11-m thick sequence of fossiliferous sediments. The diversity and tropical affinity of these taxa suggest a more-forested environment than the thornscrub desert habitat present in this region today.Isotopic analyses of tooth enamel carbonate from ancient mammalian herbivores suggest that the Sonoran desert has undergone considerable climate change since the Late Pleistocene. Bulk carbon (δ13C) and oxygen (δ18O) isotopes from nine mammalian fossils indicate a habitat mosaic with variations in diet that include browsers, mixed feeders and C4 hyper-grazers (δ13C range of − 10‰ to 2‰). Unique to this site are δ13C tooth enamel values of − 6.1 and − 5.6 ‰ for the deer Odocoileus, which suggest a more variable diet than strict browsing, including possibly feeding on CAM and/or C4 plants. Serial sampling of carbon (δ13C) and oxygen (δ18O) isotopes for ancient mammal teeth with hypsodont dentitions (fossil Equus and Bison,) as well as δ18O meteroric water estimates from well-supported climatic models suggest a cooler and more equable environment at Térapa during the Late Pleistocene. These results also support previous habitat reconstructions inferred from the macrobotanical and packrat midden records of northern Sonora (Mexico). High-resolution stable isotope geochemistry indicates that: 1) ancient Térapa was covered with forest and grassland habitats that extended northward into Mexico by about 350 km relative to their present-day northern limits during the Late Pleistocene; and 2) an Amount Effect (AE) is demonstrated in the fossil record at Térapa even though the climate was less seasonal compared to the modern desert habitat.  相似文献

Aims Both dominance distribution of species and the composition of the dominant species determine the distribution of traits within community. Leaf carbon (C) and nitrogen (N) isotopic composition are important leaf traits, and such traits of dominant species are associated with ecosystem C, water and N cycling. Very little is known how dominant species with distinct traits (e.g. N-fixing leguminous and non-leguminous trees) mediate resource utilization of the ecosystems in stressful environment.Methods Leaves of 81 dominant leguminous and non-leguminous trees were collected in forest (moist semi-deciduous and dry semi-deciduous ecosystems) and savanna (costal savanna, Guinean savanna and west Sudanian savanna ecosystems) areas and the transitional zone (between the forest and the savanna) along the transect from the south to the north of Ghana. We measured leaf traits, i.e. leaf δ 13 C, leaf δ 15 N, leaf water content, leaf mass per area (LMA) and C and N concentration. Correlation analyses were used to examine trait–trait relationships, and relationships of leaf traits with temperature and precipitation. We used analysis of covariance to test the differences in slopes of the linear regressions between legumes and non-legumes.Important findings Leaf δ 13 C, δ 15 N, leaf water content and LMA did not differ between leguminous and non-leguminous trees. Leaf N concentration and C:N ratio differed between the two groups. Moreover, leaf traits varied significantly among the six ecosystems. δ 13 C values were negatively correlated with annual precipitation and positively correlated with mean annual temperature. In contrast, leaf δ 15 N of non-leguminous trees were positively correlated with annual precipitation and negatively correlated with mean annual temperature. For leguminous trees, such correlations were not significant. We also found significant coordination between leaf traits. However, the slopes of the linear relationships were significantly different between leguminous and non-leguminous trees. Our results indicate that shifts in dominant trees with distinct water-use efficiency were corresponded to the rainfall gradient. Moreover, leguminous trees, those characterized with relative high water-use efficiency in the low rainfall ecosystems, were also corresponded to the relative high N use efficiency. The high proportion of leguminous trees in the savannas is crucial to mitigate nutrient stress.  相似文献

To understand the impact of alternative translation initiation on a proteome, we performed a proteome‐wide study on protein turnover using positional proteomics and ribosome profiling to distinguish between N‐terminal proteoforms of individual genes. By combining pulsed SILAC with N‐terminal COFRADIC, we monitored the stability of 1,941 human N‐terminal proteoforms, including 147 N‐terminal proteoform pairs that originate from alternative translation initiation, alternative splicing or incomplete processing of the initiator methionine. N‐terminally truncated proteoforms were less abundant than canonical proteoforms and often displayed altered stabilities, likely attributed to individual protein characteristics, including intrinsic disorder, but independent of N‐terminal amino acid identity or truncation length. We discovered that the removal of initiator methionine by methionine aminopeptidases reduced the stability of processed proteoforms, while susceptibility for N‐terminal acetylation did not seem to influence protein turnover rates. Taken together, our findings reveal differences in protein stability between N‐terminal proteoforms and point to a role for alternative translation initiation and co‐translational initiator methionine removal, next to alternative splicing, in the overall regulation of proteome homeostasis.  相似文献

