Background
Pectinase enzymes present a high priced category of microbial enzymes with many potential applications in various food and oil industries and an estimated market share of $ 41.4 billion by 2020.Results
The production medium was first optimized using a statistical optimization approach to increase pectinase production. A maximal enzyme concentration of 76.35 U/mL (a 2.8-fold increase compared with the initial medium) was produced in a medium composed of (g/L): pectin, 32.22; (NH4)2SO4, 4.33; K2HPO4, 1.36; MgSO4.5H2O, 0.05; KCl, 0.05; and FeSO4.5H2O, 0.10. The cultivations were then carried out in a 16-L stirred tank bioreactor in both batch and fed-batch modes to improve enzyme production, which is an important step for bioprocess industrialization. Controlling the pH at 5.5 during cultivation yielded a pectinase production of 109.63 U/mL, which was about 10% higher than the uncontrolled pH culture. Furthermore, fed-batch cultivation using sucrose as a feeding substrate with a rate of 2 g/L/h increased the enzyme production up to 450 U/mL after 126 h.Conclusions
Statistical medium optimization improved volumetric pectinase productivity by about 2.8 folds. Scaling-up the production process in 16-L semi-industrial stirred tank bioreactor under controlled pH further enhanced pectinase production by about 4-folds. Finally, bioreactor fed-batch cultivation using constant carbon source feeding increased maximal volumetric enzyme production by about 16.5-folds from the initial starting conditions.332.

Lupus nephritis (LN) is a major cause of morbidity and mortality in systemic lupus erythematosus (SLE). Previous studies suggest that mutant A20 binding inhibitor of NF-κB 1 (ABIN1) protein encoded by tumor necrosis factor alpha-induced protein 3 interacting protein 1 (TNIP1) gene is associated with LN via NF-κB dysregulation. The aim of the current study was to evaluate the association of TNIP1 gene SNP rs7708392 with SLE and LN in Egyptian patients. 5′ nuclease Allelic discrimination was used to evaluate the frequency of TNIP1 SNP rs7708392 in 53 patients with LN, 57 SLE patients without nephritis and 85 healthy controls. The genotyping analysis revealed that the CC genotype was more frequent in controls than SLE patients, while GC and GG genotypes were more common in SLE patients. Moreover, the GG genotype and the G allele were significantly more prevalent among LN patient than non-LN patients (P?<?0.001). In LN patients, the most common genotype was GG (56.6%), while among the non-LN patients; the CG genotype was the most common (59.6%). Regression analysis demonstrated that SLE patients carrying only one G allele had a 3.4 folds increased risk for LN. Our results suggested that TNIP1 SNP (rs7708392) might be associated with the LN in Egyptian SLE patients. TNIP1 SNP (rs7708392) might be used to identify patients at risk of developing LN, which could help in early detection and treatment before progression to end-stage renal disease, improving patients’ outcome and quality of life. 相似文献

Phytohormone auxin plays an indispensable role in the plethora of plant developmental process starting from the cell division, and cell elongation to morphogenesis. Auxins are transported to different parts of the plant by different sophisticated transporter molecules known as ‘auxin transporters’.There are four auxin transporter families that have been reported so far in the plant kingdom which includes AUX/LAX (AUXIN-RESISTANT1–LIKES), PIN (PIN-FORMED, auxin efflux carriers), ABCB ((ATP-binding cassette-B (ABCB)/P-glycoprotein (PGP)) and PILS (PIN-Likes). Auxin influx and efflux carriers are distributed in a polar fashion in the plasma membrane whereas ABCB and PILS are present in a non-polar fashion. Other than AUX/LAX, other auxin transporters harbor N-and C-terminal conserved domains along with a variable hydrophilic loop in the transmembrane domain. The AUX/LAX, ABCB and PIN transporters mediate long distance auxin transport whereas PILS and PIN5 protein involved in intracellular auxin homeostasis. 相似文献

Knowledge regarding the 3D structure of a protein provides useful information about the protein’s functional properties. Particularly, structural similarity between proteins can be used as a good predictor of functional similarity. One method that uses the 3D geometrical structure of proteins in order to compare them is the similarity value (SV). In this paper, we introduce a new definition of the SV measure for comparing two proteins. To this end, we consider the mass of the protein’s atoms and concentrate on the number of protein’s atoms to be compared. This defines a new measure, called the weighted similarity value (WSV), adding physical properties to geometrical properties. We also show that our results are in good agreement with the results obtained by TM-SCORE and DALILITE. WSV can be of use in protein classification and in drug discovery. 相似文献

