Opposing effects have been ascribed to nitric oxide (NO) on retinal microvascular survival. We investigated whether changes in the redox state may contribute to explain apparent conflicting actions of NO in a model of oxygen-induced retinal vasoobliteration. Retinal microvascular obliteration was induced by exposing 7-day-old rat pups (P7) for 2 or 5 days to 80% O(2). The redox state of the retina was assessed by measuring reduced glutathione and oxidative and nitrosative products malondialdehyde and nitrotyrosine. The role of NO on vasoobliteration was evaluated by treating animals with nitric oxide synthase (NOS) inhibitors (N-nitro-l-arginine; L-NA) and by determining NOS isoform expression and activity; the contribution of nitrosative stress was also determined in animals treated with the degradation catalyst of peroxynitrite FeTPPS or with the superoxide dismutase mimetic CuDIPS. eNOS, but not nNOS or iNOS, expression and activity were increased throughout the exposure to hyperoxia. These changes were associated with an early (2 days hyperoxia) decrease in reduced glutathione and increases in malondialdehyde and nitrotyrosine. CuDIPS, FeTPPS, and L-NA treatments for these 2 days of hyperoxia nearly abolished the vasoobliteration. In contrast, during 5 days exposure to hyperoxia when the redox state rebalanced, L-NA treatment aggravated the vasoobliteration. Interestingly, VEGFR-2 expression was respectively increased by NOS inhibition after short-term (2 days) exposure to hyperoxia and decreased during the longer hyperoxia exposure. Data disclose that the dual effects of NO on newborn retinal microvascular integrity in response to hyperoxia in vivo depend on the redox state and seem mediated at least in part by VEGFR-2.  相似文献

Roots of a wide range of plant species exude carboxylates, e.g. citrate, into the rhizosphere, to mobilise sparingly available phosphate. We investigated the carboxylates in root exudates of Banksia grandisWilld. (Proteaceae), which occurs on severely phosphate-impoverished soils in Western Australia. Plants were grown in pots with a nutrient-poor quartz sand, with phosphate, at 25 g P g–1, added as either K-phosphate, glycerol phosphate, Fe-phosphate or Al-phosphate.Plants grown on Fe-phosphate or Al-phosphate formed `proteoid' or `cluster' roots, and exuded significant amounts of carboxylates. Plants grown on K-phosphate did not form cluster roots; their leaves were chlorotic, and some of these plants died during the experiment. Plants grown on glycerol phosphate did have cluster roots, but their leaves also became chlorotic, albeit later in the experiment.Tri- and dicarboxylates (citrate, 60%; malate, 25%; trans-aconitate, 14%) were the major carboxylates in root exudates when P was supplied as Al-phosphate. The same tri- and dicarboxylates were also exuded when P was supplied as Fe-phosphate (31, 14 and 12%, respectively). In addition, these plants exuded monocarboxylates (lactate, 30%; acetate, 12%). We analysed the effect of the different carboxylates on the mobilisation of phosphate and Fe in two different types of soils. The ecological significance of the difference in exudate spectrum for the mobilisation of nutrients and for the detoxification of aluminium is discussed.Because the leaves of plants grown with K-phosphate or glycerol-phosphate appeared chlorotic, we analysed the concentrations of P, Fe, Zn, Mn and Cu in these leaves. Only the concentration of total P was considerably higher in leaves of plants grown with K- or glycerol-phosphate than that in leaves of plants grown with Fe- or Al-phosphate. Both the concentration of total Fe and that of reduced Fe was the same in chlorotic leaves as that in leaves of plants grown with Fe- or Al-phosphate, which had a healthy appearance. It is concluded that P-induced chlorosis was not due to a lack of total or reduced Fe; it may have been due to precipitation of Fe by phosphate.  相似文献