Cannabinoid receptors are G-protein-coupled receptors comprised of seven transmembrane helices. We hypothesized that the extended helix of the receptor interacts differently with POPC bilayers due to the differing distribution of charged amino acid residues. To test this, hCB1(T377-E416) and hCB2(K278-H316) peptides were studied with 31P and 2H solid-state NMR spectroscopy by incorporating them into 1-palmitoyl-2-oleoyl-sn-glycerophosphocholine bilayers. Lipid affinities of the 40- and 39-residue peptides were analyzed on the basis of 31P and 2H spectral line shapes, order parameters, and T1 relaxation measurements of the POPC bilayers. Lipid headgroup perturbations were noticed in the 31P NMR spectra in the lipid/peptide mixtures when compared with the pure lipids. 2H order parameters were calculated from the quadrupolar splitting of the de-Paked 2H NMR spectra. At the top of the acyl chain, pure lipids had an average S(CD) approximately = 0.20, whereas S(CD) approximately = 0.16 and S(CD) approximately = 0.18 were found in the presence of hCB1(T377-E416) and hCB2(K278-H316), respectively. S(CD) values decreased in the central part of the acyl chains when compared to the pure POPC lipids, indicating a change in the dynamic properties of the lipid membrane in the presence of the cannabinoid peptides. R(1Z) vs S2(CD) plots exhibited a linear dependency with and without the peptides, with an increase in slope upon addition of the peptides to the POPC, indicating that the dynamics of the lipid bilayer is dominated by fast axially symmetric motion. This study provides insights into the interaction of cannabinoid peptides with the membrane bilayer by investigating the headgroup and acyl chain dynamics.  相似文献

The intrinsic qualities of lanthipeptides for their use as therapeutic drugs present several challenges because of their properties, which include stability, solubility and bioavailability, which, under physiological conditions, are very low. Researches have encouraged clinical evaluation of a few compounds, such as mutacin 1140, microbisporicin, actagardine and duramycin, with pharmacokinetic profiles showing rapid distribution and elimination rates, good bioavailability and fecal excretion, as well as high protein binding. Local and parenteral administration are currently suitable to minimize environmental influences on lanthipeptides and ensure efficient activity. Nevertheless, valuable improvements on pharmacodynamic and pharmacokinetic properties may also permit systemic applications via enteral routes. Understanding how rational modifications influence the desired pharmacological and pharmacokinetic properties of these biomolecules would help to answer some specific questions about their susceptibility to environmental changes, mechanism of action and how to engineer other peptides of the same group to improve their clinical relevance.  相似文献

Cannabinoids are compounds that can modulate neuronal functions and immune responses via their activity at the CB1 receptor. We used 2H NMR order parameters and relaxation rate determination to delineate the behavior of magnetically aligned phospholipid bilayers in the presence of several structurally distinct cannabinoid ligands. THC (Δ9-Tetrahydrocannabinol) and WIN-55,212-2 were found to lower the phase transition temperature of the DMPC and to destabilize their acyl chains leading to a lower average SCD (≈ 0.13), while methanandamide and CP-55,940 exhibited unusual properties within the lipid bilayer resulting in a greater average SCD (≈ 0.14) at the top of the phospholipid upper chain. The CB1 antagonist AM281 had average SCD values that were higher than the pure DMPC lipids, indicating a stabilization of the lipid bilayer. R1Z versus |SCD|2 plots indicated that the membrane fluidity is increased in the presence of THC and WIN-55,212-2. The interaction of CP-55,940 with a variety of zwitterionic and charged membranes was also assessed. The unusual effect of CP-55,940 was present only in bicelles composed of DMPC. These studies strongly suggest that cannabinoid action on the membrane depends upon membrane composition as well as the structure of the cannabinoid ligands.  相似文献

Neutral models are often used as null models, testing the relative importance of niche versus neutral processes in shaping diversity. Most versions, however, focus only on regional scale predictions and neglect local level contributions. Recently, a new formulation of spatial neutral theory was published showing an incompatibility between regional and local scale fits where especially the number of rare species was dramatically under‐predicted. Using a forward in time semi‐spatially explicit neutral model and a unique large‐scale Amazonian tree inventory data set, we show that neutral theory not only underestimates the number of rare species but also fails in predicting the excessive dominance of species on both regional and local levels. We show that although there are clear relationships between species composition, spatial and environmental distances, there is also a clear differentiation between species able to attain dominance with and without restriction to specific habitats. We conclude therefore that the apparent dominance of these species is real, and that their excessive abundance can be attributed to fitness differences in different ways, a clear violation of the ecological equivalence assumption of neutral theory.  相似文献

Abstract

Young children are considered critical receptors of potentially toxic trace elements (PTEs) by non-dietary ingestion of contaminated soil. The study assessed the potential enrichment of soil and the health risk of PTEs to 471 children less than seven years via non-dietary soil ingestion at six Early Childhood Development Centers (ECDCs) in urban low-income settings. The total concentrations of PTEs were determined by ICP-AES after wet acid digestion. The extent of soil contamination with PTEs and their source apportionment were assessed by the enrichment factor (EF). The US-EPA risk assessment model was used to determine the risk of PTE exposure by children. Multivariate statistical analyses and the EF suggested anthropogenic origin of PTEs in playgrounds and indoors, especially Cd and Pb from atmospheric deposition. Indoor floor dust at ECDCs was enriched (significant to extreme) with PTEs of anthropogenic origin imported from the outside environment. Children at the six ECDCs were not at significant non-carcinogenic risk of PTEs in soil and dust through non-dietary ingestion. The study setting is typical of urban child play centers in low-income countries which needs regular risk assessment and the enforcement of legislation in order to reduce the exposure of children to PTEs.  相似文献