Increased demand for energy worldwide has resulted in increasing interest in alternative renewable sources of biofuels. Demand for improved systems of bioenergy generation, environmental remediation, and coproduction of high value bioactive compounds has led to the potential use of algae in biomass utilization. In Malaysia, palm oil industries generate high amount of solid wastes. Palm Oil Mill Effluent (POME) is estimated to be three times of the amount of crude palm oil produced. POME is a heavily polluting wastewater due to its high chemical oxygen demand (COD), high biochemical oxygen demand (BOD), and high contents of minerals such as nitrogen and phosphorus that can cause severe pollution to the environment and water resources. Gender exponence in underived French nouns is particularly opaque. In this paper, we identify two subsystems of gender assignment: monogendered nouns, bi-gendered nouns; we show what they share and where they diverge. We show how the notorious floating final consonants of the language are handled in each subsystem for purposes of gender exponence. Our analysis and proposals develop in two steps: (a) we elaborate on the internal structure of √P, arguing that it is more richly articulated than had been assumed in previous work; (b) we propose that a gender feature sitting on n probes into its complement √P for valuation. Our proposal opens a range of selectional options of √P by n. These selectional options along with the probing device they feed, are shown to account for the French evidence. 相似文献

The apical complex of Toxoplasma gondii enables it to invade virtually all nucleated cells in warm‐blooded animals, including humans, making it a parasite of global importance. Anti‐T. gondii cellular defence mechanisms depend largely on interferon (IFN)‐γ production by immune cells. However, the molecular mechanism of IFN‐β‐mediated defence remains largely unclear. Here, mouse peritoneal macrophages and murine embryonic fibroblasts (MEFs) primed with recombinant IFN‐β and IFN‐γ showed different pathways of activation. Treatment of these cells with IFN‐β or IFN‐γ inhibited T. gondii (type II PLK strain) growth. Priming macrophages with IFN‐β had no effect on inflammatory cytokine expression, inducible nitric oxide synthase or indoleamine 2,3‐dioxygenase, nor did it have an effect on their metabolites, nitric oxide and kynurenine respectively. In contrast, IFN‐γ stimulation was characterized by classical macrophage activation and T. gondii elimination. IFN‐β activation recruited the immunity‐related GTPase M1 (IRGM1) to the parasitophorous vacuole in the macrophages and MEFs. Anti‐toxoplasma activities induced by IFN‐β were significantly reduced after IRGM1 knockdown in murine macrophages and in IRGM1‐deficient MEFs. Thus, this study unravels an alternative pathway of macrophage activation by IFN‐β and provides a mechanistic explanation for the contribution of IRGM1 induced by IFN‐β to the elimination of T. gondii. 相似文献

Due to their RNA-N-glycosidase activity, ribosome-inactivating proteins (RIPs) are attractive candidates as antitumor and antiviral agents in biomedical and agricultural research. We have isolated and characterized two such proteins, foetidissimin II and texanin, from two Cucurbitaceae species. Foetidissimin II, obtained from the roots of Cucurbita foetidissima, was identified as a type-2 RIP, with a molecular weight of 61 kDa, as estimated by gel electrophoresis. It is composed of two chains, a 29-kDa chain A, and a 32-kDa chain B. Texanin, isolated from the fruits of Cucurbita texana, is a type-I RIP, with a single chain of molecular weight 29.7 kDa, as estimated by MALDI-TOF-MS. Both proteins exhibit RNA-N-glycosidase activity, with aniline playing a critical role in rRNA cleavage. The IC50 value of foetidissimin II, determined by cell-free protein-synthesis inhibition, was 0.251 muM. In an in vitro cytotoxicity assay, foetidissimin II exhibited IC50 values of ca. 70 nM to both adenocarcinoma and erythroleukemia cells. Texanin exhibited a weaker anticancer activity against erythroleukemia cells, with an IC50 value of 95 microM, but no activity against adenocarcinoma cells. Thermoluminescence glow curves of gamma-irradiated samarium-doped lithium borosilicate glass were investigated. The number of overlapping peaks was determined using the repeated initial rise method. The glow curves were deconvoluted into four overlapping peaks. The trapping parameters such as activation energy E, frequency factor (s), and kinetic order (b) for each peak were determined. The obtained results indicated that the lithium borosilicate glass doped with samarium had four electron trap levels with the average activation energies of 0.82, 1.01, 1.21, and 1.31 eV. Thermal fading analysis of the individual peaks based on the deconvolution data was performed. The obtained results showed high thermal fading of the first peak, but high thermal stability of the second and third peaks compared with the other peaks. These results could be used to explain some observed properties such as high thermal fading and light sensitivity for this thermoluminescent material. Moreover, the obtained results may be helpful in minimizing fading corrections in dosimetric applications. 相似文献