Epidemiology in host meta-populations depends on parasite ability to disperse between, establish and persist in distinct sub-populations of hosts. We studied the genetic factors determining the short-term establishment, and long-term maintenance, of pathogens introduced by infected hosts (i.e. carriers) into recipient populations. We used experimental populations of the freshwater ciliate Paramecium caudatum and its bacterial parasite Holospora undulata. Parasite short-term spread (approximately one horizontal transmission cycle) was affected mainly by carrier genotype, and its interactions with parasite and recipient genotypes. By contrast, parasite longer term spread (2-3 horizontal transmission cycles) was mostly determined by parasite isolate. Importantly, measures of parasite short-term success (reproductive number, R) were not good predictors for longer term prevalence, probably because of the specific interactions between host and parasite genotypes. Analogous to variation in vectorial capacity and super-spreader occurrence, two crucial components of epidemiology, we show that carrier genotype can also affect disease spread within meta-populations.  相似文献

Plants have been used to produce many diverse and valuable recombinant proteins, including subunit vaccines, antibodies and antibody fragments, hormones, blood products, cytokines, and enzymes. Different plant species and platforms have been explored as production hosts, each with unique properties in terms of the gene transfer method, production time, environmental containment, scalability, downstream processing strategy, protein folding and accumulation, and overall costs. Seed-based systems have many advantages because they exploit the natural storage properties of seeds, which facilitate batch processing and distribution. Seeds possess specialized storage organelles that may be used to accumulate recombinant proteins, offering stability both in planta and after harvest in the final preparation/formulation. The post-harvest stabilizing effect of seeds allows recombinant subunit vaccines and antibodies to be delivered via the mucosal route because they are better able to withstand this harsh microenvironment when protected by the plant matrix. Native storage organelles such as starch granules and protein bodies offer this protective effect, but protein storage organelles can also be induced ectopically in vegetative tissues. In this paper, we discuss the technical capabilities of storage organelle-based expression platforms and their potential applications.  相似文献

Background

Chagas disease prevention critically depends on keeping houses free of triatomine vectors. Insecticide spraying is very effective, but re-infestation of treated dwellings is commonplace. Early detection-elimination of re-infestation foci is key to long-term control; however, all available vector-detection methods have low sensitivity. Chemically-baited traps are widely used in vector and pest control-surveillance systems; here, we test this approach for Triatoma spp. detection under field conditions in the Gran Chaco.

Methodology/Principal Findings

Using a repeated-sampling approach and logistic models that explicitly take detection failures into account, we simultaneously estimate vector occurrence and detection probabilities. We then model detection probabilities (conditioned on vector occurrence) as a function of trapping system to measure the effect of chemical baits. We find a positive effect of baits after three (odds ratio [OR] 5.10; 95% confidence interval [CI95] 2.59–10.04) and six months (OR 2.20, CI95 1.04–4.65). Detection probabilities are estimated at p≈0.40–0.50 for baited and at just p≈0.15 for control traps. Bait effect is very strong on T. infestans (three-month assessment: OR 12.30, CI95 4.44–34.10; p≈0.64), whereas T. sordida is captured with similar frequency in baited and unbaited traps.

Conclusions/Significance

Chemically-baited traps hold promise for T. infestans surveillance; the sensitivity of the system at detecting small re-infestation foci rises from 12.5% to 63.6% when traps are baited with semiochemicals. Accounting for imperfect detection, infestation is estimated at 26% (CI95 16–40) after three and 20% (CI95 11–34) after six months. In the same assessments, traps detected infestation in 14% and 8.5% of dwellings, whereas timed manual searches (the standard approach) did so in just 1.4% of dwellings only in the first survey. Since infestation rates are the main indicator used for decision-making in control programs, the approach we present may help improve T. infestans surveillance and control program management.  相似文献

Studies on members of the crow family using the "Aesop's Fable" paradigm have revealed remarkable abilities in these birds, and suggested a mechanism by which associative learning and folk physics may interact when learning new problems. In the present study, children between 4 and 10 years of age were tested on the same tasks as the birds. Overall the performance of the children between 5-7-years was similar to that of the birds, while children from 8-years were able to succeed in all tasks from the first trial. However the pattern of performance across tasks suggested that different learning mechanisms might be being employed by children than by adult birds. Specifically, it is possible that in children, unlike corvids, performance is not affected by counter-intuitive mechanism cues.  相似文献
